Higher cyclobutane pyrimidine dimer and (6-4) photoproduct yields in epidermis of normal humans with increased sensitivity to ultraviolet B radiation.

Cyclobutane pyrimidine dimer (CPD) and (6-4) photoproduct induced in the epidermis of five Japanese volunteers exposed to ultraviolet (UVB) radiation were measured with monoclonal antibodies specific for each photoproduct. The volunteers comprised two individuals who are sensitive to solar irradiation (low minimal erythema dose [MED]) and three who are less sensitive. The yields of CPD and (6-4) photoproduct were within similar ranges after 1 MED or 3 MED doses. The yields of both photoproducts after the same dose of irradiation (120 mJ/cm2) were higher in UV-sensitive individuals than in less sensitive individuals. By 24 h after irradiation, an average of 60% of CPD had been removed after the 1 MED dose, 27% after the 3 MED dose and 34% after 120 mJ/cm2. The (6-4) photoproduct was removed within 24 h, independently of the dose of UVB tested. These data suggest that DNA photoproducts participate in initiating UVB-induced erythema, and partially explain why individuals with higher sensitivity to UVB have a higher risk of UV-induced skin cancer.

Retinal detachment in identical twins with Stickler syndrome type 1.

BACKGROUND: The high incidence of retinal detachment and its poor surgical prognosis in patients with Stickler syndrome are well known. However, the vitreoretinal relation to retinal detachment in this syndrome is uncertain. METHODS: Vitreoretinal examination with a binocular indirect ophthalmoscope and a Goldmann three mirror contact lens was performed on identical twin boys with Stickler syndrome. Each had retinal detachment in the left eye, and many aspects of their fundus findings were similar. The vitreous showed distinct abnormalities consistent with congenital vitreous anomaly of type 1 Stickler syndrome. The twins were followed up for 4 years after undergoing a successful operation to reattach their left retinas. RESULTS: Rhegmatogenous detachment with multiple tears occurred in the right eye of only one twin during the follow up period, despite the similar condition of their fundi. Although vitreous body was not present in most parts, slightly opaque vitreous cortex was attached to the retina near the ora serrata, and neither twin had posterior vitreous detachment during the follow up period. CONCLUSION: Multiple retinal tears appeared simultaneously in the right eye of one twin, indicating some tractional force had acted on the retina. It is believed that this force was caused by very thin vestigial vitreous cortex attached to the retina. Although these observations have been limited, vitreoretinal findings of the twins and their father were consistent and suggested presence of thin vitreous cortex attached to the retina without posterior vitreous detachment.

Expression of platelet derived growth factor B chain and beta receptor in human coronary arteries after percutaneous transluminal coronary angioplasty: an immunohistochemical study.

OBJECTIVE: To evaluate whether expression of platelet derived growth factor B (PDGF-B) protein is associated with expression of its receptor protein in human coronary arteries after angioplasty and to identify cells involved. BACKGROUND: PDGF is considered an important growth factor in the repair process of the vessel wall after angioplasty. In situ hybridisation has revealed expression of PDGF-A and -B chain messenger ribonucleic acid (mRNA) in human coronary arteries at sites of postangioplasty injury. METHODS: Target and non-target sites of eight coronary arteries were studied immunohistochemically for PDGF-B and PDGF-beta receptor proteins in relation to macrophages, T lymphocytes, smooth muscle cells, and HLA-DR positive cells. RESULTS: The PDGF-B and PDGF-beta receptor proteins were expressed in areas with distinct repair, containing alpha actin negative spindle cells, macrophages and, at later stages, alpha actin positive smooth muscle cells as well. When the neointima was composed mainly of alpha actin smooth muscle cells, PDGF-B expression was rare and PDGF-beta receptor expression was negative. CONCLUSIONS: There is expression of PDGF-B and PDGF-beta receptor proteins at sites of postangioplasty repair in human coronary arteries. The associated cells are mainly macrophages and alpha actin negative spindle cells; the latter may be dedifferentiated smooth muscle cells. A link between PDGF expression and the postangioplasty time interval suggests a relation with cell differentiation as part of the maturation of the repair tissue. Mutual expression of both the growth factor and its receptor protein strongly suggests that in humans a PDGF mediated repair process occurs, with involvement of smooth muscle cells and macrophages.

Granulosa cells express integrin alpha 6: possible involvement of integrin alpha 6 in folliculogenesis.

To identify differentiation antigens of granulosa cells, we have raised two monoclonal antibodies against porcine granulosa cells (POG-2 antibody) and human granulosa cells (OG-1 antibody). Analysis of N-terminal amino acid sequences of purified OG-1 and POG-2 antigens revealed that they were identical to human integrin alpha 6 and its porcine homolog, respectively. An immunohistochemical study showed that integrin alpha 6 was highly expressed on granulosa cells in preovulatory follicles in the human ovary, whereas it was highly expressed on granulosa cells in small follicles in the porcine ovary. As a common characteristic of integrin alpha 6 expression in both ovaries, granulosa cells located in inner layers, which are not in contact with the basal lamina, expressed integrin alpha 6 on the cell surface. This expression profile suggests that the interaction between integrin and extracellular matrices occurs in the granulosa cells located in inner layers. This cell surface interaction is reported to modulate soluble signals to the cells. Since the expression of integrin alpha 6 on inner-layer granulosa cells is related to stages of differentiation, this interaction may be involved in folliculogenesis.

The effect of atrial natriuretic peptide on pulmonary acid injury in a pig model.

The effect of exogenous atrial natriuretic peptide (ANP) on pulmonary injury induced by aspiration of hydrochloric acid (0.1 N HCl, 2.0 ml/kg) was examined in anesthetized pigs ventilated with 100% oxygen. Group-specific diuretic treatment was used 60 min after aspiration and followed for a further 120 min. No diuretics were used for Group 1, 0.1 microg/kg/min of ANP was infused intravenously for 60 min in Group 2, and 0.05 mg/kg of furosemide was injected (intravenous bolus) in Group 3. Total urine volume after diuretic treatment did not differ significantly between Groups 2 and 3. However, the increase in PaO2 (to 386.3 +/- 67.5 mm Hg) after infusion of ANP was significantly higher than the increase in PaO2 (to 275.9 +/- 63.3 mm Hg) after furosemide treatment, and in the no treatment control (to 171.1 +/- 31.5 mm Hg). Pulmonary hypertension induced by acid aspiration was significantly reduced (p < 0.01 versus Groups 1 and 3) during ANP infusion. ANP has a beneficial effect on acute lung injury, possibly through its diuretic and/or pulmonary vasodilating action.

Immunoelectron microscopic observation of the behaviors of peroxisomal enzymes inducibly synthesized in an n-alkane-utilizable yeast cell, Candida tropicalis.

We reported that immunoelectron microscopy was an excellent tool for determining the subcellular localization of thiolase isozymes, acetoacetyl-CoA thiolase (T-I) and 3-ketoacyl-CoA thiolase (T-III) in n-alkane-grown Candida tropicalis cells (KAMASAWA, N. et al., (1992). Cell Struct. Funct., 17: 203-207). Current investigation on the visualization of other peroxisomal enzymes, acyl-CoA oxidase (ACO), catalase (KAT), carnitine acetyltransferase (CAT), isocitrate lyase (ICL) and malate synthase (MS), showed that ACO localized in peroxisomes, KAT in peroxisomes and cytoplasm, and CAT in peroxisomes, mitochondria and cytoplasm. Most of ICL and MS were found in peroxisomes. These results agreed with previous biochemical studies and supported the presumed roles of these enzymes. The same technique was applied to study the process of synthesis and localization of these enzymes early in the cultivation period in n-alkane medium when peroxisomes began to proliferate. ACO and T-III were rapidly induced after transfer of cells from glucose- to n-alkane-media. There was a drastic change of their location from cytoplasm to peroxisomes between 1 h and 2 h after the transfer, while T-I, KAT and CAT were moderately induced in cytoplasm and their location was gradually changed to each organelle. ICL and MS, the key enzymes in the glyoxylate cycle, were already localized in peroxisomes in the glucose-grown cells and respective inducible enzymes also were gradually localized there. This visual analysis is useful for the vivid elucidation of the process of peroxisome proliferation and enzyme transport within a cell.

Expression of the chitinase III gene of Aeromonas sp. no. 10S-24 in Escherichia coli.

The chitinase III gene of Aeromonas sp. No. 10S-24 was expressed in Escherichia coli. Production of chitinase III by E. coli was 3-fold higher than that of chitinases in the culture supernatant of Aeromonas sp. The enzyme from E. coli was purified and characterized. The molecular weight of the chitinase III from E. coli was estimated by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) to be 55,000. This agreed with the calculated molecular weight of the mature chitinase III (54,111). However, it was different from that of the enzyme from Aeromonas sp. It showed that the chitinase III from Aeromonas sp. had an additional sugar chain, causing the higher molecular weight. Chitinase III from E. coli had almost the same enzymatic properties as chitinase III from Aeromonas sp., but the specific activity of the chitinase III from E. coli is slightly higher than that of the native chitinase III. Both enzymes hydrolyzed chitosan (80% deacetylated) well.

[Demonstration and characterization of CD 46, membrane cofactor protein, in gastric cancer tissue].

CD 46, membrane cofactor protein, is a membrane protein which shows different expression and phenotypes with the organ or cell in the same individual. Previously, we reported that gastric CD 46 was expressed strongly in the mucosal epithelium, mucosa and endothelial cells of vessels in the submucosal layer. Western blot analysis revealed that gastric CD 46 was expressed as one broad band with a molecular weight ranging from 60 kDa to 69 kDa, which was different from that of lymphocytes. In this study, we investigated the differences in expression and characterization of CD 46 in gastric cancer obtained by surgery as compared with non-cancerous mucosa. Expression of CD 46 was greater in gastric cancer than in non-cancerous mucosa. In some cases, the phenotypes of CD 46 in gastric cancer were different from those in the non-cancerous tissue.

Human betacellulin, a member of the EGF family dominantly expressed in pancreas and small intestine, is fully active in a monomeric form.

Betacellulin (BTC) was found to be expressed mainly in human pancreas and small intestine. This finding suggests that BTC possesses some specific function distinguished from the other members of epidermal growth factor (EGF) family. To clarify this function, the released form of human BTC has been expressed in E.coli, purified, and characterized. The recombinant human BTC was produced as an inclusion body. This material was dissolved in guanidine-HCl under reducing conditions, refolded, and purified through sequential liquid chromatography. Purified BTC was electrophoresed under reducing conditions and a molecular size of 18 kDa was determined, which is the supposed size of a dimer of the peptide. However, chemical analysis failed to show a covalently linked dimer. The molecular mass of BTC analyzed by mass spectrometry revealed it to be 9 kDa, which is consistent with theoretical value for a monomer. Recombinant BTC showed growth promoting activity for mouse fibroblasts and rat aortic smooth muscle cells which was equivalent to EGF On the other hand, BTC was found to exhibit a growth inhibitory effect on the cells overexpressing EGF receptor.

Localization of recurrent disease after extended lymph node dissection for carcinoma of the thoracic esophagus.

BACKGROUND: The role of aggressive locoregional dissection in the treatment of carcinoma of the thoracic esophagus is controversial. The extent of disease spread for which resection can be performed is not yet fully established. STUDY DESIGN: Of 230 patients who underwent extended lymph node dissection, including the cervical nodes, the overall hospital mortality rate was 4.8 percent. Recurrent carcinoma was clinically confirmed by periodic follow-up examination with diagnostic imaging in 83 patients. We examined the exact anatomic sites of recurrent lesions and the clinical courses of patients. Recurrence patterns were classified into locoregional, distant, and mixed types. RESULTS: Recurrence of the carcinoma was locoregional in 35 patients, distant in 38 patients, and mixed in ten patients. The frequency of locoregional recurrence was significantly lower in patients with less than eight positive nodes and in patients without invasion of neighboring organs. Locoregional recurrence in the dissected area was mainly found in the region around the recurrent nerves and the main bronchi. Lymph nodes outside of the dissected area developed recurrence only in cases of markedly advanced disease apart from the abdominal para-aortic nodes. The group with locoregional recurrence and the group with distant recurrence had similar clinical courses over time. In patients in whom recurrent lesions could be treated with resection and adjuvant therapy, the one-year survival rate after recurrence was 83 percent. CONCLUSIONS: Unless the disease was markedly advanced, systematic lymph node dissection in our procedures yielded good locoregional control. The relatively low rate of distant recurrence with acceptable hospital mortality rates favors an extensive operation. When recurrent lesions were localized macroscopically, surgical removal of the recurrent lesions offered good palliation.

Hematopoietic clone with karyotypic abnormalities of host origin after bone marrow transplantation: two case reports.

A patient with non-Hodgkin's lymphoma in remission developed a myelodysplastic syndrome (MDS) 12 years after ABMT. This patient had undergone bone marrow harvesting prior to any chemoradiotherapy. He had received the autograft following conditioning with high-dose CY and TBI. Chromosomal analysis of BM cells revealed complicated abnormalities. Similar karyotypic abnormalities in host-derived BM cells were found in another patient with AML who had received allogeneic BM following conditioning with CY plus TBI 15 months previously. These findings suggest that MDS or clonal karyotypic abnormalities following ABMT may derive from endogenous hematopoietic stem cells that survive the BMT preparative regimen.

A study of the bone morphogenetic protein derived from bovine demineralized dentin matrix.

We tried to extract and purify bone morphogenetic protein (BMP) from bovine demineralized dentin matrix (DDM). Crude dentin BMP (d-BMP) was extracted from DDM in 3 sequential steps. Purification of crude d-BMP was carried out by liquid chromatography. The molecular weight and the isoelectric point of the purified d-BMP were determined by sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE) and electrofocusing. All fractioned samples were bioassayed in the thigh muscle pouches of AKR strain mice to test their ability to induce new bone formation. A sequence of 3 extraction steps worked effectively to obtain crude d-BMP. The purified d-BMP was shown to be homogeneous on high performance liquid chromatography (HPLC) and SDS-PAGE. The molecular weight and the pI were 25 kDa and 6.5, respectively. The amino acid composition was different from that of known bone-derived BMP. The purified d-BMP induced new bone formation in the thigh muscle pouches. The molecular weight, pI and amino acid composition were different from those of bone-derived BMP.

Thermochemotherapy for cancer of the tongue using magnetic induction hyperthermia (implant heating system: IHS).

A 55-year-old patient with cancer of the tongue (T2N0M0) was treated by thermochemotherapy using interstitial magnetic induction hyperthermia (Implant Heating System: IHS). The patient received 2 courses of hyperthermia, each of which was 45 minutes long. At the same time, the patient received 2 courses of chemotherapy, which included intra-arterial infusion of 100 mg of cisplatin (CDDP) and 25 mg of peplomycin (PEP). The patient showed complete response (CR) to this therapy. To date, 1.5 years after completion of treatment, the patient has shown no recurrence. This therapy, which makes surgery and radiotherapy unnecessary, is promising, because it is expected to improve the quality of life (QOL) of cancer patients.
