Characterization of antigens of Paragonimus miyazakii by ELISA.

The characterization of adult worm and excretory-secretory (ES) antigens of P. miyazakii was performed with a sandwich enzyme-linked immunosorbent assay (ELISA) IgG and Fab' fragments were prepared from P. miyazakii-infected rat sera. The former was purified by affinity column chromatography. The latter was prepared from IgG2c by passage through a Protein A affinity column, then conjugated to horseradish peroxidase. Cross-reactivity of the ELISA with Fasciola hepatica. Fischoederius elongatus. Taenia saginata. Schistosoma mansoni and S. Japonicum was negligible. With Paragonimus westermani, the same genus as P. miyazakii, cross-reactivity was below 30%. The molecular weight of both antigens was comparable (36,000) as determined by Sephadox G-100 gel filtration. Isoelectric points in both antigens had many peaks in a pH range of 3.9-4.7. In adult worm, pI of the major peaks were 4.6 and 4.7 and minor part of peaks had the common isoelectric points with four ES peaks in a pH range of 4.1-4.6. Antigenicity per protein concentration of both antigens was examined by the ELISA. The ES antigen was about five times as active antigenically as adult worm antigen. These results suggest that ES is one of the major antigens inducing antibody synthesis in rats infected with P. miyazakii, and that other antigens may be present within the adult worm.

[Alterations in collagen synthesis after transection of peripheral nerves].

In order to elucidate the mechanisms of fibrosis during Wallerian degeneration after peripheral nerve injuries, the biochemical and immunohistochemical changes in collagen from posterior tibial nerves of albino rabbits have been analyzed after transection of the sciatic nerves. The total amount of collagens in the posterior tibial nerve was found to increase progressively after 2 weeks post-neurectomy. The amount of total soluble collagen and the reducible cross-links were also increased as time advanced after surgery. There was no significant change in the synthesis of genetically distinct types of collagens after neurectomy. The immunohistological study showed that Type I and Type III collagens were increased remarkably in epi- and endoneurium. These findings indicate that the fibrosis of peripheral nerve during Wallerian degeneration is most likely due to the progressive formation of immature collagen fibers in the tissues, rather than the decrease in the amount of nerve proteins as previously discussed.

Application of the latex agglutination test for human paragonimiasis in South America.

The latex agglutination test (LAT) was compared with the complement fixation test (CFT), double diffusion test (DDT) and enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of South American paragonimiasis. Sera from 17 Peruvian and Ecuadorian patients with paragonimiasis, from 15 Japanese patients with other helminthiases, and from 100 healthy students were analyzed. The tests were also compared using serum samples from patients treated with bithionol. There was remarkable agreement among the methods and it was concluded that the LAT could be a useful serodiagnostic technique for paragonimiasis, especially in field surveys.

Development of spindle-shaped cells and chondroid cells from androgen-dependent Shionogi carcinoma 115. A light and electron microscopic study.

Androgen-dependent Shionogi carcinoma 115 (SC115) is an undifferentiated medullary carcinoma consisting of compact round cells. However, when host male DS mice were castrated 2 weeks after tumor transplantation, tumors composed of compact round cells, spindle-shaped cells and chondroid cells grew 4 weeks after castration. Compact round cells with desmosomes were arranged in solid nests and exhibited immunoreactivity for keratin protein. Spindle-shaped cells had prominent rough endoplasmic reticulum, and appeared to secrete collagen. Chondroid cells had the characteristics of chondrocytes. The light and electron microscopic features were highly suggestive of a transition from compact round cells to spindle-shaped cells, and from spindle-shaped cells to chondroid cells. The histology of this tumor thus suggests that SC115 cells are able to change into chondroid cells via spindle-shaped cells.

An evaluation of serum high density lipoproteins-phospholipids.

Phospholipids in high density lipoproteins (HDL) is being used as a negative risk indicator of atherosclerosis. Phospholipids in HDL may not demonstrate the actual level of HDL-phospholipids when determined by the precipitation or ultracentrifugal methods, because HDL fractions contain very high density lipoproteins (VHDL) and albumin. In the present study, the true level of phospholipids in HDL was estimated using high performance liquid chromatography (HPLC), and it was compared with the level of phospholipids in HDL determined by the precipitation method. Sera from 18 healthy subjects were used as materials. In the HPLC method, the HDL fraction was extracted making sure that it contained no free albumin, which is albumin not bound to phospholipids. The HDL fraction was separated into subfractions. It was found that phospholipids in the VHDL fraction make a 20.2 +/- 7.3% (mean +/- S.D.) part of the total HDL-phospholipids. A large part of the VHDL fraction was constituted of albumin-bound phospholipids. A significant correlation was observed between HDL-phospholipids determined by the precipitation method, which contain albumin, and the actual HDL fraction phospholipids determined by HPLC, which do not contain VHDL (r = 0.903, p less than 0.01). These results suggest that HDL-phospholipids values determined by the precipitation method give useful clinical data.

Eosinophilic leucocytes and arylsulfatase activity in bronchoalveolar lavage fluid of patients with bronchial asthma.

The arylsulfatase activity and histamine concentration of bronchoalveolar lavage fluid (BALF) were examined in patients with bronchial asthma in relation to the eosinophil count and asthma type (atopic and non-atopic). The BALF arylsulfatase activity and histamine concentration were significantly higher in atopic asthmatics than in non-atopic asthmatics. In atopic asthmatics, the activity of arylsulfatase was significantly increased in patients with a higher eosinophil count (10% or more). However, the BALF histamine concentration did not correlate with the eosinophil count. In non-atopic asthmatics, there was no significant correlation between arylsulfatase activity and the eosinophil count. The results show that arylsulfatase participates in IgE-mediated allergic reactions.

Immunochemical relationship of three antigens purified from Pasteurella multocida strain P-1059.

Three antigens were prepared from a type-3 avian strain of Pasteurella multocida, and their chemical and immunologic characteristics were studied. An antigen, designated 2.5S, was extracted with 2.5% NaCl solution and purified by chromatography. Lipopolysaccharide (LPS) was extracted with phenol-water, and a third antigen, designated FS, was extracted in 0.3% formalin solution containing 0.85% NaCl and purified by differential centrifugation. The 2.5S and the FS antigens consisted of 40% protein and 15% carbohydrate, whereas LPS did not contain a substantial amount of protein. A major protein component with a molecular weight of 44,000 was detected in the 2.5S antigen, as well as in the FS antigen. Of the 3 antigens, LPS had the highest activity in mouse lethality and Limulus lysate tests. Antigenic cross-reactions among the 3 antigens were demonstrated by immunodiffusion tests. The 2.5S antigen was indistinguishable from the FS antigen, as both antigens contained the LPS component of approximately 45%. Treatments with various reagents indicated that the 2.5S and FS antigens contained at least 2 antigenic determinants. The first was a heat-stable protein sensitive to protease or phenol-water, and the second was a periodate-sensitive carbohydrate, which was a major antigenic determinant on the LPS antigen.

Evaluation of relationship among three purified antigens from Pasteurella multocida strain P-1059 and of their protective capacities in turkeys.

Three antigens were prepared from Pasteurella multocida strain P-1059, and their immunogenicity and antigenic relationships were investigated. The 3 antigens were a soluble antigen purified from a 2.5% NaCl extract (2.5S), a similar antigen purified from an extract in 0.3% formalin solution containing 0.85% NaCl (FS), and lipopolysaccharide (LPS). The antigens were treated with various chemicals and enzymes to study their antigenic and immunogenic determinants. Antigenic analyses with ELISA inhibition tests indicated that 2.5S and FS were similar LPS-protein complex antigens. The 2.5S and FS antigens induced protective immunity in turkeys with high antibody titers against LPS antigen. Although LPS was a component of 2.5S and FS, LPS itself was poorly immunogenic in turkeys. The antigenicity of protein compounds in 2.5S was deteriorated by protease treatment, which, however, did not significantly diminish the protective immunogenicity. Treatment of 2.5S with sodium periodate, altering its carbohydrate moieties, decreased its immunogenicity. The immunogenicity of 2.5S also was abolished by phenol-water treatment, owing to dissociation of the LPS-protein complex. These findings suggest that a certain form of LPS-protein complex is essential for the induction of immunity against the P multocida infection in turkeys.
