[Comparative study of adrenaline-induced lipolysis of human deltoid and trochanter adipose tissue].

In vitro lipolysis by adrenaline, adrenaline + phentolamine (alpha-blocker) and isoprenalin (beta-agonist) was studied in deltoid and trochanter adipose tissues of sixteen obese females (10 android and 6 gynoid). Lipolysis differs in the upper and lower parts of the body. The more the obesity predominates in the upper part of the body, the more this difference is significant. If the actions of adrenaline in gynoid obese women are identical in deltoid and trochanter adipose tissues, the alpha-antilipolytic effect is more powerful than the beta-antilipolytic effect on the trochanter adipose tissue of android obese women.

Assay of blood and tissue aldehydes by HPLC analysis of their 2,4-dinitrophenylhydrazine adducts.

A new method for the assay of blood and tissue acetaldehyde is described. Samples are reacted with a methanolic solution of 2,4-dinitrophenylhydrazine (DNP) and the DNP-aldehyde adducts extracted into CHCl3. DNP-[14C]formaldehyde is added as internal standard. The CHCl3 extracts are washed with HCl and H2O and purified by aluminium oxide chromatography. The eluate is dried down, re-dissolved in methanol and subjected to quantitative analysis by HPLC. The acetaldehyde adduct was identified by co-chromatography with the authentic derivative and by mass spectrometry. Recoveries of added acetaldehyde were 85% and addition of 20 mmol ethanol to the sample gave no apparent increment in acetaldehyde content. This technique is suitable for assessment of acetaldehyde levels in clinical and experimental studies of ethanol metabolism.

A relationship between ascorbic acid and threonic acid in guinea-pigs.

Threonic acid is a major breakdown product of ascorbic acid used as a food additive. When administered orally to guinea-pigs (100 mg/kg body weight) for periods of 4 or 28 days, it produced a significant fall in the ascorbic acid concentration of certain organs but was without effect on other physiological and biochemical characteristics. The lifespan of scorbutic guinea-pigs was significantly reduced by dietary threonic acid (100 mg/kg body weight). The results indicate that threonic acid may modify the metabolism of ascorbic acid in guinea-pigs.

[HBs antigen in the blood donors of Calvados. Epidemiologic study and surveillance of the patients].

175 Chronic HBs Ag carriers have been discovered in the blood donors of the Calvados blood transfusion center from 1971 to 1979. 72 of them (41%) gave their consent for a clinical and biological study at the end of 1979, after receiving a convocation letter. This work had two aims: - to study the epidemiological factors in this population. - to evaluate the clinical and biological consequences of persistent antigenemia. 1. Epidemiological Study. Most results agree with the literature (higher prevalence in male, age, stay in endemic countries) but some results disagree for several reasons: our donors are all volunteers, HBs Ag prevalence is low in our region, most of the patients are caucasian and with life conditions and habits which may explain some particularities in contagion. Furthermore, the relative number of blood donors found every year as chronic HBs Ag carriers, does not increase in our country. 2. The Clinical and Biological follow-up of 62 HBs Ag carriers (alcoholics excluded) was carried out for 4,3 years on average. No patients developed clinical and biological features of chronic liver disease. After a mean term follow-up, we conclude that the asymptomatic HBs Ag carriers state seems not to be of bad prognostic. Since long term complications cannot be excluded, the follow-up of these patients must be maintained.

Effect of blackgram fiber (Phaseolus mungo) on hepatic hydroxymethylglutaryl-CoA reductase activity, cholesterogenesis and cholesterol degradation in rats.

Rats maintained on cholesterol-free diet and fed neutral detergent fiber (NDF) from blackgram showed lower concentrations of cholesterol in the serum, liver and aorta, when compared to those fed isocaloric, fiber-free diet. Incorporation of labeled precursors, [1,2(-14)C]acetate and [U-14C]glucose, into cholesterol of liver was higher in these rats. The activity of hepatic hydroxymethylglutaryl coenzyme A (HMG-CoA) reductase was also increased. Concentration of cholic acid and chenodeoxycholic acid in the liver was also more in rats fed blackgram NDF. The decreased concentration of cholesterol in the tissues may be due to its higher rate of hepatic degradation to bile acids.

Interpretation of the effects of propranolol on coronary reactive hyperaemia.

The effect of propranolol on the hyperaemic response after transient coronary arterial occlusion has been evaluated. The excess volume of blood delivered to the myocardium after an occlusion is diminished after propranolol in proportion to the reduced coronary blood flow. From the findings it is not necessary to postulate a change in the intrinsic mechanisms of reactive hyperaemia after propranolol has been given.

Ceftriaxone pharmacokinetics in newborn infants.

Ceftriaxone pharmacokinetics were determined in 40 newborn infants who were 1 to 45 days of age. Mean peak plasma concentrations of 136 to 173 micrograms/ml were observed at the completion of a 15-min intravenous infusion of 50 mg of ceftriaxone per kg. Mean half-life values were 5.2 to 8.4 h, and mean plasma clearances were 0.7 to 1.8 ml/min. Rectal swab cultures from 14 of 16 infants had either reduced numbers of aerobic and anaerobic bacteria or no growth during therapy. A once-daily dosage schedule is suggested for ceftriaxone therapy in newborn infants.

Flow cytometric studies of oxidative product formation by neutrophils: a graded response to membrane stimulation.

We have developed a quantitative assay to monitor the oxidative burst (H2O2 production) of polymorphonuclear leukocytes (PMNL) using single cell analysis by flow cytometry, and have examined whether PMNL respond to membrane stimulation with an all-or-none oxidative burst. During incubation with normal neutrophils, dichlorofluorescin diacetate diffused into the cells, was hydrolyzed to 2',7'-dichlorofluorescin (DCFH) and was thereby trapped within the cells. The intracellular DCFH, a nonfluorescent fluorescein analogue, was oxidized to highly fluorescent 2',7'-dichlorofluorescein (DCF) by PMNL stimulated by phorbol myristate acetate (PMA). That the oxidative product was DCF was shown by excitation/emission spectra and by mass spectrometry of the product from PMA-stimulated PMNL. Normal resting and PMA-stimulated PMNL oxidized 6.9 +/- 0.7 and 160 +/- 13 attomoles DCF per cell, respectively, in 15 min. Absence of calcium and magnesium ions and/or addition of 2 mM EDTA did not inhibit DCF formation by PMNL stimulated by 100 ng/ml PMA. Since EDTA prevented aggregation of PMNL (even when stimulated by 100 ng/ml PMA), which would prevent accurate flow cytometric analysis, further experiments were performed with EDTA in the medium. A close correlation between average DCFH oxidation and hexose monophosphate shunt stimulation was demonstrated using cells from patients whose PMNL had oxidative metabolic defects of varying severity. Intracellular DCFH was also oxidized by reagent H2O2 or oxygen derivatives generated by glucose oxidase + glucose or by xanthine oxidase + acetaldehyde; DCFH oxidation by these systems was inhibited by catalase but unchanged by superoxide dismutase. The data indicate that the DCFH oxidation assay is quantitatively related to the oxidative metabolic burst of PMNL, and they strongly suggest that the reaction is mediated by H2O2 generated by the PMNL. Incubation of PMNL with varying concentrations of PMA caused graded responses by all PMNL present; i.e., 1 ng/ml PMA caused a mean response of 34% maximal with a single population of responding PMNL (rather than 66% resting and 34% fully stimulated as predicted by the all-or-none hypothesis). Thus, with these assay conditions, oxidative product formation by PMNL occurs as a graded response to membrane stimulation by PMA.

Fructose: incomplete intestinal absorption in humans.

Fructose is an increasingly important commercial sweetener. However, some patients report abdominal symptoms after ingesting fructose-containing foods. The completeness of fructose absorption by the small intestine was assessed by breath hydrogen analysis in 16 healthy volunteers and incomplete absorption was defined as a peak rise in breath hydrogen of greater than 20 parts per million. Fructose, 50 g as a 10% solution, was incompletely absorbed in 6 of 16 subjects (37.5%). Incomplete absorption was associated with symptoms of cramps or diarrhea, or both in 5 of these 6 individuals. Incomplete absorption was both concentration- and dose-related. Three subjects incompletely absorbed 37.5 g of fructose. In comparison, all 15 subjects who were studied after ingestion of sucrose, 50 g and a 10% solution, completely absorbed this sugar load. Incomplete absorption of fructose should be considered as a possible case of gastrointestinal symptoms.

[Value of neuroleptoanalgesia in preventing recurrence of fetal shunts during treatment of diaphragmatic hernias].

A leptanalgesia (Chlorpromazine-morphine) was used in the treatment of postero lateral diaphragmatic hernia in six neonates. The treatment which was begun immediately after the arrival of the child and continued during and after surgery seemed to contribute to the favorable outcome observed in five of the babies. In spite of the empirical nature of their argument, the authors suggest the possible mechanism of action of the treatment. However they recognize that its effectiveness can only be assessed after careful hemodynamic studies.
