The laboratory diagnosis of Nairobi sheep disease.

The laboratory methods available for the isolation and identification of Nairobi sheep disease virus have been compared. The results show that inoculation of tissue culture (BHK 21 C 13) with suspensions of infected organs or plasma followed by fluorescent antibody tests on coverslip preparations gave the quickest means of identification. This test did not depend on the production of a cytopathic effect. Primary isolation of the virus in infant mouse brain and identification either by fluorescent antibody methods or by complement fixation with antigen prepared from the mouse brain offers a slightly more sensitive isolation system and would be recommended where no tissue culture facility exists.

An analysis of nucleotides and catecholamines in bovine medullary granules by anion exchange high pressure liquid chromatography and fluorescence. Evidence that most of the catecholamines in chromaffin granules are stored without associated ATP.

The catecholamine and nucleotide content of chromaffin granules from bovine adrenal medulla was analyzed both qualitatively and quantitatively. Nucleotides were examined by separation on anion exchange high pressure liquid chromatography and ultraviolet absorption, as well as by anion exchange thin-layer chromatography and luciferin-luciferase light emission analysis. CPATHECHOLAMINES WERE QUANTIFIED VIA AUTOMATED CONTINUOUS-FLOW ANALYSIS USING FLUORESCENCE. We found the average catecholamine/ATP ratio to be 8.2:1 with an SD of 1.98 and an SE of 0.50 for 16 separate glands. Relative percentages of epinephrine and norepinephrine were found to be 79.8 and 20.2, respectively. The granules contained 80.9% ATP, 12.3% GTP, and 6,8% UTP, as well as detectable amounts of ADP, AMP, GDP, and UDP. If we accept the catecholamine/ATP ratio of 4:1 proposed by Hilliarp earlier, then those catecholamines constituting the remainder of our ratio of 8.2:1 should represent a nucleotide-unassociated pool. In view of recent evidence of a direct agonistic function of nucleotides, we propose that the granule-associated nucleotides may act locally as coagonists with certain biogenic amines, and may additionally provide acirculating pool of purines and pyrimidines for use by the heart and lungs.

A weight illusion produced by lifting movements.

The perceived relative heaviness of objects was manipulated by instructing 13 subjects to lift them gently or vigorously, as was predicted by the motor theory of weight judgment and the known behavior of proprioceptors. The time-order error was shown to be significantly related to lifting movements.

Micromethods for induction and assay of mouse mixed lymphocyte reactions and cytotoxicity.

A new method in which induction of mixed lymphocyte responses and cytotoxicity can be performed under identical conditions is described. This enables the investigation of the relationship between MLR and cytotoxicity. A micro method for the 51Cr release assay for cytotoxicity is described using both mitogen-induced blasts and tumors as target cells. Under the conditions used data obtained is suitable for linear regression analysis, enabling quantitative comparisons to be made.

Immunological reactivity of B mice reconstituted with various numbers of syngeneic bone marrow cells.

Adult thymectomized mice were sublethally irradiated and reconstituted with various doses of syngeneic bone marrow. Immunological reactivity was assessed by allogeneic skin grafting at various intervals after reconstitution. It was found that reconstitution with high doses (1 X 10(7), 2 X 10(7)) of bone marrow was associated with an ability to reject allogeneic skin grafts and that this reactivity increased with time. Following rejection of a first skin graft, second grafts were rejected in an accelerated manner. These mice had measurable levels of Thy 1.2 positive cells in the spleen. In contrast, low-dose bone marrow reconstitution (1 X 10(6), 2 X10(6)) produced mice which did not reject either first or second allogeneic skin grafts, and such mice had at most only very low levels of Thy 1.2 positive cells in their spleens.

A case of drowning.

A case of near drowning is described. Despite full consciousness on early admission, and apparently good immediate recovery following intensive therapy for delayed pulmonary effects, the patient died of overwhelming necrotising pneumonia on the tenth day after the accident.

Serological identification of Ia antigens: report of a British region Ia workshop.

In preparation for the 7th International Histocompatibility Workshop 13 laboratories in the British Region participated in a local workshop. One hundred and twenty-three sera which had been previously shown to have activity on either normal B cells, CLL cells or B cell lymphoid lines in the absence of HLA-A, B or C activity were exchanged between the laboratories. These sera were tested on a total of 212 B cells, 101 CLL cells, 76 T cells and 76 lymphoid cell lines. The data was collected and analyzed in Oxford. The analysis showed that six groups of sera could be distinguished. When these groups were compared with the D locus typing of some of the lymphoid lines which were derived from individuals used as MLC typing cells, they were seen to have significant associations with D locus antigens. The serological groups defined were therefore given numbers corresponding to the D locus numbers they associate with, i.e. UK1 is associated with DW1 and so on for UK2, 3, 4, 5 and 7. Comparison of typing techniques showed that long incubation both with antiserum and then with complement, 1 hour + 2 hours gave the best and most reproducible reactions on normal B cells. Residual anti-HLA-A, B or C activity in some of the sera even after platelet absorption showed the importance of adequate checking on T cells after absorption.

Effects of L-tryptophan and L-methionine on activity in the rat.

1 The effects of an intraperitoneal dose of 20 mg/kg L-tryptophan and doses of 100 and 150 mg/kg of L-methionine were investigated on activity in the rat. 2 Activity was measured by a time sampling behaviour categorization procedure, and began 15 min after compound administration, lasting for a total of one hour. 3 Total active behaviour in the first half hour was reduced after 20 mg/kg tryptophan, 100 or 150 mg/kg methionine. Only 20 mg/kg tryptophan led to a significant reduction in active behaviour during the second half hour. 4 Administration of 100 mg/kg methionine plus 20 mg/kg tryptophan, and 150 mg/kg methionine plus 20 mg/kg tryptophan had no discernible effect on activity. 5 The results draw attention to the role of another amino acid in the modification of the behavioural effects of tryptophan.

Gentamicin dosage in children.

The results of 49 serum gentamicin assays from children of 2 weeks to 11 years of age are presented. The dose of gentamicin given 8-hourly intramuscularly ranged from 0-7 mg/kg-4-9 mg/kg. Doses of 1-7 mg/kg or less did not give adequate serum concentrations. As a result of these observations it is suggested that an intramuscular dose of 2-5 mg/kg given 8-hourly is suitable for most children in the absence of renal failure, but that adjustment of the dose according to serum gentamicin concentration is necessary.

Acute sedative properties of SKF 525A in rats: implications for its use as a metabolism inhibitor in the study of psychoactive drugs.

SKF 525A was found possess sedative properties in rats at doses of 25 and 50 mg per kg, when injected intraperitoneally. The behavioural effects of the drug were assessed in two ways. Firstly, by "Time Sampling Behavioural Categorisation" of explatory behaviour; and secondly by activity measurements obtained with an ultrasonic motion recorder. The results clearly demonstrate that SKF 525A has sedative properties in rats at doses which are conventionally used to inhibit metabolism of a wide range of drugs. The implications of these results for the use of SKF 5251 in the study of the actions of psychotropic drugs are discussed.
