Differential expression of two Xenopus c-myc proto-oncogenes during development.

Two distinct Xenopus c-myc cDNA clones have been characterized from an oocyte cDNA library. This allowed a comparison of the c-myc protein sequence across the vertebrate phylum to be made and prominent conservations to be identified. The majority of the sequence differences between the two Xenopus c-myc cDNAs are in the 5' and 3' untranslated regions. Sequence-specific oligonucleotide probes from the 5' untranslated region were used to demonstrate the differential expression of the two c-myc mRNAs during development. One of the mRNAs corresponds to the Xenopus c-myc gene previously reported expressed as a stable maternal mRNA uncoupled from cell division during oogenesis (c-myc I). It is the major mRNA species expressed during oogenesis and is expressed again from the zygotic genome in post-gastrula embryos. In contrast, the second c-myc mRNA (c-myc II) is expressed only from the maternal genome during oogenesis. Primer extension experiments show that in the oocyte the transcriptional initiation sites for c-myc I and c-myc II are at different distances from the translational start site. The 'oocyte-specific' and 'somatic-type' developmental regulation of c-myc is reminiscent of polymerase III 5S RNA gene expression in Xenopus, and may provide new insights into the developmental regulation of genes transcribed by RNA polymerase II.

Routine cleaning and the elimination of Campylobacter pylori from endoscopic biopsy forceps.

The effect of routine cleaning in removing Campylobacter pylori from the biopsy forceps of endoscopes has been examined in a series of 50 patients. Campylobacter pylori was isolated from the biopsies of 15 of the patients, while one of the 50 biopsy forceps washings yielded the organism after routine cleaning. This study suggests that there is a small chance of transmitting C. pylori by endoscopic equipment if cleaning is the only method of decontamination adopted.

Congenital mesoblastic nephroma, renin production, and hypertension.

Between 1964 and 1987, 12 patients with mesoblastic nephroma were treated. The BP was measured preoperatively in five patients, four of whom were hypertensive. Following nephrectomy, the BP returned to normal. Plasma renin levels were measured in one patient; they were markedly elevated pre-operatively, but returned to normal following tumour excision. Immunoreactive renin staining, using a polyclonal antibody to human renin, was performed in the 12 patients. Staining was positive in ten patients. The most intense staining was noted in the areas of cortex entrapped within the tumour, and in perivascular spaces not associated with entrapped cortex. These findings suggest that hypertension secondary to increased tumour associated renin production is a feature of congenital mesoblastic nephroma.

Endotoxin induces bacterial translocation and increases xanthine oxidase activity.

Previously, we documented that endotoxin induces bacterial translocation from the gut and that inhibition or inactivation of xanthine oxidase activity reduces endotoxin-induced bacterial translocation. Consequently, experiments were performed to correlate endotoxin-induced bacterial translocation with changes in intestinal mucosal structure and xanthine dehydrogenase and oxidase activity. Segments of the jejunum, ileum, cecum, proximal colon, distal colon, and liver were harvested from ICR mice 24 hr after IP administration of E. coli 0111:B4 endotoxin (0.1 mg). Xanthine dehydrogenase and oxidase activities were measured in these samples and correlated with intestinal morphology. Bacteria translocated from the intestines to extraintestinal organs in 70% of the mice receiving endotoxin, while the organs of control mice were sterile (p less than 0.01). Endotoxin injured primarily the ileal and cecal mucosa and increased ileal and hepatic xanthine dehydrogenase and cecal oxidase activities (p less than 0.05). These results suggest that xanthine oxidase-induced mucosal damage plays a role in endotoxin-induced bacterial translocation.

Muscle-specific (CArG) and serum-responsive (SRE) promoter elements are functionally interchangeable in Xenopus embryos and mouse fibroblasts.

The Xenopus cardiac actin gene contains four copies of a promoter element, the CArG box, which is conserved amongst striated muscle actin genes and is essential for tissue-specific expression in the developing Xenopus embryo. Our aim is to identify embryo and muscle proteins that interact with the CArG box as a step towards understanding the molecular basis of this developmentally regulated gene expression. The CArG box shares some sequence similarity with the Serum Response Element (SRE), which mediates the transcriptional activation by serum of genes such as c-fos and cytoskeletal actin. We show here that the most proximal cardiac actin CArG box is recognized by the same binding activity as the cytoskeletal actin SRE in nuclear extracts from both Xenopus embryos and mammalian muscle cells. This activity is indistinguishable from the previously characterized HeLa cell SRE-binding activity, Serum Response Factor (SRF). Importantly, we extend these in vitro studies to demonstrate that the CArG box and SRE are functionally interchangeable, both in Xenopus embryos and mouse fibroblasts. This implies that the CArG box and SRE can bind the same protein in vivo, as well as in vitro. Our results identify an SRF-like protein as a CArG box-binding factor and we discuss the implication that a common mechanism may be utilized in both muscle-specific gene expression and serum-responsive transcription.

Renin messenger RNA localization in congenital mesoblastic nephroma using in situ hybridization.

Renin messenger (m) RNA distribution was studied in congenital mesoblastic nephroma, a usually benign renal tumour of early infancy which may be associated with excess renin production and hypertension. Using in situ hybridization with synthetic radiolabelled oligonucleotide probes combined with immunohistochemical studies, renin expression was found in areas of tumours containing recognizable cortical structures including glomeruli and tubules. Renin mRNA was also detected in vessels and larger vascular spaces within the tumour not associated with cortical structures. Cells in the tumour vessel walls and sinusoids which expressed renin also stained positively for vascular smooth muscle-specific alpha actin.

Incorporating new words into the lexicon: preliminary evidence for language hierarchies in two-year-old children.

In 4 experiments, we examined how young children incorporate new word meanings into their lexicons. 2-year-olds were each taught a new noun for an object that already had a known label (e.g., a "fep" for a dog). Children's interpretations of the new nouns were assessed by asking subjects to select the named toy from an array of 4 toys (e.g., "Point to a fep"). The experiments were designed to determine which of several possible semantic relations between novel and familiar words was most consistent with children's performance. It was found that children often seemed to interpret the new word as referring to a subordinate of the known category. This tendency was reduced when the named object could sensibly receive a proper name (e.g., when the named object was a stuffed animal), particularly when children had to consider both the familiar and the novel label for the object in the same session. Although not all alternative explanations have been ruled out, these results suggest that, from a very young age, children may spontaneously form language hierarchies when they hear a novel work for an object that already has a familiar name.

Cytogenetic and chemical detection of human exposure to polyhalogenated aromatic hydrocarbons.

Peripheral lymphocytes from Taiwanese women (n = 35) exposed to polychlorinated aromatic hydrocarbons and from matched controls (n = 24) were assessed for the levels of sister chromatid exchanges (SCEs) after a 72-hour incubation of whole blood in the presence or absence of alpha-naphthoflavone (ANF) and for chromosome aberrations after 48 hours of incubation. Serum levels of polychlorinated biphenyl (PCB) congeners were measured for all individuals, and serum levels of several polychlorinated dibenzofurans (PCDFs) were measured for 12 exposed individuals by gas chromatography-mass spectometry. Blood concentrations of total PCBs in the exposed population averaged approximately 15 ppb, whereas mean PCDF values were 14 ppt. Major PCB congeners detected were 2,2' 4,4', 5,5'-hexa CB and 2,2'3,4,4',5-hexa CB. PCDFs detected were primarily 1,2,3,4,7,8,-hexachlorodibenzofuran (10.8 ppt) and 2,3,4,7,8-pentachlorodibenzofuran (2.7 ppt). Average SCE frequencies were 7.61 for controls and 7.30 for exposed individuals when assays were conducted in the absence of ANF, whereas respective values were 8.85 and 10.75 in the presence of ANF. Differences in the level of ANF-induced SCEs between the two populations were highly significant (P less than .001). Moreover, the ANF-induced SCEs were highly correlated with the serum concentrations of total PCBs and of several PCB congeners (P less than .001). Increases in ANF-induced SCEs appeared to be linear up to a PCB concentration of approximately 30 ppb. Chromosome aberration frequencies were similar in control and exposed populations. These studies demonstrate that in vivo exposure to PCBs and PCDFs result in an enhanced sensitivity of lymphocytes to the SCE-causing actions of ANF.

A study of the value of electrophoretic and other techniques for typing Acinetobacter calcoaceticus.

Forty-four isolates of Acinetobacter calcoaceticus var anitratus collected during hospital outbreaks were studied using polyacrylamide gel electrophoresis (PAGE), plasmid analysis, antibiograms and biochemical tests to determine their degree of similarity. Reproducibility tests were also carried out on the PAGE and biochemical techniques to determine their validity when used to compare bacteria of the same type isolated intermittently. PAGE data was analysed densitometrically and isolates compared using a similarity matrix. All methods were able to subdivide the isolates, but results did not always correlate well between methods. Reproducibility data indicated that careful attention to technique is required when organisms are examined by PAGE sequentially. Results suggest that no single biotyping technique is likely to be adequate and that electrophoretic, biochemical and antibiogram data may complement one another and other epidemiological data in the typing of these organisms.

Neurotoxicity associated with deferoxamine therapy.

We have documented visual and auditory neurotoxicity in 42 of 89 patients with transfusion-dependent anemia who were receiving iron chelation therapy with subcutaneous deferoxamine (DFO). Of the affected groups, 13 presented with visual loss or deafness or both, and ophthalmologic, audiologic, and visual evoked potential studies (VEPs) uncovered abnormalities in 29 more. Four patients with visual loss had optic neuropathy with a marked decrease in acuity and loss of color vision. These 4, and 16 other asymptomatic patients, had abnormal VEPs. When DFO was stopped, 3 of 4 with visual problems regained normal visual function but VEPs remained abnormal. Of the other 16 with abnormal VEPs, 9 became normal or improved and 7 did not change; on restarting DFO, the 9 became abnormal again. There were 22 abnormal audiograms that showed a high-frequency sensorineural deficit; 13 patients were symptomatic and 4 needed hearing aids. On stopping DFO, 9 became asymptomatic but 15 audiograms remained abnormal and 2 deteriorated further on restarting the drug. An analysis of the clinical data showed that members of the affected group were younger, had lower serum ferritin values, and were self-administering higher doses of DFO/kg body weight. Significantly lower doses of DFO were being taken by patients without abnormalities than by those with visual symptoms, abnormal audiograms, or prolonged VEPs (P less than 0.001, less than 0.006, and less than 0.04, respectively). The data implicate high-dose DFO as a central factor in the pathogenesis of the neurotoxicity. Our serial studies provide the basis for effective yet safe DFO administration for patients who require the agent.
