Metabolic detoxication of bis-(2-hydroxy-3, 5-dichlorophenyl)-sulfoxide in man.

The metabolic detoxication of bis(2-hydroxy-3, 5-dichlorophenyl)sulfoxide (BTS) in man was investigated. Bis(2-hydroxy-3, 5-dichlorophenyl)sulfide (BT) was identified in beta-glucuronidase treated urine following the administration of BTS by thin-layer chromatography, gas chromatography, ultraviolet spectrum and quantitative analysis. No other metabolites were detectable. BT-glucuronide was also identified in urine. It was assumed that BTS was reduced to BT and successively conjugated with glucuronide in man, and excreted as BT-glucuronide in the urine.

Is there any interaction between cadmium and ouabain effects on Na transport?

Dermally applied ouabain decreased the short circuit current (SCC) in frog skin, whereas epidermal cadmium increased it. The percent increase in SCC induced by Cd2+ was never modified by the presence of ouabain. This suggests that Cd2+ and ouabain may exert their effects on Na transport independently.

Interaction of cadmium, calcium, and amiloride in the kinetics of active sodium transport through frog skin.

The epidermal effects of activator (cadmium) or inhibitors (calcium and amiloride) as well as their interactions in active Na transport through bullfrog abdominal skin were analyzed kinetically. Cadmium at 1 mM increased the short circuit (SCC) to 126%. The Hill coefficient (n) for the relation between [Cd2+] and SCC was 0.737, which indicated negative cooperation. Calcium at over 8 mM decreased the SCC to 80% at its maximum effect. Calcium inhibited the Cd2+-induced SCC at lower [Cd2+], and facilitated it at higher [Cd2+]. Calcium acted uncompetitively on the binding reaction of Na+ with a Na entry channel. Cd2+ alone or Cd2+ under the influence of Calcium also acted uncompetitively on the reaction. Amiloride at 5 X 10-5 M inhibited the SCC almost completely. The Hill coefficient in the relation between amiloride and SCC was 0.757, indicating the presence of negative cooperation. In contrast to the effect of calcium, the Cd2+-induced SCC was much reduced at higher [amiloride]. Amiloride inhibited the channel-Na binding reaction in a mixed manner (competitive + noncompetitive), while Cd2+ activated the reaction under the influence of amiloride noncompetitively. From the SCC model based on the hypothesis that the kinetic constants are invariable even under interaction of the activator and inhibitors, the percentage of the various forms of channel.ion complex were calculated. The SCC was then calculated, assuming that is was directly proportional to the sum of the rate of decomposition of channel.Na complex and the higher rate of decomposition of channel.Na.Cdn complex. The above assumption is supported by the fact that there is good agreement between the calculated SCC and experimentally obtained SCC in the mixture of the activator and the inhibitor.

Epstein-Barr virus-induced lymphoblastoid cell lines from seropositive adult donors and their HLA-antigens.

Epstein-Barr virus (EBV)-induced lymphoblastoid cell lines (LCL) were established from seropositive adult Japanese donors. HLA-A, -B, -C and -DR antigens expressed on these EBV-LCL cells were identical to those of peripheral blood lymphocytes. There were no extra reactions upon DR typing; the extra reactivity in A, B and C typing was due to contaminating DR antibodies in the typing sera. In one-way mixed lymphocyte reactions with a stimulator : responder ratio between 1 : 10 and 1 : 20, autologous EBV-LCL exhibited low stimulation which could be distinguished from that of unrelated combinations. EBV-LCL may be useful for the screening of anti-DR antisera and as targets in cell-mediated lympholysis. Furthermore, EBV-LCL established from homozygous typing cells may be useful in HLA-D typing.

An animal model system for the immunotherapy of lung carcinoma using sensitized lymphocytes: a preliminary study.

An animal model system was established for immunotherapy of lung cancer using line-1 lung carcinoma in Balb/cR mice and xenogeneic lymphocytes from New Zealand white (N2W) rabbits. The immunization schedule consisted of footpad and subcutaneous injections of 1 X 10(8) viable line-1 cells into two rabbits, twice at a weekly interval. Two other rabbits were maintained as source of unsensitized lymphocytes. All rabbits were bled on day 14 at which time 45 mice were divided into three groups of 15 for both transplantation of 1 X 10(4) viable line-1 cell (subcutaneously) and treatment with 1) control saline, 2) 2.5 X 10(6) unsensitized lymphocytes, and 3) 2.5 X 10(6) sensitized lymphocytes. These treatments were repeated twice at weekly intervals. Results were evaluated four weeks after transplant and initial treatment. Tumor developed in 13/15 control mice, 15/15 treated with unsensitized lymphocytes, and 7/15 with sensitized-lymphocyte injection. The mean tumor weight for group 1 (control mice) was 1.38 +/- 0.22 gm (SEM), group 2 was 2.09 +/- 0.25 gm, and 0.56 +/- 0.28 gm for group 3. There was a statistically significant difference between control and group 3 for both tumor appearance (P = 0.014) and tumor weight (P = 0.026).

Cranioplasty utilizing a preserved autogenous bone flap coated with acrylic resin.

Cranioplasty utilizing a resin-coated preserved autogenous skull flap was devised and used in 36 cases, 14 of them being followed up radiologically. It has been confirmed that this method is advantageous in that it repairs the skull defect satisfactorily both from surgical and cosmetic points of view. Furthermore this method causes no anxiety to the patient because of the autogenous origin of the bone flap. We recommend this method for cranioplasty when an autogenous bone flap is preserved.

Effect of cadmium on active sodium transport by the abdominal skin and the isolated epidermis of the bullfrog: differences in effects between epidermal and dermal cadmium applications.

Epidermal application of 2 mM-cadmium (Cd2+) to the SO42---Ringer's solution bathing the whole frog skin increased its short circuit current (SCC) significantly, whereas the dermal application did not, in most cases. When Cd2+ was applied to the epidermal side, the uptake of cadmium by the skin was approximately 150 microM/kg wet weight, but the uptake by the skin from the dermal side was about eight times as large. In the isolated epidermis, the epidermal application of Cd2+ increased the SCC, whereas the dermal application decreased it. The amount of cadmium taken up by the isolated epidermis was approximately the same when the cation was applied to either epidermal or dermal side. These seemingly contradictory results can probably be explained by the barrier-hypothesis which is supported by the results of an electron probe X-ray microanalysis. In the case of the epidermal application, the junction between s. corneum and s. granulosum may act as an outside barrier for cadmium translocation. For the dermal application of Cd2+, the sieve layer in the dermis, where a large amount of calcium is deposited, may probably exert its effects as an inside barrier by the mechanism of Cd-Ca exchange process. Tela subcutanea, the basement membrane and its adjacent tissue may be also inside barriers, though their role as a diffusion barrier may be considered to be weaker than that of the sieve layer.

Biosynthesis of leupeptin. IV. Is protein turnover in leupeptin producer cells affected by leupeptin?

There was no significant difference between the rates of protein degradation in cells of a leupeptin-producing strain and that of a leupeptin-nonproducing strain, the latter being derived from the former by mutation. Protein autodigestion in a cell homogenate of the leupeptin producer was sensitive to EDTA and chymotrypsin and less sensitive to leupeptin. On the contrary, protein degradation caused by exogenous trypsin in a similar homogenate was highly sensitive to leupeptin. A labeling experiment with [14C]-arginine of a culture of the leupeptin producer strain revealed that leupeptin was accumulated mostly in the medium and only slightly in the cells; the ratio between the amount in the medium and that in the cells was about 250:1. In contrast, leupeptin acid, the proximal intermediate having no antiplasmin activity, showed a ratio of 5:1.

Lymphoproliferative response to membrane extracts of nasopharyngeal carcinoma.

The tumor-associated antigen (TAA) was extracted from cultured cells obtained from nasopharyngeal carcinoma (NPC), and the specific immune reaction was studied by utilizing the proliferation reaction of lymphocytes from NPC patients. The following results were obtained. (1) The stimulation of lymphocytes by membrane soluble antigen (MS-Ag) was significantly different in the group of patients with NPC and the control groups (patients with malignant tumors of the head and neck and healthy individuals). Lymphocytes from the NPC patients gave a significantly higher stimulation index (SI) value with NPC MS-Ag, but a lower value with PHA, as compared with those of normal controls. (2) No definite correlation was observed in anti-Epstein-Barr virus (EBV) antibody titers. (3) As for the correlation with various histological types, apparently enhanced reaction was noted irrespective of the histological type of the cancer. (4) In connection with stage classification, the reaction was always negative in patients in stage IV. A particularly close correlation was noted between the degree of the response and the N classification. Some inhibitory factor affecting the LP reaction might appear with the progress of N and M.
