Purification and properties of chromosomally mediated beta-lactamase from Citrobacter freundii GN7391.

Both a penicillinase and a cephalosporinase are present in a strain of Citrobacter freundii (GN7391) resistant to beta-lactam antibiotics. The penicillinase was identical to the type Ia penicillinases (Type III by Richmond classification), mediated by Rms212 and R-TEM. A cephalosporinase, typical of enterobacteriaceae chromosomal beta-lactamase (Type I by Richmond classification), was purified from the strain. It gave a single protein band on polyacrylamide gel electrophoresis and immunoelectrophoresis; the pI was 8.6 and its molecular weight was approximately 38 000. Cysteine was not found among its amino acids. The specific activity was 388 units (mg protein)-1 for the hydrolysis of cephaloridine, and the optimal pH was 8.0. Rabbit antiserum obtained against the purified enzyme showed cross-reaction with cephalosporinases produced by strains of Enterobacter cloacae in a neutralization test.

An ultrastructural study on the interaction of Mycoplasma gallisepticum with the chicken tracheal epithelium.

Seven-day-old chickens wee intratracheally inoculated with Mycoplasma gallisepticum. The tracheas collected 6 and 14 days after chickens were inoculated were subjected to titration of mycoplasma and examination by light and electron microscopy. The mycoplasma organisms grew well; 10(7) to 10(8) color-changing units in a milligram of tissue were determined. Tracheal lesions occurred in close association with the presence of mycoplasmas and were characterized by degeneration of the epithelial cells and inflammatory cellular infiltration of the mucosa. Mycoplasmas were predominantly found extracellularly and only rarely in phagocytic vacuoles of the epithelial cells. Although the mycoplasmas exhibited considerable pleomorphism in size and shape, most of them were oval or round, and the largest diameters were between 300 and 700 mn. Elongated and irregular forms were also observed, particularly in those mycoplasmas adhering to the epithelial cells. The organism had a limiting unit membrane, the fibrillar nuclear area, the peripheral cytoplasmic area containing numerous ribosomes, and a terminal bleb structure. Mycoplasmas attached to the epithelial cells by their blebs close to the host cell membrane. At the attachment site, neither fusion of the membranes of the mycoplasma and host cell nor injury to the host cell membrane could be demonstrated. Nevertheless, seemingly, the intimate association between the adhering mycoplasmas and the epithelial cells might be an important factor in pathogenesis of the disease.

[Craniolacunia in newborns with myelomeningocele and encephalocele (author's transl)].

Craniolacunia is characterized by clusters of deep pits with steep edges in the cranial vault of newborn infants. And also craniolacunia is anomaly of the skull of the newborn infants usually associated with spinal meningocele, myelomeningocele and occasionally with encephalocele. The purpose of this paper is to report the results of the skull roentgenologic study of 34 newborn infants with myelomeningocele and 11 newborns with encephalocele who were treated within 7 days after birth, and, to discuss the correlations with the head circumference, level of myelomeningocele, location of encephalocele, Arnold-Chiari malformation and hydrocephalus. Roentgenograms of the skull were obtained at the time of admission and were examined regarding the presence and the degree of craniolacunia. The radiographic changes were classified according to the extension of the craniolacunia, from grade 0 to grade 3. Among 34 cases of myelomeningocele, 28 cases (82%) were observed to have craniolacunia and among 11 cases of encephalocele, 4 cases (36%) had findings of craniolacunia. In our cases, the coincidence of craniolacunia with myelomeningocele was 82% and this is higher than in previous reports. But the coincidence of craniolacunia with encephalocele was same as in previous reports. There has been no report about the incidence of craniolacunia within 7 days of life.

Craniolacunia in newborn with myelomeningocele.

Plain skull films were taken within 7 days of birth in 34 newborn with myelomeningocele. There appears to be a correlation between hydrocephalus or Arnold-Chiari malformation and the severity of craniolacunia. Craniolacunia was observed in 28 cases (82%). The incidence of craniolacunia in myelomeningocele in this series is higher than in previous reports. One of the reasons is that plain skull films of newborn with myelomeningocele are usually not taken early at birth. We would like to emphasize that plain skull films be taken early at birth in newborn with myelomeningocele and that the presence of craniolacunia be considered as being associated with hydrocephalus and the Arnold-Chiari malformation.

Role of calcium ions in the thermostability of thermolysin and Bacillus subtilis var. amylosacchariticus neutral protease.

The stabilizing effect of calcium ions on thermolysin and Bacillus subtilis var. amylosacchariticus neutral protease has been investigated. Calcium and zinc ions were removed from the proteases by gel filtration over Sephadex G-25 equilibrated with metal chelating agents. Using these enzymes with different metal content, heat inactivation kinetics were studied at various temperatures. Removal of calcium ions caused a sharp decrease in thermostability and diminished the values of the activation enthalpy (deltaH*) and entropy (deltaS*) for heat inactivation. There was little difference in stability between thermolysin containing 0.3 g-atom/mol and B. subtilis neutral protease containing 1.4 g-atoms/mol. Calcium binding isotherms of the proteases were obtained by equilibrium gel chromatography with various concentrations of free calcium ions. Thermolysin had four independent calcium binding sites with an identical intrinsic binding constant (K) of 2.0 X 10(4) M-1. B. subtilis neutral protease had four independent sites. The K value for three sites was 1.1 X 10(5) M-1 and the binding constant for the other site was 1.5 X 10(3) M-1. There was little difference in total free energy change for calcium binding between these proteases. From these results it is concluded that the stabilizing effect of calcium on these enzymes is almost equal, and the extra thermal stability of thermolysin is likely to come from its polypeptide chain structure.
