Morphological studies of the lungs in chronic hypobaric hypoxia.

The aim of the present study was to evaluate the effect of hypobaric hypoxia on the changes observed within lung parenchyma, particularly in the surfactant system-forming structures. The experiment used male Wistar rats, 180-220g b.w. The animals were kept in a hypobaric chamber for 3, 10 and 30 days. Pressure in the chamber (380mm Hg) corresponded to air rarefaction at an altitude of 5500m. Control animals were kept in the same room near the chamber at normal atmospheric pressure. The changes observed in the lungs were evaluated basing on ultrastructural analysis by transmission electron microscopy. The present results indicate phasic character of the changes occurring within lung parenchyma, including the surfactant system-forming structures. In the early phase (3 days), destructive-exudative changes predominated (delamelation of type II pneumocytes; oedematous changes, damage to the alveolar lining layer, accumulation of alveolar macrophages). In the later phases, repair-proliferative processes were predominant (increased number of type II pneumocytes, focal intensification of fibroplasia). The changes within the surfactant system-forming structures were accompanied by the accumulation of granulocytes and monocytes (hypobaric conditions for 3 and 10 days) and blood platelets (hypobaric conditions for 30 days) in the vascular bed of the lungs. The pathomechanism of the changes observed in the surfactant system was discussed with regard to the vascular changes. The possibility of formation of blood platelets within the vascular system of the lungs was considered.

Pathological changes in rats hepatocytes on acute lindane poisoning.

Histological, histochemical and ultrastructural examinations of the liver were carried out on rats. The animals were given crystalline lindane intragastrically, in a dose evoking acute intoxication, (1/3 LD50). Then the rats were decapited after 1, 3, 10, 20 and 36 days following pesticide administration. The obtained results indicate, that acute lindane intoxication produces morphological changes in the rat liver, which evidence a disturbance inenergetic processes of hepatocytes and they are reversible and adaptative.

Primary intracranial malignant fibrous histiocytoma--ultrastructural study.

Malignant Fibrous Histiocytoma is the most common soft tissue sarcoma in adults with the majority of cases that occur in patients between 50-70 years and 64% of cases occurs in male. Most commonly it arises in extremities (lower 49% and upper 19%), less often in the abdomen (16%) and very rare in head localization (3%) [5]. Typically macroscopically it is a multilobulated, gray-white, fleshy mass between 5-20 cm of diameter. In light microscopy this tumour has a highly variable morphologic pattern and manifests a broad range of histological appearance. It has been classified into the following subtypes: storiform-pleomorphic, myxoid, giant cell, inflammatory and angiomatoid. The first two subtypes are more frequent than others. The aim of this report is to present the electronmicroscopic features of giant cell type of MFH localized in head.

Cyclophosphamide in diffuse lung damage.

Some cyclophosphamide toxic effects on lung tissue are presented. Cyclophosphamide metabolism, pathogenesis of lung damage and morphological lung tissue changes caused by that agent were characterized. Attention was focused on BAL evaluation as a useful method in the monitoring of lung tissue damage degree.

Effects of pentoxifylline on some peripheral blood parameters and haemostasis in acute pulmonary tissue injury after cyclophosphamide.

Cyclophosphamide (CP) is one of the widely used cytostatic drugs, with a strong toxic influence on pulmonary tissue. Experimental works have shown that a single high dose of CP causes injury to all elements of the interalveolar septum, especially to the alveolar endothelial and epithelial cells. The aim of this work was to evaluate the influence of pentoxifylline (PTXF) (30 mg/kg b.w.) on the ultrastructure of lungs capillaries and blood cell count as well as plasma fibrinogen levels in Wistar rats after intraperitoneal injection of 150 mg/b.w. CP. We established that in the doses applied PTXF had no statistically significant influence on the number of leukocytes and erythrocytes determined in the left ventricular blood of rats receiving CP, while the number of white cells from animals given PTXF only was higher than in controls. There was a smaller decrease in the number of platelets (p < 0.05) and smaller reduction in fibrinogen level (p < 0.01) in the serum of PTXF-CP animals than in the CP group. The results obtained suggest a protective effect of PTXF on CP induced changes, which have been evidenced in some of the parameters examined. Ultrastructural examinations found the lungs to be the organ of extramedullary thrombocytopoiesis in CP-treated animals and revealed that platelet accumulation in the system of lung capillaries was a potential cause of the decrease observed in the number of blood platelets following CP administration.

Microenvironment of Morris hepatoma 5123 metastases to the lungs--ultrastructural analysis of intratumour injections of rhTNF-alpha.

We analysed the effect of rhTNF-alpha on the development of Morris hepatoma 5123 spontaneous metastases to the lungs of Buffalo rats. After intratumour administration of the cytokine the lungs were found to contain scarce metastatic foci. In precapillaries and capillaries neoplastic cells were single or in small clusters, without contact with endothelial cells. Endothelial cells showed high pinocytic activity and frequently tightly closed vascular lumen. The stroma of interalveolar septa and the capillary bed contained multiple eosinophils. Moreover, eosinophils, histiocytes and monocytes mixed with abundant collagen fibres were found on the margin of single small metastatic foci and among necrotic neoplastic cells.

Ultrastructure of bone marrow megakaryocytes in experimental haemorrhagic shock in rats. I. Correlation between ultrastructure of MK nuclei and DNA ploidy.

The correlation was described between the ultrastructural picture and DNA mass content of MK nuclei in experimental haemorrhagic shock in rats. Significant disproportions were revealed between the morphological pictures and the ploidy of MK nuclei in the successive phases of the shock. Abnormalities of the maturation of marrow MK nuclei were found in the first hours of the shock, being most pronounced in the 24th hour.

Ultrastructure of bone marrow megakaryocytes in experimental haemorrhagic shock in rats. II. Correlation between ultrastructure of MK cytoplasm and thrombopoiesis.

Ultrastructural evaluation was performed of the MK cytoplasm in the successive phases of experimental haemorrhagic shock in rats. Qualitative abnormalities were revealed in the intracytoplasmic structures of MK. Platelets in the circulating blood were evaluated. A correlation was found between the changes in the intracytoplasmic organelles and the number and biological activity of blood cells. The results obtained indicate significant disturbances in the functioning of the thrombopoietic system in experimental haemorrhagic shock, which are caused by alterations within the intracytoplasmic structures of MK and lead to platelet production from morphologically differentiated but functionally immature MK.

A comparison of the peritoneal cell population of pregnant rabbits after LPS or TNF-alpha induced septic shock.

Septic shock is a catastrophic consequence of invasive infection. Unfortunately, recent advances in surgical and medical sciences have not significantly reduced the overall mortality from septic shock. Bacterial antigens stimulate a cascade of cytokine release; each cytokine helps the host to overcome infection, but their excessive production causes them to trigger events that lead to septic syndrome and shock. Tumour necrosis factor (TNF-alpha) has a pivotal role in orchestrating the events leading to septic shock. Intraperitoneal administration of certain substances can increase the number and phagocytic activity of cells, which reach naturally the site of infection. Activation of the immunity cells in the peritoneal cavity and their immunocompetence are found to be responsible for the organism protection against abdominal cavity infections. Macrophages, lymphocytes and granulocytes of low activity in the non-stimulated peritoneal cavity become significant due to the influence of numerous biologically active substances. This study was designed to determine the peritoneal response to local administration of LPS or TNF-alpha in the course of experimental septic shock.

Changes of myocardial capillary density in progression of experimental lung emphysema.

A morphometrical analysis of changes in the heart of rats with experimental lung emphysema due to single intratracheal administration of papain solution has been done. Special attention has been paid to quantitative changes of capillaries and to the cardiomyocyte diameter in respective regions of the myocardium after one, three and six months following the administration of papain. A histological and morphometrical analysis of changes in the lungs has also been carried out and partial pressures of oxygen and carbon dioxide in the arterial blood of the examined animals have been marked. It has been found that a single administration of papain caused emphysematous changes in the lungs, growing more intense until the third month of the experiment. The progression of these changes corresponded to a capillary density increase in the right ventricle, interventricular septum and in the subendocardial layer of the left ventricle. The increase in the number of capillaries was interpreted as the first adaptive stage of cor pulmonale development and explained by the occurrence of angiogenesis. The changes co-existed with a decrease in partial pressure of oxygen in the arterial blood. After six months of the experiment, a reduction in capillary density in the heart regions mentioned above and a simultaneous increase in the cardiomycyte transversal diameter were observed, being the exponents of evident myocardial hypertrophy. Development of ultrastructural changes in the hearts of rats has been analysed too. Simultaneous development of changes in the right ventricle muscle and in the subendocardial layer of the heart left ventricle myocardium was observed. The role of hypoxia as a one of essential factors responsible for the observed morphologic changes has been discussed.

Type II alveolar epithelial cells and free alveolar cells after intratumor TNF-alpha administration.

The experiment used Morris hepatoma 5123 series growing in muscles of the Buffalo rats. A suspension of 3 x 10(6) neoplastic cells was injected into the right hind leg of the animals. After fourteen days, TNF-alpha was administered into the tumour in a dose of 1.5 x 10(4) U/24 hours in 0.5 ml PBS solution. The group I animals were injected for 4 days and group II for 8 days. Control groups consisted of rats with injected Morris hepatoma which were given PBS solution instead of TNF-alpha (group III A and B) and animals without the hepatoma, given 4 or 8 TNF-alpha, respectively (groups IV A and B). In the present study, we have explored the effect of intratumor TNF-alpha administration on the composition of cells isolated from the lungs through multiple bronchoalveolar lavages (BAL). Ultrastructural evaluation of the pulmonary tissue was done using a transmission electron microscope (TEM), with special attention paid to type II alveolar epithelial cells and free alveolar cells. Examinations in TEM in groups I, II and IV (A and B) found, in the lumen of alveoli, an increase in the number of alveolar macrophages (AM) with morphological features of intensified activity and AM with numerous secondary lysosomes containing material of phospholipid structure. Also, numerous type II alveolar epithelial cells with emptied lamellar bodies were observed. The above mentioned changes were especially marked after eightfold TNF-alpha administration. In groups I, II and IV (A and B), compared with group III, a significant increase was found in the total number of cells isolated by BAL as well as in the number of cells with positive reaction in staining according to Beckstead's method. It may indicate that the changes in the parameters mentioned above are related to TNF-alpha action. The results obtained indicate the possibility of systemic effect of TNF-alpha after its administration into the experimental Morris hepatoma.

Ultrastructural analysis of pulmonary capillaries in the course of experimental lung emphysema.

Processes which occur within the capillaries of interalveolar septa play a major role in many lung diseases. In the present study, ultrastructural analysis of pulmonary capillaries was made at 7 days after intratracheal instillation of a proteolytic enzyme - papain, that is when macrophage cumulation within pulmonary tissue was maximal. Experimental animals (male Wistar rats) were injected with BCG vaccine, in doses that would stimulate the macrophage system to produce TNF-alpha. All the experimental groups, compared with the control, showed cumulation and increased adherence of monocytes, granulocytes and blood platelets to endothelium. Changes within endothelial cells manifesting their activation were observed. In the animals receiving papain or BCG, focal destruction of endothelial cells was found. The picture of vascular changes induced with papain and by simultaneous stimulation with BCG vaccine revealed a number of features similar to those found in the adult respiratory distress syndrome. The study proved usefulness of vascular perfusion as a method to analyze the vascular system, particularly in the processes progressing with activation and increased adhesion of inflammatory cells to the endothelium of pulmonary capillaries.

The effect of local hrec TNF-alpha administration upon the spontaneous lung metastases in rats with Morris 5123 hepatoma.

The experiment used Morris hepatoma 5123 series growing in muscles of the right hind limb of Buffalo rats. The group I animals were given intratumor 4 doses of TNF-alpha and group II-8 doses of TNF-alpha (10 micrograms/day). Control groups (III and IV) consisted of rats with injected Morris hepatoma, which were given PBS solution instead of TNF-alpha. A decrease in the volume of neoplastic metastases was observed in groups I and II, compared with groups III and IV. At the same time an increase was found in the volume of metastatic tumors in group II (8 x TNF-alpha), compared with group I (4 x TNF-alpha). Histological and ultrastructural analysis of the pulmonary tissue revealed intensified fibrotic reactions and inflammatory infiltrations around the metastatic tumors. The change were much more enhanced in group II, which might affect the results of neoplastic metastatic volume measurements. We concluded that multiple human recombinant TNF-alpha, hrec TNF-alpha, local injections inhibited dissemination of tumor cells and prolonged the survival time of rats up to the 76th day of the follow-up.

The effect of a herbicide--sodium salt of 2,4-dichlorophenoxyacetic acid on guerin carcinoma.

The effect of sodium salt of 2,4-dichlorophenoxyacetic acid, being an active component of herbicide "PIELIK", upon the development of Guerin carcinoma implanted in male Wistar rats, was studied. 192 animals were divided in to 6 equal groups: I-animals which obtained physiological salt solution; II-rats exposed to the herbicide in postlactational period; III-animals with Guerin carcinoma, non exposed to the herbicide; IV- rats exposed to the herbicide in postlactational period+Guerin carcinoma; V-animals exposed to the herbicide from prenatal period to the end of an experiment, without Guerin carcinoma; VI-the same as in V group, but with Guerin carcinoma. The effect of the herbicide on tumor growth dynamism (diameters and mass), degree of tumour malignancy (metastases to lymph nodes), animals survival time and morfological changes in the primary tumour and in metastases was evaluated. Basing of the results obtained, it was stated that this herbicide accelerates the development of Guerin carcinoma and reduces the survival time in the rats exposed to it in the prenatal and postnatal period. However, it does not significantly influence the growth of the carcinoma in the rats exposed only in the postlactational period.

Studies on pulmonary tissue after administration of mutein VI-HREC TNF-alpha into implantable experimental Morris hepatoma.

Experiments were carried out on Buffalo rats with implantable Morris hepatoma 5123 growing in the skeletal muscles of the limbs. Mutein VI (a protein which differs from the native TNF-alpha molecule in its N-terminal amino acid composition) was administered at a dose of 10 micrograms per rat once a day in a cycle of 8 days. Control animals were given saline (PBS). Ultrastructural changes within the pulmonary tissue were evaluated with an electron transmission microscope (TEM), with special attention paid to endothelial cells and alveolar epithelial cells. Quantitative analysis of neoplastic metastases to the lungs was carried out. The animals given mutein VI compared to those injected with PBS demonstrated a decrease in the number of metastases. TEM pictures showed accumulations of eosinophilic granulocytes and monocytes in the lumen of the blood vessels. Enhanced activity of endothelial cells was observed. In pulmonary alveoli conglomerates of fibrin, and fragments of damaged cells were found, with erythrocytes, granulocytes and macrophages in their vicinity. The epithelium of pulmonary alveoli showed signs of considerable damage, including necrosis. The mutein VI-hrec TNF-alpha was found to block the neoplastic process, illustrated by a reduction in the volume of lung parenchyma occupied by neoplastic metastases. Also, the ultrastructural changes observed in the pulmonary tissue indicate the possibility of peripheral action of mutein VI after its administration to rats carrying the Morris hepatoma.

[Morphologic analysis of congenital central nervous system malformations in children from the first of life dying in the years 1986-1990].

An incidence and morphology of the CNS congenital malformations in newborn babies and infants were analysed in the consecutive autopsies carried out in 1986-1990, i.e. following Tscharnobyl disaster. The obtained results were compared to those seen in the two earlier periods (1976-1980 and 1981-1986). In 1986-1990, a percentage of autopsies showing congenital CNS malformations increased approximately by two-fold (15%). The highest percentage of such malformations in specific years of the analysed period was noted in 1990 (20%). Central nervous system malformations were more frequent in female sex (57%) than in male sex (43%). In 64% of cases newborn babies were affected. A percentage of CNS malformations coexisting with other congenital malformations increased to 40% in the analysed period of time (from 29.4% in 1976-1985). Meningomyelocele (41%), congenital hydrocephalus (21.5%), multiple anomalies in brain (14%), and anencephaly (12.6%) constituted the most frequent group of CNS malformations. In 1986-1990, an incidence of meningomyelocele increased by more than two-fold (if this anomaly coexisted with hydrocephaly, an increase in the incidence exceeded three-fold), and hydrocephaly as well as an increase in the incidence of anencephaly by 1.5 times in comparison with 1976-1985.
