Carcinogenicity in mice and rats of heterocyclic amines in cooked foods.

Carcinogenicities of mutagenic heterocyclic amines in cooked foods have been tested in CDF1 mice and F344 rats of both sexes. Eight heterocyclic amines--Trp-P-1, Trp-P-2, Glu-P-1, Glu-P-2, MeA alpha C, A alpha C, IQ, and MeIQ--were given to mice and/or rats at 0.02 to 0.08% in the diet continuously. In mice, all heterocyclic amines tested were demonstrated to be carcinogenic. Hepatocellular carcinomas were induced in a high incidence in all groups treated with heterocyclic amines. Hemangioendothelial sarcomas were also induced by Glu-P-1, Glu-P-2, MeA alpha C, and A alpha C. Most hemangioendothelial sarcomas were located in the interscapular brown adipose tissue. In mice given IQ, forestomach and lung tumors were also observed in a high incidence. Carcinogenicity tests on MeIQ are ongoing, and interim data by week 83 show that MeIQ also induces forestomach tumors in addition to liver tumors. In rats, hepatocellular carcinomas were induced by Trp-P-1, Glu-P-1, Glu-P-2, and IQ. In rats given Glu-P-1, Glu-P-2, and IQ, adenocarcinomas in the small and large intestines, squamous cell carcinomas in the Zymbal gland and clitoral gland were also observed in a high incidence.

Carcinogenicities in mice and rats of IQ, MeIQ, and MeIQx.

The mutagenic heterocyclic amines, 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-3,4-dimethylimidazo[4,5-f]quinoline (MeIQ), and 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) are present in broiled fish, fried beef, and beef extract. Their carcinogenicities in CDF1 mice and F344 rats were tested by their oral administration in the diet. In mice given diet containing 0.03% IQ, tumors developed in the liver (hepatocellular carcinomas or hepatocellular adenomas), forestomach (squamous cell carcinomas or papillomas), and lung (adenocarcinomas or adenomas) at high incidences. In mice given diet containing 0.04% or 0.01% MeIQ, squamous cell carcinomas and papillomas of the forestomach developed at high incidences. About 40% of the squamous cell carcinomas induced in the forestomach by 0.04% MeIQ metastasized to the liver. Clear dose-response relations were seen in the incidences of tumors in the groups given 0.04% and 0.01% MeIQ. The squamous cell carcinoma-papilloma ratios were higher in 0.04% groups than in 0.01% groups. Female mice treated with 0.04% and 0.01% MeIQ showed significantly higher incidences of liver tumors than controls. The experiment on the carcinogenicity of MeIQx at a dose of 0.06% in mice is still in progress but by experimental week 74, 4 of 16 males and 7 of 18 females autopsied were found to have liver tumors. Rats given diet containing 0.03% IQ showed high incidences of hepatocellular carcinomas, adenocarcinomas of the small and large intestines, and squamous cell carcinomas of the Zymbal gand, clitoral gland, and skin. Except for the liver, the target organs of IQ in CDF1 mice and F344 rats were different.

Changes in erythrocyte carbonic anhydrase activity due to physical exercise.

Changes in erythrocyte carbonic anhydrase (RBC-CA) activity due to physical exercise were investigated using a measuring apparatus based on the electrometric method. RBC-CA activities were slightly changed due to transient aerobic (20-min run) or anaerobic (200-m run) exercise in moderately trained subjects, but there were no fixed trends. In 3 weeks of weight training, there were no differences in RBC-CA activities between before training and after 1 day of recovery in both trained and untrained subjects. However, after 1 week of recovery, RBC-CA activities slightly decreased in both types of subjects. The trained subjects showed higher levels of RBC-CA activity than the untrained subjects before and after training. The rest values of RBC-CA activity were higher in trained subjects than in untrained subjects. It was evident that RBC-CA activities were higher in subjects (especially in long-distance runners, swimmers etc.) who had undergone long-term strenuous aerobic training.

Liver regeneration and tumor growth in the rat after partial hepatectomy.

Tumor growth of Yoshida sarcoma implanted in the remnant liver was studied in rats subjected to a hepatectomy. After 70 percent hepatectomy, the liver progressively regenerated and the total liver weight was reverted to by 10 days after the operation. Concomitantly with liver regeneration, tumor growth in the remnant liver was stimulated significantly, compared with that in the sham-operated liver. Incorporation of tritiated thymidine into tumor cells in the remnant liver was strikingly high and progressive, while that in the sham-operated liver was low and retained. Mitomycin C given to the hepatectomized rats was more effective against the tumor in the remnant liver than in the sham-operated liver. We conclude from this study that cancer cell proliferation in the remnant liver can be accelerated by the process of liver regeneration.

[An experimental study of the role of portal vein blood factors in the regeneration of the liver after partial hepatectomy].

It is generally accepted that the portal venous blood has an important role in hepatic regeneration. However, the actual factor or factors of the portal venous blood have not been fully identified. In this experimental study on dogs, 1) the effect on the amount of hepatic blood flow, 2) the effect of pancreatic hormones, and 3) the effect of amino acids in the portal vein upon hepatic regeneration at 96 hours after 40% hepatectomy were examined by 3H-thymidine labelling indices of hepatocyte nuclei. Diversion of portal blood from the liver resulted in a significant decrease of labelling index. Portacaval transposition did not influence the labelling index regardless of pancreatic venous flow into the liver. On the other hand, the direct infusion of amino acids into the liver via portal vein after portacaval transposition with or without pancreatic venous flow significantly increased the labelling index. However, additional arterialization of portacaval transposition resulted in no significant increase of the labelling index. These results indicate; 1) Adequate amount of blood flow is necessary for hepatic regeneration. 2) Pancreatic venous flow into the liver may not be a major factor in hepatic regeneration. 3) High concentration of amino acids in the portal venous blood is one of the most influential factors in hepatic regeneration. 4) Oxygen concentration in the portal vein may not the factor which effected directly hepatic regeneration.

Ten years' survey of dialysis-associated tuberculosis.

Tuberculosis associated with dialysis was studied at the Renal Unit of the Tokyo Medical and Dental University and Yokosuka Mutual Aid Hospital Kidney Center, in both of which the treatments of chronic renal failure are the same. There are 12 tuberculosis patients out of 367 patients on maintenance hemodialysis from January 1967 to December 1976, an incidence of 3.3%. This was 6-16 times greater than that in the general population of this country according to yearly statistics. The characteristics of dialysis-associated tuberculosis include a high incidence of miliary tuberculosis, especially in aged patients and difficulty in establishing the diagnosis before death. Clinical features which are helpful in the early diagnosis are intermittent high fever of unknown origin, weight loss, anorexia, abnormalities of the central nervous system, erythrocyte sedimentation rate over 100 mm/h, leukocytosis and high value of the C-reactive protein. With the increasing number of dialysis patients, an increase of dialysis-associated tuberculosis is expected and this will be one of the major problems of dialysis patients in future.

Reduction of unfavourable effects of heparin with use of gabexate mesilate in dialysis.

Gabexate mesilate (GM), a potent synthetic proteinase inhibitor has been evaluated as an alternative anticoagulant to heparin for haemodialysis. GM haemodialysis was achieved at a dose of 1,600mg/hr. GM was found to be dialysable and so rapidly degraded that dialysis with GM anticoagulation was analogous to regional heparinisation. Although heparin induced considerable lipolytic activity, GM showed no particular effect on lipids. Some patients reported mild nausea and there was evidence of slight clot formation around the top of the venous drip chamber. GM anticoagulation is easier than the conventional unreliable techniques of regional heparinisation.

Comparison of dialysis programmes on different molecular prescriptions: a preliminary study.

Five dialysis and/or adsorbent therapy programmes were compared to examine the biological toxicity of both small and middle molecular fractions from the clinical viewpoint. With combination of various surface areas, diffusion pool sizes, or use of charcoal, what can be called a 'molecular prescription' is possible. The programmes were designed to bring about different degrees of removal of both fractions. Ultrafiltrates obtained with Amicon XM-10 hollow fibre filters before and after a programme were examined for levels of BUN, creatinine, and methylguanidine, inhibitory effect on pyruvate kinase, G-6-PDH, and LDH, and on the cell mortality rate of rat embryo liver cell monolayer culture. The worst results were obtained in those programmes which retained small molecular fractions and removed the middle molecular fractions efficiently. The biological toxicity of the small molecular fraction, from the viewpoint of maintaining the 'milieu intérieur', seems to be much stronger than that of the middle molecular fraction. A dialysis or adsorbent therapy programme removing the middle molecular fraction only, but leaving the small molecular fraction should be considered as putting the cart before the horse.

Hepatic resection for hepatocellular carcinoma.

One hundred and eighteen patients underwent hepatic resection for hepatocellular carcinoma from 1979 to 1987. Ninety-eight of these patients had co-existing cirrhosis of the liver; 18 patients underwent lobectomy, 28 patients had segmentectomy, and 52 patients had subsegmentectomy. In the 21 non-cirrhotic patients, 11 patients underwent lobectomy, 5 patients had segmentectomy, and 5 patients had subsegmentectomy. The operative mortality rate of patients with cirrhosis was 11% and of patients without cirrhosis was 5%. There was no significant difference in hepatic function tests between survivors and nonsurvivors. Lobectomy of < 55% of the parenchymal hepatic resection rate was well tolerated in the patients with cirrhosis. One-year, 3-year, and 5-year survival rates of patients with hepatocellular carcinoma and co-existing cirrhosis were 57.9%, 36.8% and 20.0% following lobectomy, 82.8%, 82.8% and 57.6% following segmentectomy, and 72.0%, 46.2% and 24.0% following subsegmentectomy. The tumor recurrence rate appeared to be lower after segmentectomy than subsegmentectomy. Multiple gross lesions, tumors > 5 cm, and presence of gross vascular invasion were poor prognostic signs in terms of survival rates as well as recurrence rates. Of the 51 patients with tumor recurrence limited to the residual liver, 13 patients underwent repeat resection, and 23 patients were treated by transcatheter arterial chemoembolization. The survival rates of the patients undergoing repeat resection were significantly better than those of other groups.

epsilon-N-acetylation in the production of recombinant human basic fibroblast growth factor mutein.

During large scale preparation of recombinant human basic fibroblast growth factor (hbFGF) mutein CS23(CS23) produced in Escherichia coli (E. coli), three species, named Mf-1, Mf-2 and Mf-3 in the order of their elution, were isolated from the early fraction of Sulfated Cellulofine chromatography by high performence liquid chromatography (HPLC) on heparin-5pw. Structural analysis through HPLC peptide mapping, mass spectrometry, sequencing and amino acid composition revealed that epsilon-N-acetylation occurred at positions Lys135, Lys26 and Lys119 of CS23. Together with this observation, the results on large scale preparation of CS23 were also presented.
