Red-cell-volume distribution curves in diagnosis of glomerular and non-glomerular haematuria.

The distribution curves of urinary red-blood-cell (RBC) size were obtained from automated blood-cell analysis in 146 patients with definite causes of haematuria. In 65 of 67 patients (97%) with haematuria and glomerulonephritis demonstrated by renal biopsy, urinary RBC had an irregular and asymmetrical distribution with RBC size showing a much smaller volume than that of venous RBC. This "glomerular" distribution contrasted with the "non-glomerular" normal distribution when the peak for RBC was at a larger volume than that for peripheral RBC. In 46 of 47 patients with haematuria who had lower urinary tract lesions other than infection, a non-glomerular distribution was obtained; 30 of these cases also showed glomerular distribution, and were classified as "mixed". All 32 patients with urinary tract infection had either a glomerular or mixed distribution, suggesting that they excreted distorted and dysmorphic urinary RBC. After excluding infections, this simple, rapid, reproducible, and non-invasive technique provides reliable information in distinguishing glomerular bleeding from other causes of haematuria.

Analytical electron microscopy study of high Tc superconductor YBa2Cu3O7.

The high Tc superconducting material YBa2Cu3O7 shows a complex relationship between microstructure and oxygen content, which are controlled by length of heat treatment, atmosphere, and quench rate. An AEM investigation studying changes in the oxygen near edge features was undertaken. Electron energy loss spectroscopy (EELS) measurements have the important advantage that they can be made on single crystal grains, allowing orientation-dependent studies. Both ion-milled and crushed samples with varying O2 content were analyzed. The structure of YBaCu3O7 was determined by neutron diffraction to be orthorhombic with distinct Cu-O chains along the b-axis as well as Cu-O planes in the a-b plane. Therefore, by looking for a crystallographic dependence of the oxygen K-edge one might be able to distinguish inequivalent oxygen atoms by their core level binding energy and correlate site occupancy with varying O2 content. The EELS results on the oxygen K-edge are strongly dependent on oxygen content, most noticeably when the c-axis is parallel to the electron beam.

[5-FU and HCFU concentrations in serum and tissues in hepatocellular carcinoma after HCFU oral administration].

Before proceeding with a resection of a hepatocellular carcinoma (HCC) in patients, the concentrations of 5-FU and 1-Hexylcarbamoyl-5-fluorouracil (HCFU) have been measured in the serum, the liver, and in the cancer tissue after an oral administration of HCFU (300 mg). The maximum 5-FU value was found to be 3.45 +/- 3.21 micrograms/ml (n = 22) at 2 hours, and this value decreased linearly. The concentrations of 5-FU in the liver tissues were 0.02 +/- 0.01 micrograms/g (n = 16) and the concentrations of 5-FU in cancer tissues were 0.03 +/- 0.01 micrograms/g (n = 16). There were no statistical differences found between cirrhotic patients and non-cirrhotic patients. These results indicate that the concentrations of HCFU in the serum and tissues were not influenced by liver damage.

Carcinogenicity in mice of a mutagenic compound, 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) from cooked foods.

2-Amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), which is a mutagenic compound present in fried beef and beef extracts, was given orally to CDF1 mice at a concentration of 0.06% in the diet for 84 weeks. Liver tumors were induced in 43% of males and 91% of females fed MeIQx. The incidences of liver tumors in mice of both sexes were significantly higher in groups fed MeIQx than in control groups. The incidences of lung tumors in females fed MeIQx and of lymphomas and leukemias in both sexes fed MeIQx were also significantly higher than in the respective controls.

[5-Fluorouracil, tegafur, and uracil concentrations in serum and tissue of the patients with hepatocellular carcinoma after UFT administration].

UNLABELLED: UFT is a preparation consisting of tegafur and uracil at the molar ratio 1 : 4. In 22 resected patients with hepatocellular carcinoma (HCC), the concentrations of 5-fluorouracil (5-FU), tegafur and uracil were estimated in the serum, liver and cancer tissues after oral administration of UFT (tegafur, 300mg) before operation. The concentrations of 5-FU, tegafur and uracil in the serum were highest at 2 hours, and reduced thereafter. The maximum 5-FU level was 0.040 +/- 0.034 microgram/ml. The concentrations of 5-FU in the cancer tissue were 0.101 +/- 0.070 microgram/g in all patients, 0.085 +/- 0.050 microgram/g in cirrhotic patients and 0.144 +/- 0.105 microgram/g in non-cirrhotic patients. These concentrations were significantly higher than those in the liver tissue. In 10 out of 13 patients, 5-FU concentrations was higher than 0.056 microgram/g which was reported as minimum effective concentration. CONCLUSION: the concentration of 5-FU after oral administration of UFT prior to resection of HCC was estimated in the serum, liver and cancer tissues. The concentration of 5-FU in cancer tissues was satisfactorily higher regardless of cirrhotic or non-cirrhotic conditions UFT seemed to be a safe and useful drug since tissue 5-FU concentration was higher than those in the serum and liver.

Emergence of malignant lesions within an adenomatous hyperplastic nodule in a cirrhotic liver. Observations in five cases.

Five cases of an adenomatous hyperplastic nodule or a similar lesion resected from a cirrhotic liver in which early malignant foci were seen as small nodule-in-nodule lesions are described. These hyperplastic lesions were detected by imaging diagnosis in patients with nonalcoholic cirrhosis, mostly during routine clinical follow-up. In 2 patients, recurrence of hepatocellular carcinoma occurred 11 mo and 15 mo postresection. Thus, these nodule-in-nodule lesions in an adenomatous hyperplastic nodule seem to represent an early stage of hepatocarcinogenesis in humans. In nonalcoholic cirrhotic patients from Japan and Southeast Asia, in whom hepatocellular carcinoma is endemic, an adenomatous hyperplastic nodule or a similar hyperplastic lesion that occurs in cirrhotic livers may be preneoplastic and already committed to malignant transformation.

Induction of hepatocellular carcinoma and highly metastatic squamous cell carcinomas in the forestomach of mice by feeding 2-amino-3,4-dimethylimidazo[4,5-f]quinoline.

2-Amino-3,4-dimethylimidazo[4,5-f]quinoline (MeIQ), a potent mutagen which was isolated from grilled sardines, was tested for carcinogenicity in CDF1 mice. Mice were given diet containing 0.04 or 0.01% MeIQ for 91 weeks. The incidences of liver tumors (hepatocellular carcinomas and hepatocellular adenomas) in female mice given 0.04 and 0.01% MeIQ were significantly higher than those in controls. The incidences of forestomach tumors (squamous cell carcinomas and papillomas) were also significantly higher in both sexes of mice given MeIQ. About 40% of the squamous cell carcinomas that developed in mice given diet containing 0.04% MeIQ metastasized to the liver.
