Quantitative analysis of oxepinac in human plasma, urine and saliva by gas chromatography--mass fragmentography.

A sensitive and specific method is described for the quantitative analysis of 6,11-dihydro-11-oxo-dibenz[b,e]oxepin-3-acetic acid (oxepinac) in human plasma, urine and saliva. Oxepinac and internal standard are extracted from acidified plasma, urine or saliva, converted to the corresponding n-propyl esters and analysed by gas chromatography--mass fragmentography using selected ion monitoring. The method is accurate and precise over the range 100 microgram/ml to 1.0 ng/ml. The method has been applied to the analysis of plasma, urine and saliva from healthy volunteers receiving therapeutic doses of oxepinac.

Analysis of drugs by pyrolysis. II--An improved method for the determination of carpronium chloride in plasma by selected ion monitoring.

A sensitive and specific method has been devised for the determination of carpronium chloride, a parasympathomimetic agent, in plasma. The method is based on selected ion monitoring combined with the pyrolysis reaction of (3-isopropoxycarbopropyl)trimethylammonium chloride derived by transesterification of the drug. Carpronium chloride-[2H9] is used as an internal standard. The drug and the internal standard are separated from deproteinized plasma by ion pair extraction, and converted into isopropyl ester derivatives. The derivatives are pyrolysed to produce the respective isopropyl N,N-dimethyl-gamma-aminobutyrates available for selected ion monitoring. Measurement is carried out in the electron impact mode by monitoring the fragment ion [M-OC3H7]+ (m/z 114, 120). The method is applicable to 5 ng ml-1 plasma with about 10% coefficient of variation, and has been utilized successfully for the pharmacokinetics of carpronium chloride in man.

Metabolsim of vasodilator 4-(3,4,5-trimethoxycinnamoyl)-1-(1-pyrrolidinyl)carbonylmethylpiperazine (cinepazide): isolated and identification of metabolites in the rat and man.

Further study on identification of metabolites of cinepazide, 4-(3,4,5-trimethoxycinamoyl)-1-(1-pyrrolidinyl)carbonylmethylpiperazine, in rats and men was carried out. New metabolites of cinepazide in rat urine were 4-[1-[4-(3,4,5-trimethoxycinnamoyl)-piperazinly]acetamidol-1-butanol (M-II), 4-(3,4,5-trimethoxycinnamoyl)-1-[1-(hydroxypyrrolidinyl)]carbonylmerthylpiperazine (M-IV) and 4-[1-[4-(3,4,5-trimethoxycinnamoyl)piperazinyl]acetamidol]-1-butyric acid (M-VI). Structures of two mono-O-demethylated metabolites of cinepazide predicted in the previous paper were assigned as 3-hydroxy-4,5-dimethoxycinnamoyl and 4-hydroxy-3,5-dimethoxycinnamoyl compounds (M-III and M-V) by the FT-NMR spectrometry measured in a mixture of CDC13-C6D6 (1:1, v/v) after methylation with dimethylsulfare-d6. The corresponding pyrrolidone metabolite of cinepazide was undetectable in rat urine. New metabolites of cinepazide in rat feces were found to be 4-(3,5-dihydroxy-phenethylcarbonyl)-1-(1-pyrrolidinyl)carbonylmethylpiperazine (M-VIII) and 4-(3,5-dihydroxycinnamoyl)-1-(1-pyrrolidinyl)carbonylmethylpiperazine (M-IX). Metabolites of cinepazide in human urine were studied by both methods--high performance liquid chromatography and mass spectrometry with ion cluster technique. Unchanged drug was mainly excreted in the urine, and M-VI was the major urinary metabolite. The existence of M-IV and M-V was suggested by high performance liquid chormatography. The corresponding pyrrolidone was also undetectable in human urine.

Role of the ovary in the sexual differentiation of prolactin and growth hormone cells in the mouse adenohypophysis during postnatal development: a stereological morphometric study by electron microscopy.

To clarify the role of the ovary in the sexual differentiation of prolactin and growth hormone cells, the anterior pituitary glands of mice ovariectomized before or after puberty were studied by a stereological morphometric technique with the electron microscope. A marked sex difference was observed in the relative proportions of these two types of cells in normal adult control animals. In male mice about 52% of anterior pituitary cells were growth hormone cells and about 10% were prolactin cells, while in female mice prolactin cells represented about 39% and growth hormone cells about 37% of the anterior pituitary cell population. Ovariectomy before puberty reduced the proportion of prolactin cells to about 10% and increased growth hormone cells to about 50% of the cell population. The size of prolactin cells and the development of their cell organelles was also reduced. Ovariectomy after puberty had less marked effects. These results suggest that ovariectomy before puberety induced the male phenotype by inhibition of the differentiation of prolactin cells and stimulation of the differentiation of growth hormone cells. The significance of these results in relation to the sexual differentiation of the pituitary gland is discussed.

Studies on the examination of imported laboratory monkey, Macaca fascicularis for E. histolytica and other intestinal parasites.

Infection rate of imported monkeys, under this study, with intestinal protozoa is higher than the infection rate with intestinal nematodes. Mostly encountered protozoa were E. coli, E. nana, E. histolytica and I. butschlii. Formalin-Ether concentration method yielded higher % positive for E. histolytica than the culture method using Tanabe-Chiba medium, but the results obtained from these 2 methods correlate perfectly well with each other. Efficacy of thiabendazole against intestinal nematodes such as Strongyloides, Trichuris, Physaloptera, Oesophagostomum and Capillaria was very satisfactory.

Experimental light infection of Angiostrongylus cantonensis in hamsters.

An experimental infection with a small number of larvae of A. cantonensis in hamster was carried out. When hamsters were infected with one larva, infection rate and recovery rate were 66.7%. In the case of infection with more than two larvae, all hamsters were infected had recovery rates were around 65%. The recovered worms were all matured and showed no important morphological differences. It was concluded that the hamster would be able to serve as a final host for A. cantonensis only in the case of a small number of larvae infection.

Effect of timiperone on 3H-spiroperidol binding to rat striatal dopamine receptors.

The effects of timiperone, its metabolites and related compounds on specific 3H-spiroperidol binding to dopamine receptors in the rat corpus striatum were studied to clarify the affinity of timiperone, a new butyrophenone, for the receptors and whether timiperone itself was active in vivo. Timiperone had an approx. 0.6, 5 and 30 times greater affinity for the receptors than did spiroperidol, haloperidol and chlorpromazine, respectively. This affinity was observed specifically for antipsychotic drugs but not for diazepam and trihexyphenidyl. Timiperone metabolites had little or no affinity for the receptors. Radioreceptor assay values agreed well with the radiochemical assay for timiperone in the plasma and brain of rats after i.v. injection of the 14C-labeled drug. Thus, it is conceivable that timiperone itself exerts its potent antipsychotic activity by blockade of cerebral dopamine receptors.

Experimental studies of schistosomal pigment from Schistosoma japonicum.

The schistosomal and malarial pigments were distinguishable before and after extraction from the host liver. Presence of iron in both pigments was ascertained by the elemental X-ray analysis. Histochemically, however, schistosomal pigment was similar to that of malarial pigment.

Primary adenoid cystic carcinoma of the esophagus: report of a case.

A case of primary adenoid cystic carcinoma of the esophagus is reported. A 51-year-old male patient had a tumor in the lower third of the esophagus which was incidentally found during an examination for cholelithiasis, and resected successfully. The tumor exhibited a polypoid appearance covered by normal esophageal epithelium, localized entirely in the submucosal layer, and morphologically identical to adenoid cystic carcinoma in the salivary glands. The patient is still alive and well three and one-half years after surgery. This seems to be a typical case of adenoid cystic carcinoma of the esophagus arising from the submucosal esophageal gland.
