[The measuring of the spontaneous migration of granulocytes in capillaries].

The spontaneous migration of polymorphonuclear granulocytes can be measured exactly in capillaries of 50 mm length, 0.92 mm innerdiameter and capacity of 33 +/- 0.5 microliters. The following conditions revealed to be the optimum: 1 x 10(6) cells per capillary and incubation at 37 degrees C for 3 h.

4,5-Dimethylangelicin effects on lymphocytes with and without UV-radiation.

Compared to the linear derivative 8-methoxypsoralen (8-MOP) the angular structured furocoumarin 4,5'-dimethylangelicin (4,5'-DMA) causes smaller photosensitization effects (loss of viability, inhibition of phytohemagglutinin (PHA)-induced transformation rate and E-rosette formation of T-cells, respectively) on cultured human lymphocytes. When 4,5'-DMA or 8-MOP are added to the cultures without additional UV-irradiation, increased or decreased PHA-stimulation rates are observed, respectively. In addition, 4,5'-DMA is able to reduce the binding of sheep erythrocytes to lymphocytes in a higher degree than 8-MOP. These findings suggest different action mechanisms on cytoplasma membranes and intracellular structures of lymphocytes by different furocoumarins.

[Chronic mucocutaneous candida mycosis (CMCC) caused by a T-cell defect].

It is reported on a 42-year-old book-keeper with the granulomatous variant of the chronic mucocutaneous candidiasis which could be followed up for 28 years. The intensive systemic treatment with nystatin, 5-fluorocytosin and miconazol combined with the subcutaneous injection of transfer-factor and the local application of ointments containing nystatin and clotrimazol did not only lead to the complete clearing of the lesions (4 years without any relapse), but also to the normalization of the T-lymphocyte count and the reconstitution of the formerly negative delayed type skin reactivity to candidin.

[Comparison of the drop and capillary technics of the leukocyte migration inhibition test in the detection of sensitization to candidin].

15 test persons with a positive and 3 with a negative intradermal test of candidin were examined with two modifications of the migration inhibition test. In these cases the agarose droplet test proved to be somewhat more sensitive in comparison to the capillary method. There were no correlations between the size of the cutaneous test reaction and the adequate migration index. The droplet test using culture residues (lymphokin assay) was less evident in comparison to the direct method. The two techniques (droplet test, capillary method) are well suited for the proof of cell-mediated immune reactions to candidin and other comparable antigens, respectively.

Impairment to viability of neutrophils and lymphocytes by angular psoralens and UV-radiation.

The angular psoralen derivative, 4,5'-dimethylangelicin (Dm Agn), both without and with UV-radiation induces under adequate conditions a higher percentage of lethally damaged cultured human polymorphonuclear neutrophils (PMN) as compared with angelicin (Agn). In general, PMN photosensitized with Dm Agn are less resistant than similarly photosensitized lymphocytes. These results suggest that Dm Agn might be used to improve the photochemo (PUVA-)therapy of skin diseases where PMN are of pathogenic importance.
