The blastogenic response of rabbit lymphocytes stimulated with autologous cells.

Rabbit WBC and spleen cells respond with marked blastogenesis and mitosis when cultured with mitomycin-C treated autologous sacculus rotundus, appendix and Peyer's patches cells in the one-way rabbit mixed leucocyte reaction (MLR). A lesser response is observed using stimulating cells of the other lymphoid organs. The blastogenic response in the one-way 'autologous MLR' cannot be attributed to mitogenic factors released from the mitomycin-C treated stimulator cells since it is also observed in the 'two-way autologous MLR', using untreated stimulator cells. Several explanations are proposed to account for the 'autologous MLR'.

The cells involved in cell-mediated and transplantation immunity in the normal outbred rabbit. XIII. The identity of the responder cells and the role of phagocytic cells in the mixed leucocyte culture reaction.

The ability of rabbit spleen cells depleted of thymus, bone marrow or appendix-derived cells to respond in the one-way mixed leucocyte reaction (MLR) has been investigated. The specific subpopulations of lymphoid cells were eliminated by lysis of these cells in the presence of specific antisera and complement. Phagocytic cells (monocytes and heterophils) were removed with a strong magnet after incubation with carbonyl iron particles. The results indicate that the MLR-responding cells are thymus-derived and that neither bone marrow-derived cells nor appendix-derived cells are essential for the blastogenic response following stimulation with allogeneic cells. A minimum number of phagocytic cells is required for a significant response. These cells, however, exhibit a non-specific (accessory) role and can be supplied by either the responding or the stimulating cell population.

[The Silcadraht adhesive splint--a new methods for the treatment of luxated teeth].

Splints for the immobilization of dislocated teeth have been attached to teeth with the enamal-cauterization-cement technique for the last six years at the Innsbruck Hospital. Semicircular silica wires proved to be so satisfactory with 92 patients that this method has now replaced the traditional splinting methods at our hospital. The secondary advantages are the simple production of the splint, painless application and removal, and complete protection of the periodontium. Occlusal disturbances are avoided, because the splints do not cover any of the occluding tooth surfaces.

[Animal experiments on the pharmacokinetics of 4-amidinophenylpyruvic acid (APPA) (author's transl)].

The pharmacokinetics of the synthetic protease inhibitor APPA was studied in rabbits. After intravenous or oral application of 1 and 10 mg of (carboxyl-14C)-4-amidinophenylpyruvic acid hydrochloride/kg to rabbits, the plasma concentration was determined over several days and the excretion in urine during 24 hours. The course of the plasma concentration curves showed no dose-dependence of the pharmacokinetic behaviour. A three-phase course of the plasma concentration curves was observed after intravenous application, the slowest phase having a half-life of about 20 h. This very slow elimination is considered to be caused by tubular re-absorption of APPA. There is neither accumulation in organs, tissues or erythrocytes nor binding with plasma proteins. The thin-layer chromatographic analysis of the plasma revealed no labelled metabolites, but only unchanged APPA. APPA, when applied orally (in the form of an aqueous solution), is absorbed by more than 80%, the maximum plasma level being reached within 2--3 h. Intravenous and oral applications at 24-h intervals result in a cumulative increase in APPA concentration in the plasma. If APPA is applied to pregnant rabbits, it passes into the fetal circulation.

[The value of the urethral pressure profile in urinary incontinence].

For the differentiation of the incontinence the tonometry of bladder and urethra has become indispensable in diagnostics. The author deals with the advantages of the possibility of a combined examination, the technical performance of the urethra-pressure profile and with the necessity of the same outer conditions. Taking into consideration the functional length of the urethra, the maximum pressure of the urethra as well as the difference between pressure of urinary bladder and urethra in rest and under conditions of stress the criteria for the degrees of incontinence are represented and discussed on the basis of own examinations. Not the position and the absolute length are responsible for the continence, but the relation between the attainable urethra-pressure in contrast to the appearing pressure of the urinary bladder.

Immunocompetent cells in malignant disease in man. II. Clinical and immunologic correlates in a case of non-Hodgkin's lymphoma.

The immunocompetence of the circulating lymphocytes of a patient with non-Hodgkin's lymphoma was evaluated before, during and after treatment. The immunologic features investigated were (a) the degree of blastogenic transformation induced by phytomitogens (phytohemagglutinin-M, concanavalin A and pokeweed mitogen); (b) the formation of E, EA and EAC rosettes; and (c) antibody-dependent, cell-mediated cytotoxicity. Prior to treatment the subnormal in vitro responsiveness of the cells and the very low concentrations of circulating immunoglobulins suggested immunoincompetence. A gradual shift towards normal in vitro values was observed during treatment, which corresponded to the clinical improvement in the patient's condition.

Leukemic reticuloendotheliosis: polyclonal surface immunoglobulin on "hairy" cells.

The cytochemistry, surface markers and functional properties of purified mononuclear cells obtained from the peripheral blood and spleen of a patient with leukemic reticuloendotheliosis were studied. Nonspecific esterase activity, a monocyte marker, was demonstrable in 83% of the peripheral blood mononuclear cells and 84% of the splenic mononuclear cells. Rosetting techniques failed to detect T or B lymphocyte surface markers on the majority of the cells. Direct immunofluorescence revealed capped, noncytophilic surface immunoglobulin on the cells with all immunoglobulin classes being detectable. Since noncapping conditions ahd been used during immunofluorescence staining, the observed caps were attributed to in vivo binding of autoantibodies to the "hairy" cells. This conclusion was supported by the demonstration of susceptibility of the "hairy" cells to lysis mediated by normal allogeneic lymphocytes. It is postulated that the "hairy" cells in this patient are leukemic monocytes which bear autoantibodies directed against leukemia associated antigens.

The effect of fluoride and strontium application on dentin: in vivo and in vitro studies.

The effectiveness of topical 2% sodium fluoride applied with and without 10% strontium chloride pretreatment was compared in patients complaining of tooth hypersensitivity. In addition the radiopacity of 2% sodium fluoride and 25% strontium chloride treated dentin was studied in vitro. Reduction of sensitivity to cold and mechanical stimuli was evaluated during a period of 3 months by pain scoring in the exposed cervical dentin of homologous teeth on the two sides of the jaws. There was a significant decrease in the sensitivity of the exposed cervical dentin after treatment with sodium fluoride solution with or without pretreatment with strontium chloride solution. At the end of the experimental period the combined strontium and fluoride treatment was found to be more effective than that of fluoride alone in reducing sensitivity to cold. The increase in radiodensity of dentin samples immersed in strontium chloride was statistically significant, while there was no significant increase in density for the samples immersed in sodium fluoride solutions. A mineralization process is possibly the effective means by which fluoride or strontium diminishes sensitivity of dentin.

Individualization of drug dosage during long-term treatment with phenprocoumon (Falithrom).

Pharmacokinetic studies were carried out with 3H-phenprocoumon (3H-PPC) during long-term treatment with phenprocoumon (PPC, Falithrom) in 10 female and male patients with thromboembolic diseases. Additionally, 12 mg 3H-PPC were administered per os to 6 male volunteers. Half-life and pattern of urine metabolites were measured and compared with those in patients. The radioactivity decreased independently of the total PPC plasma concentration, which was estimated fluorometrically. An exponential regression with a high coefficient of determination was found between the mean maintenance dose (PPC) and the area under plasma concentration curve of radioactivity. This procedure is considered to be a method to improve long-term treatment with PPC.
