A high-resolution, fluorescence-based, semiautomated method for DNA fingerprinting.

We describe a fluorescence-based method for the automated analysis of DNA fragments on polyacrylamide gels. A single-stranded oligonucleotide primer (18-mer) with a fluorochrome covalently bound to its 5'-end is annealed to a synthetic oligonucleotide to create a double-stranded oligonucleotide linker with a 5'-overhang complementary to a restriction enzyme site. Cosmid or plasmid DNA is digested with the appropriate restriction enzyme and then ligated to the fluorochrome-labeled linker. The labeled restriction fragments are loaded on a denaturing polyacrylamide gel in a commercially available DNA sequencer. As the restriction fragments migrate through the gel, they intersect a laser beam which excites the fluorochrome-labeled fragment. Fluorescence emission data are captured on a computer in real time and analyzed after the completion of electrophoresis. Fragment length is nearly linearly related to migration time. This method offers very near single-base resolution up to 400 bases and the ability to quantitate fragment size up to 2000 bases. The fluorochrome-labeling chemistry relies on straightforward enzymatic reactions and can be performed in a single reaction tube. Because four different fluorochromes can be used, each of 16 lanes on the gel can be used to analyze four different digest reactions, one in each color. One of the fluorochromes can be used to label size standards in each lane, eliminating interlane variability and allowing more precise estimates of fragment size. We apply the method to the analysis of overlapping cosmids.

Specific dermatoglyphic patterns: a characteristic manifestation of acantholytic dyskeratotic dermatoses.

Palmar and finger prints were obtained in patients with acantholytic dyskeratotic dermatoses. Identical patterns were found with Darier's disease, Grover's disease, Galli-Galli disease, acrokeratosis verruciformis of Hopf, and warty dyskeratoma. Because dermatoglyphic patterns are a genetically determined individual characteristic, we propose that acantholytic dyskeratotic dermatoses with identical dermatoglyphic patterns are variable manifestations of a single disorder, despite their broad spectrum of clinical manifestations.

[Pachydermoperiostosis. Clinical aspects, classification and pathogenesis].

There are 2 different forms of the rare disorder of connective tissue "pachydermoperiostosis": a primary form called Touraine-Solente-Gole (TSG) syndrome, and a secondary one, called Bamberger-Pierre-Marie syndrome (BPM). The manifestations of the two forms are very similar, but differential diagnosis is possible because BPM syndrome occurs in association with certain diseases of internal organs and effective treatment of these is followed by regression of the skin manifestations. Therefore, it seems reasonable merely to refer to primary and secondary forms of pachydermoperiostosis, rather than distinctly named syndromes. The pathogenesis of pachydermoperiostosis is unknown. Obviously a hereditary disturbance of metabolism in the connective tissue is involved, and manifestation of the disease can be either primary or secondary as a result of still unknown triggering factors.

The screening of propofol in MHS swine.

This study investigated the use of propofol in swine genetically susceptible to malignant hyperthermia (MH). Thirteen animals were exposed to 2% halothane in inspired air, a propofol infusion of 12 mg.kg-1.hr-1 for up to 45 min, or a combination of both. When MH was triggered the animals were treated with dantrolene. Mean onset time of malignant hyperthermia on exposure to halothane alone was 7.2 min +/- 3.0 min and onset with propofol and halothane was 5.0 +/- 2.5 min. In no instance did propofol alone trigger the syndrome nor was there a statistical difference (P less than 0.05) in onset time when the drug was used in conjunction with halothane. On the basis of these results, we conclude that propofol is almost certainly safe for use in humans who are susceptible to MH.

Dermatoglyphics and creases in patients with neurofibromatosis von Recklinghausen.

Fingerprint and palmar dermatoglyphics and creases were investigated in 60 patients (20 males and 40 females) with generalized neurofibromatosis. Like previous investigators, we found a significantly increased frequency of digital central pocket patterns. Furthermore, affected males and females had an increased frequency of monocentric whorls (P = 0.0037), higher quantitative values on digit II of both hands (P = 0.04), more often a reduction of main line C (P less than 0.05) with decreased frequencies of patterns in the 3rd and 4th interdigital area of the left hand (P less than 0.05), and a lower ab ridge count (males P less than 0.005; females P less than 0.001) than control individuals. On the right hand of males the frequency of high endings (5' or 5'') of line A was decreased (P less than 0.05). A significantly increased frequency of Sfl (Sydney line) was found in female patients (P less than 0.001). Male and female patients often showed a high number of secondary creases (P less than 0.001).

PC-PE hollow-fiber membrane. Structure, performance characteristics and manufacturing.

Polyether-polycarbonate hollow fibers, spun by the phase inversion method, yield a dialysis membrane with a limited ultrafiltration coefficient but high diffusive permeability. Dialysers are made out of this membrane and are sterilized by gamma radiation. The dialysers show controlled ultrafiltration and clearance values for smaller molecular weight substances in the range of cellulosic membranes, but for larger molecular weight substances the clearances are higher. Correlation for ultrafiltration and clearance values, measured in saline and whole blood, indicate low interactions between blood and membrane.

[The importance of adrenergic receptors for dermatology].

Receptor research is a rather new but most promising field of research. This review summarizes the data available on the adrenergic receptors. in addition, information is provided about the important function of these membrane receptors as control or modulator of dermatophysiology and dermatopathy.

GABA uptake by purified rat schwann cells in culture.

Purified rat Schwann cells maintained in culture for up to 6 months retained their ability to take up the neurotransmitter gamma-aminobutyric acid (GABA) by a high-affinity mechanism. Although cultured fibroblasts also accumulated GABA, they did so by a low affinity mechanism. These results indicate that Schwann cells continue to express a high affinity GABA transport system in the absence of signals from neurons.

13-cis-Retinoic acid in rosacea. Clinical and laboratory findings.

The paper presents clinical and laboratory data on 13-CIS-retinoic acid treatment in rosacea. Good-to-excellent results are reported for severe forms of rosacea. Side effects were tolerable and could easily be controlled. The role of sebaceous gland function was documented by both histological and skin sebum parameters. Diminution of sebaceous gland size and the decrease of skin sebum with alteration of single sebum fractions correspond with findings in acne. According to the findings the alteration of sebaceous gland function is one model of drug action in rosacea. Thus sebaceous gland dysfunction may play an important role in the pathogenesis of rosacea.

Angiosarcoma of the superior vena cava.

The first reported case of an angiosarcoma apparently arising from the superior vena cava is presented. The patient, a 20-year-old white man, was treated by surgical excision and reconstruction with dacron grafts from the left and right brachiocephalic veins to the right atrium. This was followed by a course of mediastinal irradiation. The patient remained clinically disease-free at 24 months post-treatment. The patient had occasional occupational exposure to polyvinyl chloride.

Do sexual steroids contribute to the pathomechanism of rosacea? A study of estrogen and androgen receptors in acne rosacea.

Estrogen- and androgen receptor levels were investigated in the skin of 11 male and 4 female rosacea patients. No significant difference in receptor distribution or in receptor levels could be found between rosacea lesions and normal skin of the same patients. Moreover, hormone serum levels were in the normal range. Data from the present investigation suggest that rosacea formation is not governed by endocrine, although clinical and electron microscopic observations indicate a possible endocrine influence at the vascular level.

[13-Cis-retinoic acid: a new form of treatment of rosacea (author's transl)].

13-cis-retinoic acid was given with good results to 7 patients with exceptionally severe rosacea. Side effects were transitory and of minor consequence and subsided quickly on reduction of the dosage and topical application of mild cosmetics. The long-lasting remissions recorded to date indicate that 13-cis-retinoic acid may prove a new and successful approach to the treatment of rosacea.

[Unilateral nevoid telangiectatic syndrome].

In recent years the unilateral nevoid teleangiectasia syndrome (UNTS) has been mainly reported in the Anglo-American literature. This rare disease is not well-known in the German literature, where hitherto it has not been published. Based on the observation of a new case the clinical and histologic features of UNTS are reviewed and the pathogenesis discussed in detail.

[Oculodermal melanocytosis (nevus of ota) and blue nevus (author's transl)].

Report is made of a patient having concurrently an Ota nevus and a blue nevus. The dermal melanocytes showed a marked formation of large intracytoplasmatic vacuoles. Reference to the important role of an "extracellular sheath" is made. The nearly identical ultrastructural characteristics of dermal melanocytes in various dermal melanocyte disorders seems to imply a common pathological origin in their development.

[Subcutaneous Dirofilaria (Nochtiella) repens infection in man--report of the first case in Austria and review of the literature (author's transl)].

A 39-year-old female patient developed migratory skin nodules 4 months after a vacation in Greece. The patient observed a total of 17 nodules moving over the trunk to the left thigh, where a subcutaneous lesion was excised, yielding a parasite measuring 0.5 mm X 7 cm. It was classified as Dirofilaria (Nochtiella) repens. Since this is the first case observed in Austria, an exact description of subcutaneous dirofilariasis, its geographical distribution and a review of the literature is presented.
