Adjusted recovery ratios of somatosensory evoked potentials were computed using Callaway's amplitude/standard deviation measure of electrocortical activity in post-stimulus epochs of 15-30, 31-50, 51-90, and 90-240 msec. Significant differences in amount of recovery and shape of the recovery functions between young and elderly adults were observed only in early (15-90 msec) phases of the responses; subjects aged 60-70 did not differ from those aged 71-80. The results demonstrate a complex interaction between age and recovery, with stability of recovery functions after age 60.
Amoebae of Naegleria fowleri nN68 became elongated flagellated cells 150 to 180 min after subculture to non-nutrient buffer. N. fowleri NF69 did not become elongated or flagellated under these conditions. Electron microscopic examination of N. fowleri confirmed that it is a typical eucaryotic protist with a distinct nuclear envelope and prominent nucleolus, numerous vacuoles and cytoplasmic inclusions, pleomorphic mitochondria, and some rough endoplasmic reticulum. During incubation in non-nutrient buffer, both strains lost ultraviolet-absorbing material to the medium, and the number of vacuoles decreased. In strain nN68, basal bodies, a rootlet, and flagella are formed quickly after an initial lag of 90 min. Initially, the rootlet is not associated with the nucleus but they become associated subsequent at the leading end of the elongated cell. In elongated cells, the rootlet lies in a furrow or groove extending the length of the nucleus. Flagella of N. fowleri nN68 exhibit the typical 9 + 2 arrangement of filaments and are surrounded by a sheath which is continuous with the plasma membrane. The enflagellation process in N. fowleri can be manipulated reproducibly.
The intracellular fate of 3H-labeled exogenous herpes simplex virus type 2 DNA following the exposure of recipient cells to selected DNA facilitators and the ultrastructural changes which ensued in transfected cultures were examined. In most cases, electron microscopic radioautography demonstrated that exogenous DNA reached the nucleus within 1 h postinoculation. At later time periods, enlargement of nuclei and margination of chromatin were noted. Cells treated with polyornithine differed significantly from those treated with other facilitators in that large cytoplasmic accumulations of labeled material associated with electron-dense, membrane-bound bodies persisted after 1 h postinoculation. In contrast, labeled DNA released by intact virions was detected in the nucleus as early as 20 min postinfection (p.i.), and progeny virus was evident by 8 h p.i. These results suggest that the efficiency of each facilitator is related to the rapidity with which exogenous DNA penetrates the recipient cell, traverses the cytoplasm, and the amount of DNA which is finally associated with the nuclear compartment.
Experimental non-A, non-B hepatitis infection has been shown to produce two distinct ultrastructural changes in chimpanzees: intranuclear particles and intracytoplasmic tubular structures. We report our observations of intranuclear particles in humans with acute non-A, non-B hepatitis. These particles appear to be specific for at least one type of non-A, non-B hepatitis.
Peripheral blood mononuclear cells from pregnant and postpartum women were cultured in vitro with erythropoietin. Burst-forming unit (BFU-E)-derived erythroid colonies composed of immature erythroblasts with low hemoglobin contents were observed by day 8 of culture. By day 12 of culture, numerous BFU-E-derived erythroid colonies with high hemoglobin contents were present. The gamma/(gamma + beta) globin synthetic ratio was approximately 12% in the early cultures and 6% in the late cultures, indicating that the proportion of fetal hemoglobin synthesis decreases during erythroid cell maturation. These studies also reveal that the capacity fof fetal hemoglobin production by peripheral blood BFU-E in vitro is not altered during pregnancy.
Messenger RNA (mRNA) was isolated from adult as well as normal and mutant f/f fetal mouse reticulocytes by poly-U affinity chromatography. mRNA from normal adult reticulocytes directed equal synthesis of alpha and beta globin chains in wheat germ cell free translational system. mRNA from either normal or mutant f/f fetal reticulocytes directed 40% more alpha globin synthesis than beta globin synthesis. These observations are suggestive that there is significantly more alpha mRNA than beta mRNA in normal and mutant f/f fetal mouse reticulocytes.
The diagnosis of amoebiasis presents problems, particularly if one relies on finding the organism. Thus, serologic tests are expedient. A gel diffusion precipitin test (GDP), commercially available, simple to perform, and inexpensive, was compared with the indirect hemagglutination test (IHA). 257 Patients' sera were tested; 14 had amoebic colitis, 21 had amoebic liver abscess, 63 had suspected amoebic liver abscess, and 46 had inflammatory bowel disease. GDP tests were positive in 85% of amoebic colitis and 95% of amoebic liver abscess patients; IHA was positive in 91% of amoebic colitis and 94% of abscess patients. Within 6 mo, GDP tests became negative in 66% of patients. IHA tests were observed positive up to 20 yr. The performance characteristics of diagnostic methods in amoebiasis, fecal examination, IHA and GDP, show serologic tests have superior sensitivity and predictive value in recognizing invasive disease.

Pure populations of large, nucleated erythrocytes derived from yolk sac blood islands were obtained during normal fetal mouse development. Embryonic hemoglobins were present in these cells early in gestation. Later in gestation, an increasing amount of adults hemoglobin was also synthesized and accumulated in this population of primitive nucleated erythrocytes, as demonstrated by both biochemical and immunocytochemical techniques.

A 10-year retrospective analysis of 15 patients with amebic liver abscess is reviewed and represents a continuation of the previous 2-decade experience at our institution (37 patients). Records were studied to determine the population affected, presenting symptoms, physical and laboratory findings, type and response to therapy. Most patients were rural males of lower socioeconomic status in the third to fifth decade of life. They presened as a febrile illness with abdominal pain for an average of 2 1/2 months duration. Significanct physical abnormalities were tender hepatomegaly (93%), right-sided pulmonary changes (40%), and fever (66%). All patients had abnormal liver scan, positive amebic serology, and hypoalbuminemia. All patients promptly responded to amebicidal therapy except one whose therapy was delayed. Clinical suspicion, liver scanning, serologic testing, and response to therapy are the keys to diagnosis.
