[Blood-brain barrier opening microcirculation in human brain tumor].

The changes of the human tumor microcirculation after blood-brain barrier (BBB) opening was studied. BBB opening was performed by retrograde injection of hyperosmotic solution, such as mannitol or glycerol, from superficial temporal artery in the operation procedure. Serial fluorescein micro angiography of human tumor surface was taken by motor-driven camera equipped to surgical microscope. Two glioblastomas, two astrocytomas and one metastatic tumor were studied. In glioblastoma multiforme, after BBB opening, fluorescein sodium perfusion was increased with the enlargement of the venules and extravasation of fluorescein sodium was also observed from venules by the late venous phase of fluorescein microangiography. In low grade astrocytoma, after BBB opening, fluorescein sodium perfusion was relatively decreased in the vasculature which diameter was from 5 micron to 10 micron and no extravasation of fluorescein sodium was observed from the venules. In metastatic brain tumor, extravasation of fluorescein sodium was observed, ever before BBB opening, especially from arterial capillary or venous capillary. After BBB opening, extravasation of fluorescein sodium from those capillaries was increased and the tumor staining of fluorescein sodium was observed after BBB opening. But in low grade astrocytoma, BBB disruption was not observed. The effect of BBB disruption was more marked in malignant tumor, but little effect was observed in low grade astrocytoma and normal brain tissue. The degree of BBB disruption is depend on the volume and speed of hyperosmotic solution. Using our methods of volume and speed, we could observed the different BBB disruption according to the malignancy of the tumor. The difference of BBB reaction to hyperosmotic solution between low grade astrocytoma and malignant tumors is their stability of the BBB.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of concomitant posterior fossa and supratentorial hemangioblastomas].

The patient was a 31-year-old female who complained of headache and gait disturbance and admitted to our hospital on January 14, 1983. Familiar history revealed that her mother died of hemorrhage from hemangioblastoma in area postrema. Neurological examination revealed bilateral papilledema, right cerebellar sign and ataxia. Other neurological and clinical examinations were normal. CT-scan showed a right cystic cerebellar lesion and a right intraventricular lesion suggestive of hemangioblastomas. Right vertebral angiography demonstrated two vascular tumors lying in the right cerebellar hemisphere and right trigone of the lateral ventricle. Operation was performed on January 28, 1983. The histological diagnosis were hemangioblastomas.

[Statistical analysis of nutrients intake affecting serum lipid values of workers in an industry].

Relationships between serum lipid values and the quantity and quality of nutrient intakes of the employees in a steel factory were studied, and the following results were obtained. High density lipoprotein cholesterol (HDL-CL) and alpha-CL separated by electrophoresis in office workers were significantly lower than those of field workers and they were especially lower in cases of hypertriglyceridemia. HDL-CL, alpha-CL and Apo A values of workers with abnormal circulatory findings were demonstrated to drop down with increased cigarette smoking or carbohydrate intake. On the other hand, these values were apt to be elevated with increased alcohol or energy intake. beta-Lipoprotein and beta-CL separated by electrophoresis in workers with hyperlipidemia or abnormal electrocardiogram findings were shown to be at significantly high levels. Furthermore, beta-CL values in workers with abnormal circulatory findings were especially inclined to be elevated with increased cigarette smoking or fat intake.

Acyl derivatives of 16-membered macrolides. I. Synthesis and biological properties of 3"-O-propionylleucomycin A5 (TMS-19-Q).

Using leucomycin A5 (1), 3"-O-propionylleucomycin A5 (7) was synthesized by the following synthetic route: 2"-O-acetylation, 3,9-di-O-trimethylsilylation, 3"-O-propionylation, 3,9-di-O-detrimethylsilylation and 2'-O-deacetylation. Acylation of the 3"-tertiary hydroxyl group of 2'-O-acetyl-3,9-di-O-trimethylsilylleucomycin A5 with propionyl chloride in the presence of tribenzylamine at 70 degrees C gave a 3"-O-propionyl derivative in 96% yield. The structure of the final compound, 3"-O-propionylleucomycin A5 (7) was confirmed by means of mass, 1H-NMR and 13C-NMR spectrometry and chemical degradations. 3"-O-Propionylleucomycin A5 (7) showed higher antibacterial activity in vitro and higher serum levels than its mother antibiotic. The biological properties of 7 also were compared with those of josamycin and midecamycin.

Studies on neplanocin A, new antitumor antibiotic. I. Producing organism, isolation and characterization.

Neplanocin A. C11H13N5O3, is a novel carbocyclic analog of adenosine with cyclopentene. It was isolated from the culture filtrate of Ampullariella regularis A11079 by means of ion-exchange, carbon, silica gel adsorption, or partition chromatography. Neplanocin A forms crystals, and is stable at acidic or alkaline pH. Neplanocin A has cytotoxicity against L5178Y cells in culture and showed a remarkable effect on the life prolongation of mice infected with L1210 leukemia.
