Identification of species specific hemoglobin by isoelectric focusing.

A method for the discrimination of human hemoglobin from animal sources is described. Hemoglobins from 34 species with and without treatment by p-chloromercuribenzoate (PCMB) were examined by thin-layer isoelectric focusing (IEF) in polyacrylamide gel (PAG) plates containing an Ampholine gradient of pH 3.5 - 9.5. PCMB treatment was effective in distinguishing human and animal hemoglobins with their characteristic IEF patterns. Values for the isoelectric points of the major and minor hemoglobins of each species are also described.

Segmental transcatheter hepatic artery chemoembolization with iodized oil for hepatocellular carcinoma: antitumor effect and influence on normal tissue.

PURPOSE: Segmental transcatheter arterial embolization (TAE) with use of iodized oil mixed with an anticancer drug, followed by injection of gelatin sponge particles, was undertaken to evaluate its antitumor effect and its influence on normal tissue in patients with hepatocellular carcinoma (HCC). PATIENTS AND METHODS: Histologic findings in 12 patients who underwent hepatectomy after segmental TAE were compared with findings on plain radiographs and computed tomographic (CT) scans. Visualization of the portal veins contiguous to the tumor on radiographs and the pattern of iodized oil accumulation in the tumor and vicinity on CT scans after TAE were assessed. RESULTS: Complete necrosis of the tumor was achieved in 10 cases (83%), while complete necrosis of daughter nodules and capsular invasion was observed histologically in eight of these 10 patients (80%). The degree of tumor necrosis correlated with the pattern of iodized oil accumulation in and near the tumor. Partial necrosis of normal tissue near the tumor correlated with accumulation of iodized oil. CONCLUSION: Segmental TAE may be an excellent therapeutic method for treatment of HCC that is localized in one or a few segmental or subsegmental regions.

Immunohistological and biochemical detection of fucosylated polylactosamine antigen with monoclonal antibody ACFH18 in gastric cancer and normal gastric mucosa.

A mouse monoclonal antibody ACFH18, produced by immunizing with human gastric cell line MKN-74, recognizes a novel glycolipid antigen, fucosyl-lactoneodecaosylceramide. In the present study, we investigated the immunohistological localization of this antigen and its biochemical detection in glycolipid fraction in normal and malignant human gastric tissue, compared with other tumor-associated type-2 chain glycolipid antigens such as sialyl Lex, sialyl Lex-i and Ley. Reactivity with ACFH18 was detected immunohistologically in the proliferation zone of normal fundic gland region as well as in 38 of 54 cases of gastric cancer, with preferential binding to undifferentiated type cancer. Glycolipids reacting with ACFH18, especially slow-migrating glycolipids on thin-layer chromatography, were accumulated in all of four cancer specimens compared with normal mucosa obtained from the same patients. Many glycolipids with sialyl Lex or sialyl Lex-i epitope were detected in normal mucosa and almost all of these glycolipids were accumulated in cancer specimens. Sialyl LeX-i antigen was increased more specifically in cancer than sialyl LeX was. A Ley antigenic glycolipid was considerably decreased in three of four cancer cases and increased in one case. From these results, glycolipid detected by ACFH18 appeared to be a tumor-associated antigen comparable to sialyl LeX and sialyl Lex-i antigens, but with a unique character as an undifferentiated-type tumor-associated antigen.
