Ring formation by human variant endothelial cells in vitro.

Variant endothelial cells were cultured from the healthy carotid arteries of a 19-year-old woman who was killed accidentally. The cells were grown and subcultured in Medium 199 supplemented with 20% heat-inactivated fetal calf serum. The cells are still viable after 20 passages. They were recognized as variant endothelial cells by their morphology which included ring formation, and by the existence of factor VIII-related surface antigen on the cell membrane. The cultured endothelial cells produced prostacyclin when the cells were incubated with arachidonic acid in smaller amounts than does typical endothelium, but far in excess of that produced by vascular smooth muscle cells.

Injury to cultured endothelial cells from human umbilical vein by linoleic acid hydroperoxide.

The effect of linoleic acid hydroperoxide on cultured endothelial cells from human umbilical vein was examined morphologically. Incubation of the cells with a low concentration (0.5 nmol/ml) of linoleic acid hydroperoxide for 3 h caused a slight decrease in electron density of the mitochondrial matrix. Upon an increase of the concentration to 1.0 nmol/ml, the damage became more pronounced, and enlargement and dilatation of the rough-surfaced endoplasmic reticulum were observed. At higher concentrations (5 and 7.5 nmol/ml), dilatation of the rough-surfaced endoplasmic reticulum became further pronounced, and many vacuoles were observed in the cells.

[An autopsy case of malignant histiocytosis showing a phenotype of S100 +lysozyme].

A 37-year-old Japanese man was admitted to our hospital because of high fever. The peripheral blood showed pancytopenia with a few erythrophagocytic histiocytes. A bone marrow aspirate revealed 12% immature histiocytes and 7% leukoerythrophagocytic histiocytes. A postmortem examination showed an infiltrative proliferation of immature histiocytes and erythrophagocytic histiocytes in the sinusoid of the liver, the urinary bladder, etc. These findings were consistent with malignant histiocytosis. In addition, these abnormal histiocytes showed a phenotype of S 100 protein +lysozyme.- In agreement with Watanabe's report, our findings suggest that malignant histiocytosis is the neoplasm of T-zone histiocytes.

Perianal alveolar rhabdomyosarcoma in a child. Report of a long-term survival case.

The long-term survival of alveolar rhabdomyosarcoma of the perianal region in a 13-month-old infant is described. Local excision with chemotherapy consisting of vincristine, actinomycin D, and cyclophosphamide over a period of two years has resulted in eight years of tumor-free survival with normal growth and development. Our case and review of the literature suggest that local excision with chemotherapy may be an appropriate treatment for Group I perianal rhabdomyosarcoma in children.

[Case of disseminated necrotizing leukoencephalopathy following intrathecal methotrexate in acute lymphocytic leukemia].

Disseminated necrotizing leukoencephalopathy was found upon autopsy of a 32-year-old man with acute lymphocytic leukemia. He had been treated with intrathecal methotrexate for CNS involvement; no radiation therapy had been given. Autopsy findings revealed that in the white matter of the temporal lobe, there was widespread necrosis associated with numerous foam cells, demyelination, vacuolization and slight gliosis; in this lesion, leukemic cells infiltrated the Virchow-Robin space. In the white matter of other lesions we found discrete multifocal necroses of coagulative type associated with round or rod-shaped axonal swelling partly accompanied with calcification. However, in these lesions, there was no leukemic cell infiltration.

Rhabdomyosarcoma of the right ventricle.

A case of a 66-year-old male with rhabdomyosarcoma of the right ventricle who presented first with a pleuritic chest pain and later developed congestive heart failure with A-V block is described. The total period of the disease was about 9 months. At autopsy the right ventricle and wall were found to be replaced by tumor tissue which was diagnosed histologically as rhabdomyosarcoma of the alveolar type. We described one case and 52 rhabdomyosarcomas of the heart from the literature; the ages ranged from 3 months to 92 years with 29 men and 23 women (unknown 1). The primary lesions were right ventricle 29%, right atrium 26%, left atrium 24%, left ventricle 6%, and others 15%.
