Heat production by adipocytes from obese subjects before and after weight reduction.

Microcalorimetry has been employed to measure the heat production by adipocytes obtained by percutaneous biopsy from lean subjects and from obese subjects before and after weight reduction. Cellular heat production was significantly lower in obese than in lean subjects. After weight reduction cellular heat production increased in fat cells from the obese subjects but was still significantly lower than in cells from control subjects. A number of variables reflecting uptake and mobilization of depot fat have measured and correlated to the heat production values in the obese subjects. The findings are consistent with the view that a decreased total metabolic activity might contribute to the development or perpetuation of obesity.

The effect of D,L-carnitine supplementation on muscle metabolism, neuropathy, cardiac and hepatic function in hemodialysis patients. A pilot study.

Three patients on regular hemodialysis treatment were perorally supplemented with D.L-carnitine, 900 mg/d, during four weeks. Before treatment, muscle carnitine concentrations were low and the patients showed signs of hypometabolism in skeletal muscle, cardiomyopathy, impaired hepatic function and a moderate hyperlipoproteinemia. After treatment, the skeletal muscle metabolism normalized. The results further indicate improvement of the cardiac and hepatic function and of the neuropathy on the sensory side. No improvement was found, however, in plasma lipoprotein pattern or in variables reflecting plasma lipid transport (lipoprotein lipase activity and intravenous fat tolerance).

Heat production in different populations of human blood cells exposed to immune complexes in vitro: the importance of the Fc parts of immunoglobulins and the influence of active complement.

By use of a batch microcalorimeter of the thermopile type, heat production was measured in isolated populations of human peripheral blood cells exposed to defined immune complexes formed in vitro. It was found that most of the heat production recorded in whole blood after admixture of immune complexes occurs in the granulocytes. Under these conditions small but constantly higher activation values were found in the absence of active complement. It was shown that complexes consisting of antigen and F(ab)2 fragments prepared from the specific antibodies were able to initiate heat production in the cells only in the presence of active complement. These experiments indicate that immune complexes are able to induce increased heat production in the cells either by binding to Fc receptors or by activation of complement through the alternative pathway and subsequent binding of the generated C3b to C3b receptors on the heat-producing cells.

[Association of filiform hematomas of the nails and Raynaud's syndrome].

Two men (non smokers) suffer from unilateral Raynaud's phenomenon, as well as homolateral splinter hemorrhages of the nails. One of the patients has digital artery obstruction, confirmed angiographically. The other patient, in whom a thoracic outlet syndrome was diagnosed, suffers of cold-induced vascular spasm. The incidence, significance and etiology of splinter hemorrhages are discussed.

[Variations of the sum of the arterial partial pressure of inert gases during changes in the composition of inhaled gas without a variation of the ambient pressure, in a hyperbaric atmosphere].

In hyperbaric environments, when inhaled inert gas composition is abruptly modified, the sum of the arterial inert gases partial pressures is different to the sum of these same gases in the inhaled mixture. While switching from a helium-oxygen to a nitrogen-oxygen mixture of same P1O2 and total pressure, the sum of the arterial inert gas partial pressure was transiently less than the one in the inspired gases: there was an arterial under-saturation: PaHe + PaN2 = 0.68 (P1He + P1N2). During the opposite switch (from nitrogen to helium), a reversed time course, namely a transient over saturation, was observed: PaHe + PaN2 = 1.31 (P1He + P1N2). Amongst the different possible explanatory hypotheses, the most probable is that inert gas partial pressure equilibrium through the alveolo-capillary membrane is not achieved when the blood leaves the pulmonary capillary.

Erythrocyte 2,3-diphosphoglycerate and adenosine triphosphate in polycythaemia vera.

Erythrocyte 2,3-diphosphoglycerate (2,3-DPG) and adenosine triphosphate (ATP) were measured in 16 patients with polycythaemia vera and found to be normal. The determination of erythrocyte organic phosphate concentration can be helpful in the differential diagnosis of different types of polycythaemia due to the lack of 2,3-DPG rise in patients with polycythaemia vera, as compared to the 2,3-DPG increase usually observed in patients with secondary polycythaemia.

Metabolic activity of lymphoma cells and clinical course in non-Hodgkin lymphoma (NHL).

By microcalorimetry the metabolic activity of malignant cells from 2l patients with non-Hodgkin lymphoma was monitored. Heat production, a measure of the metabolic activity of cells, showed positive correlation with the clinical course of the disease. The heat produced by tumor cells from patients with progressive disease was 5.5 pW/cell, while that from patients responding to treatment was 3.l pW/cell. The difference between the 2 groups is significant (P less than 0.05). A higher heat production was obtained in tumor cells from 7 out of 10 patients who were unresponsive to the therapy when compared with cells from 11 patients who had become asymptomatic as a result of the same therapy. Also heat production from blood lymphocytes was found to be higher (P less than 0.005) in the group of patients with progressive disease as compared to the group of patients who responded to therapy. The present results indicate an association between high heat production in lymphoma cells and blood lymphocytes and a poor prognosis.
