Mosquito (Diptera: Culicidae) and predator abundance in irrigated and rain-fed rice fields in north Sulawesi, Indonesia.

Immature mosquito species composition and abundance were studied in irrigated and rain-fed rice fields of North Sulawesi, Indonesia. Irrigated rice fields were characterized by the prevalence of aquatic macrophytes and cyprinodont larvivorous fish, Aplocheilus panchax (Hamilton), but abundance per dip of most aquatic insect predators was lower than that in rain-fed rice fields. Anopheles peditaeniatus (Leicester), Culex vishnui Theobald, and Culex tritaeniorhynchus Giles, were dominant in both irrigated and rain-fed fields, but the abundance of the Culex species was lower in irrigated fields. The effect of irrigation system introduction on regional mosquito abundance cannot be evaluated by the enlarged surface water area alone. Changes in habitat quality, expressed as the abundance per dip (index of density per unit water area), also need to be considered.

Hypertriglyceridemia associated with tumor necrosis factor-alpha in hamster cheek-pouch carcinogenesis.

We demonstrated for the first time that 9,10-dimethyl-1,2-benzanthracene (DMBA)-treated hamsters showed hypertriglyceridemia followed by cachexia. Hypertriglyceridemia is believed to be caused in part by the decreased lipoprotein lipase (LPL) activity, and by cytokines such as tumor necrosis factor (TNF)-alpha. In addition, TNF-alpha action is associated with the LPL activity. Therefore, we determined the content of triglyceride (TG), LPL, and TNF-alpha in the serum from DMBA-treated hamsters. Elevated TG concentration in the serum of tumor-bearing hamsters was more remarkable and preceded the increase in other lipids, whereas the activity of LPL, the key enzyme of TG metabolism in vivo, was drastically reduced. TNF-alpha, known as an endogenous inhibitor of LPL activity, was detected in both the sera and the extract of tumors from DMBA-treated hamsters, whereas it was not detectable in any control samples. Pre-incubation of control sera with exogenous recombinant human TNF-alpha resulted in a potent inhibition of endogenous LPL activity in a dose-dependent manner in vitro. Therefore, the presence of TNF-alpha might lead to the increase in plasma TG mediated by LPL in tumor-bearing hamsters.

Macroconditions of channel segments utilized by Culex pipiens pallens immatures in Saga City, southwest Japan.

The relation between macroconditions of water channels and the breeding of Culex pipiens pallens in Saga City were examined, based on weekly census data over 3 years for ca. 2,000 channel segments. Mosquito breeding activity was greatest in channel segments between building lots and field lots in the suburban area. The segments involved ranged from narrow roadside ditches (width < 1 m) to wide irrigation canals (width > 10 m), all of which were utilized by mosquitoes. There was a weak tendency for more mosquito breeding in narrower segments. Water flow indices ranged from 0% (always stagnant) to 100% (always flowing). Mosquito breeding tended to be suppressed with an increase in flow indices. Mosquito breeding was confirmed in ca. 35% of segments yearly, and such mosquito-productive segments tended to persist during 3 study years.

Age structure of Anopheles subpictus (Diptera:Culicidae) collected by a light trap in Halmahela, Indonesia.

Age structure of Anopheles subpictus Grassi was studied at a Halmahela village, Indonesia. Ovary development of females reared with 2% sugar solution after emergence ceased at Stage I and their ovary size remained small (mean length x width = 0.68 x 0.18 mm). Females with such small ovaries occupied 21.2% of unfed Stage I females in cattle-bait samples. The remaining unfed Stage I or II females had cleary larger ovaries (mean 1.17 x 0.39 mm). Unfed Stage I or II females collected by the outdoor light trap all had large ovaries (mean 1.42 x 0.34 mm) irrespective of stages and parity. The parous rate of unfed stage I or II females collected by the light trap (86.6%) was significantly higher than that of cattle-bait samples (69.6%). Feasibility of using outdoor light trapping in malaria entomology was discussed.

Interleukin-1 beta, interleukin-6, epidermal growth factor and transforming growth factor-alpha are elevated in the brain from parkinsonian patients.

Interleukin (IL)-1 beta, IL-6, epidermal growth factor (EGF), and transforming growth factor-alpha (TGF-alpha) were measured for the first time in the brain (caudate nucleus, putamen and cerebral cortex) from control and parkinsonian patients by highly sensitive sandwich enzyme immunoassays. The concentrations of IL-1 beta, IL-6, EGF, and TGF-alpha in the dopaminergic, striatal regions were significantly higher in parkinsonian patients than those in controls, whereas those in the cerebral cortex did not show significant differences between parkinsonian and control subjects. Since these cytokines and growth factors may play important roles as neurotrophic factors in the brain, the present results suggest that they may be produced as compensatory responses in the nigrostriatal dopaminergic regions in Parkinson's disease, and may be related, at least in part, to the process of neurodegeneration in Parkinson's disease.

Tumor necrosis factor-alpha (TNF-alpha) increases both in the brain and in the cerebrospinal fluid from parkinsonian patients.

Tumor necrosis factor-alpha (TNF-alpha), a glial-cell-related factor, was measured for the first time in the brain (striatum) and cerebrospinal fluid (CSF) from control and parkinsonian patients by a sensitive sandwich enzyme immunoassay. The concentrations of TNF-alpha in the brain and CSF were significantly higher in parkinsonian patients than those in controls. Since TNF-alpha is an important signal transducer of the immune system with cytotoxic and stimulator properties, these results suggest that an immune response may occur in the nigrostriatal dopaminergic regions in Parkinson's disease and that TNF-alpha may be related, at least in part, to the neuronal degeneration.

Increased beta 2-microglobulin in both parotid and submandibular/sublingual saliva from patients with Sjögren's syndrome.

Samples of unstimulated saliva from patients with sialoadenopathy were collected by microcapillary tube (1 microliter), and their beta 2-microglobulin (B2-MG) content determined by a sandwich enzyme immunoassay. A higher than normal (control) concentration of the globulin was present in both parotid and submandibular/sublingual saliva from the patients with Sjögren's syndrome but not in the samples from the patients with sialoadenitis or diabetes mellitus. The increase in B2-MG in saliva from patients with Sjögren's syndrome may reflect that immunolopathological events are important in the degeneration of both glands in this disease. Therefore, the determination of B2-MG in saliva may be a simple, non-invasive technique for confirming the diagnosis of Sjögren's syndrome as an autoimmune disease.

Prevalence of intestinal helminthic infections in Kao District, north Halmahera, Indonesia.

A parasitological survey was conducted on the inhabitants of six villages of Kao District, Halmahera Island, North Maluku, Indonesia, in July 1993. A total of 422 fecal samples were examined by using Kato-Katz thick smear, modified Harada-Mori culture and formalin ether concentration techniques. Seven nematode species, ie Ascaris lumbricoides, Trichuris trichiura, Necator americanus, Ancylostoma duodenale, Strongyloides stercoralis, Enterobius vermicularis and unidentified rhabditoids of free-living nature, were detected. Trematode and cestode infection was not proven. Necator americanus was the predominant species of hookworm. Soil-transmitted nematode infections were highly prevalent. Among the young inhabitants aged less than 15, positive rates of Ascaris, Trichuris and hookworm infections were 32.7, 52.7 and 68.6%, respectively. Among the people aged 15 or more, the positive rate for hookworm (85.9%) was much higher than that for Ascaris and Trichuris (13.5 and 40.5%, respectively). Egg count revealed that more than 90% of inhabitants with Trichuris or hookworm had light infections. The latrines in the surveyed area seemed to have only limited effects on the improvement of the parasitological status because the prevalence of Trichuris infections was much higher in a village where most houses were provided with latrines. These conflicting conditions were considered to have been caused by many factors including the inadequate structure of the latrines.

Japanese Aedes albopictus among four mosquito species reaching New Zealand in used tires.

Since a 1988-89 survey of northern New Zealand revealed no additions to the known mosquito fauna, this country's used tire importations have much increased. Relevant entomological quarantine was thus monitored in a November 1992-January 1993 Auckland project, during which almost 1/3 of 8,549 casings from Japan proved wet on inspection. In this study and at 2 South Island ports afterwards, 5 vessels from Japan and one from Australia were found to have brought in mosquito-infested used tires. Live Aedes albopictus (all larval instars, pupae, and adults) and Aedes japonicus, and dead Tripteroides bambusa were discovered in shipments from Japan (3 interceptions each in the first 2 cases, and one in the 3rd). Live Tripteroides tasmaniensis were recorded from the Australian cargo. One of the Ae. albopictus arrivals was followed by an apprehended introduction at an Auckland importer's premises.

Effect of intermittent irrigation on mosquitoes (Diptera: Culicidae) and larvivorous predators in rice fields.

Effect of intermittent irrigation to facilitate rice production on rice-field mosquitoes and larvivorous predators was studied in Saga, Japan. Mosquitoes and associated taxa were sampled by dipping in two rice fields subject to intermittent irrigation and in one rice field that remained flooded throughout the study period of approximately 1 mo. With the onset of intermittent irrigation, larval mosquito (Culex tritaeniorhynchus Giles and Anopheles sinensis Wiedemann) abundance decreased; reirrigated fields were colonized initially by first instars. Abundance of aquatic arthropod predators and larvivorous fish also decreased drastically. Water from irrigation canals was considered to be a major source of recolonization by these predators with limited breeding seasons and longer developmental periods than mosquitoes. Richness (number of taxa) of aquatic insect communities decreased, but the percentage mosquitoes among insects increased with repeated intermittent irrigation. Manipulation of rice fields and associated canals as a whole is essential to rice-field mosquito control by water management.

Selective removal of beta 2-microglobulin from plasma specimens of long-term hemodialysis patients by high-performance immunoaffinity chromatography.

We examined the suitability of high-performance immunoaffinity chromatography (HPIAC) for removing beta 2-microglobulin (beta 2-MG) from plasma of patients (n = 26) undergoing long-term hemodialysis. We used immobilized antibody to beta 2-MG to prepare an immunoaffinity column with a 0.5-mL bed volume. Plasma samples of 0.5 mL were chromatographed at a flow rate of 0.5 mL/min and pressures < 7 kg/cm2. Although only a small amount of beta 2-MG can be removed with a conventional dialyzer, 92.1% +/- 0.4% of the globulin could be removed by HPIAC. The column could be reused after acid elution. After 50 cycles the efficiency of beta 2-MG trapping was 73% of the original efficiency.

Enhancement by cyclosporin A of metastasis from hamster cheek pouch carcinoma.

Daily intravenous administration of 50 mg/kg cyclosporin A (CsA), after excision of cheek pouch tumors induced with 9,10-dimethyl-1,2-benzanthracene (DMBA), enhanced the incidence of cervical lymph node metastasis to 93%, a significant increase over the rates observed for animals receiving a dosage of 25 mg/kg (43%), and for control animals receiving no CsA (40%). No significant effect was evident on the growth rates of the metastasized tumour cells in the lymph node.

Effect of 4-MET on bond strength and penetration of monomers into enamel.

The effect of 4-[2-(methacryloyloxy)ethoxycarbonyl]phthalic acid (4-MET) in self-curing acrylic resin initiated by BPO-amine on the bond strength to etched enamel was studied. Scanning electron microscopic (SEM) observations of resin-enamel interfaces were carried out to elucidate the bonding mechanism and the effect of 4-MET on the adhesion. 4-MET, which has both hydrophilic and hydrophobic groups, increased the bond strength of self-curing acrylic resin to etched enamel and improved the bonding stability. SEM pictures strongly suggested that 4-MET promoted interpenetration of monomers into enamel prism peripheries and their polymerization therein resulted in excellent adhesion to etched enamel.

Seasonal distribution and habitat selection by Aedes albopictus and Ae. riversi (Diptera: Culicidae) in northern Kyushu, Japan.

Temporal and spatial distribution of adult and larval Aedes albopictus (Skuse) and Aedes riversi Bohart & Ingram were studied on a small island and coastal area of northern Kyushu, Japan. Adults of Ae. albopictus attracted to human bait were distributed uniformly in an evergreen forest, a citrus orchard surrounded by forest, and an area disturbed by humans, whereas Ae. riversi was confined to evergreen forest and its adjacent area. Adults of both species occurred from May through October. Although Ae. riversi outnumbered Ae. albopictus early in the season, Ae. albopictus were much more abundant than Ae. riversi later in the season, with maximum abundance in July-September. Larval Ae. riversi occurred primarily in tree holes but also were found in bamboo stumps and artificial containers in forests. Despite the abundance of adult Ae. albopictus in the forest, larvae occurred in tree holes less frequently than Ae. riversi; it more frequently exploited bamboo stumps, stone vases, and discarded tires outside the forest.

Biological control of container-breeding mosquitoes, Aedes albopictus and Culex quinquefasciatus, in a Japanese island by release of Toxorhynchites splendens adults.

To control container-breeding mosquitoes in the small island of Minnajima (0.56 km2), northern Okinawa, Japan, laboratory-reared adults (aged 7-10 days) of Toxorhynchites splendens (Palawan strain), a mosquito with predatory larvae, were released repeatedly during 1984, 1986 and 1987. Thirteen species of mosquitoes (Diptera: Culicidae) occurred in artificial containers, ground pools or crab-holes on the island, the predominant species being Aedes (Stegomyia) albopictus and Culex (Culex) quinquefasciatus. Predatory mosquito larvae of Culex (Lutzia) fuscanus and Cx (Lt.) halifaxii were found commonly in wet containers. In the first year of study, during a period of 54 days from 13 May to 5 July 1984, totals of 879 female and 806 male adults of Tx.splendens were released on six occasions. Similarly, between 29 April and 30 August 1986, totals of 2920 female and 2878 male adult Tx.splendens were released. In the third study year, totals of 2041 female and 1783 male Tx.splendens were released on eight occasions during 199 days from 23 April to 7 November 1987. After adult releases at two sites, the immature stages of Tx.splendens were found in 164 out of 502 traps in 1984, 421 out of 933 traps in 1986, and 151 out of 502 traps in 1987. The number of immatures of Tx.splendens present in each trap varied from 1 to 40 in 1984, 1 to 29 in 1986 and 1 to 9 in 1987. Numbers of immatures of the target species found in the traps during August-September averaged 71.9/trap/month in 1984, 114.7/trap/month in 1986 and 36.0/trap/month in 1987, significantly less in the traps with Tx.splendens than in those without them. The present field studies indicated that, in this small island, approximately 250 adult female and 200 male Tx.splendens per month should be released from April to November, and the releases should be carried out every year, in order to control effectively the target mosquitoes Ae.albopictus and Cx quinquefasciatus breeding in artificial containers in Minnajima.

Super-elasticity and thermal behavior of Ni-Ti alloy orthodontic arch wires.

Bending properties and thermal behavior of twenty commercial Ni-Ti alloy orthodontic arch wires were investigated quantitatively to characterize their suitability for clinical use. There was substantial difference among the load-deflection curves obtained by a three-point bending test. Some wires exhibited super-elasticity; load decreased little with decreasing deflection. Others showed good spring-back properties only; load was nearly proportional to deflection. Thermal behavior due to phase transformation of the alloy was examined by differential scanning calorimetry (DSC). Some of the wires did not have the correct transformation temperatures to exhibit super-elasticity at body temperature. Moreover, thermal behavior was closely related to super-elasticity. There were clear thermal peaks in the DSC curves of the super-elastic wires. However, wires without super-elasticity had no peak in the DSC curves.

Northernmost occurrences of the protistan pathogen, Coelomomyces stegomyiae var. stegomyiae.

Now reported as parasitizing the ovaries of female Aedes albopictus on Kabeshima (33.33N x 129.53E), Coelomomyces stegomyiae var. stegomyiae had been found earlier in larvae of this mosquito on Mikura-jima (33.53N x 139.35E). These findings from 2 widely separated little Japanese islands constitute the parasite's most northerly records to date. Few previous reports of Coelomomyces spp., none concerning C. stegomyiae, have involved adult mosquitoes. Possible reasons for this are advanced.
