Biochemical and physical analysis of subcellular membranes in rat parotid gland enlarged by chronic isoproterenol administration.

1. Chronic administration of isoproterenol caused similar alterations of membrane lipid profile in at least two rat parotid subcellular fractions, secretory granular and microsomal. 2. Typical changes in phospholipid classes and fatty-acyl chain groups were an increase of phosphatidylcholine and a decrease of sphingomyelin, and an increase of octadecadienoyl chain and a decrease of eicosatetraenoyl chain, respectively. 3. Electron spin resonance study showed that the isoproterenol-treatment also affected a membrane physical property, which may be through these compositional changes in membrane constituents.

An inhibitor of prostaglandin biosynthesis from human decidua: partial purification and properties.

An inhibitor of prostaglandin synthetase which catalyzes the conversion of arachidonic acid into prostaglandin E2 was partially purified from the 105,000 x g supernatant fraction of the human decidual cell homogenate. By means of ammonium sulfate fractionation, Mono Q ion-exchange chromatography, and gel filtration chromatography, the inhibitor was purified about 15-fold, giving a preparation with a molecular weight of 55-60 KDa. The 50% inhibitory concentration of the purified substance was approximately 0.2 mg/ml. The inhibitor may play a role in suppression of prostaglandin production by decidua in early pregnancy.

Upbeat nystagmus influenced by posture: report of two cases.

Two cases of vertical gaze nystagmus, which were influenced by posture are reported. In both cases, upbeat nystagmus (UBN) on forward gaze in upright and supine positions changed to downbeat nystagmus (DBN) in prone position. Convergence had no influence on the vertical nystagmus. A CT scan revealed an atrophy of the anterior vermis of the cerebellum in the first case. The UBN of the second case was suspected to be associated with lesions in the brachium conjunctivum. The authors concluded that both UBN and DBN were not completely separate, but they could coexist in cases with certain lesions in the posterior cranial fossa.

Detection of human papillomavirus types 6/11, 16 and 18 in exfoliated cells from the uterine cervices of Japanese women with and without lesions.

Human papillomavirus (HPV) infection of the uterine cervices of Japanese women with and without lesions was identified by the filter in situ hybridization method. Exfoliated cervical cells from 23 cervical papillary condylomas, 70 cervical intraepithelial neoplasia (CIN) grade I/II, 26 CIN III, 31 invasive cervical cancers and 666 cervices without evidence of disease (including 53 pregnant women) were tested for the presence of HPV types 6/11, 16 and 18. The positive rates for the detection of HPV types 6/11, 16 and 18 DNA were 47.8%, 26.1% and 8.7% in cervical condylomas, 5.7%, 15.7% and 8.6% in CIN I/II, 0, 34.6% and 0 in CIN III, 3.2%, 38.7% and 9.7% in invasive cervical cancers and 0.9%, 1.8% and 0.6% in the control cervices. These data suggest that, in Japan, HPV6/11, HPV16 and HPV18 infections are also prevalent in cervical cells with normal phenotype, and the type of HPV infection of the uterine cervix is related to the histological diagnosis.

Mid-cervical malignant pleomorphic adenoma presenting with upper airway obstruction.

The case of a 1 year and 6 months old girl with pleomorphic adenoma arising from the upper middle part of the neck is reported. Although the tumor invaded the hyoid bone, it was removed completely and symptoms of upper airway obstruction disappeared. We suspect it may have originated from thyroglossal duct structures.

Epidermal growth factor-like immunoreactivity in human endometrium.

Epidermal growth factor (EGF)-like immunoreactivity was detected in human endometrial tissues. The concentrations of the immunoreactivity fluctuated during the menstrual cycle. The highest levels were observed in the late proliferative phase with the concentrations being 0.50 +/- 0.06 ng/mg protein (M +/- SE, n = 5). Considering the presence of EGF receptors in human endometrium, EGF-like peptides may act in autocrine or paracrine fashions in human endometrium.

Transvaginal sonographic assessment of the endometrium in spontaneous and induced cycles.

Transvaginal sonography was used and endometrial response patterns of 3 groups of patients (Spontaneous cycles, hMG cycles, and Clomiphene Citrate cycles) were observed based on endometrial characteristics of thickness and texture. All groups demonstrated cyclical changes evident by transvaginal sonography. Clomiphene Citrate cycles showed retardation of endometrial growth during the proliferative phase and delayed endometrial development based on textural patterns. There exists a positive correlation between estrogen levels and endometrial thickness in induced cycles, which is not observed in spontaneous cycles. There were no correlations between hormonal levels of progesterone, FSH and LH, in relation to endometrial characteristics of thickness and texture.

Anaerobic energy production and O2 deficit-debt relationship during exhaustive exercise in humans.

1. Eight subjects performed one-legged, dynamic, knee-extensor exercise, first at 10 W followed by 10 min rest, then at an intense, exhaustive exercise load (65 W) lasting 3.2 min. After 60 min recovery, exercise was performed for 8-10 min each at 20, 30, 40 and 50 W. Measurements of pulmonary oxygen uptake, heart rate, blood pressure, leg blood flow, and femoral arterial-venous differences of oxygen content and lactate were performed as well as determination of ATP, creatine phosphate (CP) inosine monophosphate (IMP) and lactate concentrations on biopsy material from the quadriceps muscle before and immediately after the intense exercise, and at 3, 10 and 60 min into recovery. 2. Individual linear relations (r = 0.95-1.00) between the power outputs for submaximal exercise and oxygen uptakes (leg and pulmonary) were used to estimate the energy demand during intense exercise. Pulmonary and leg oxygen deficits determined as the difference between energy demand and oxygen uptake were 0.46 and 0.48 l (kg active muscle)-1, respectively. Limb and pulmonary oxygen debts (oxygen uptake during 60 min of recovery - pre-exercise oxygen uptake) were 0.55 and 1.65 l (kg active muscle)-1, respectively. 3. During the intense exercise, muscle [ATP] decreased by 30% and [CP] by 60% from resting concentrations of 6.2 and 22.4 mmol (kg wet wt)-1, respectively, and [IMP] increased to 1.1 mmol (kg wet wt)-1. Muscle [lactate] increased from 2 to 28.1 mmol (kg wet wt)-1, and the concomitant net lactate release was 14.8 mmol (kg wet wt)-1 or about 1/3 of the total net lactate production. During recovery 70% of the accumulated lactate was released to the blood, and the nucleotides and CP returned to about 40 and 85% of pre-exercise values at 3 and 10 min of recovery, respectively. 4. Total reduction in ATP and CP (and elevation of IMP) during the intense exercise amounted to 16.4 mmol ATP (kg wet wt)-1, which together with the lactate production accounted for 83.1 mmol ATP (kg wet wt)-1. In addition 6-8 mmol ATP (kg wet wt)-1 are made available related to accumulation of glycolytic intermediates including pyruvate (and alanine). Estimated leg oxygen deficit corresponded to an ATP production of 94.7 mmol ATP kg-1; this value included 3.1 mmol kg-1 related to unloading of HbO2 and MbO2.(ABSTRACT TRUNCATED AT 400 WORDS)

Limb skeletal muscle adaptation in athletes after training at altitude.

Morphological and biochemical characteristics of biopsies obtained from gastrocnemius (GAS) and triceps brachii muscle (TRI), as well as maximal O2 uptake (VO2 max) and O2 deficit, were determined in 10 well-trained cross-country skiers before and after a 2-wk stay (2,100 m above sea level) and training (2,700 m above sea level) at altitude. On return to sea level, VO2 max was the same as the prealtitude value, whereas an increase in O2 deficit (29%) and in short-term running performance (17%) was observed (P less than 0.05). GAS showed maintained capillary supply but a 10% decrease in mitochondrial enzyme activities (P less than 0.05), whereas an increase in capillary supply (P less than 0.05) but unchanged mitochondrial enzyme activities were observed in TRI. Buffer capacity was increased by 6% in both GAS and TRI (P less than 0.05). A positive correlation was found between the relative increase in buffer capacity of GAS and short-term running time (P less than 0.05). Thus the present study indicates no effect of 2 wk of altitude training on VO2 max but provides evidence to suggest an improvement in short-term exercise performance, which may be the result of an increase in muscle buffer capacity.

Stimulatory effect of some therapeutic drugs used for improving cerebral insufficiency on gastric acid secretion in rats.

Baclofen, gamma-amino-beta-hydroxybutyric acid (GABOB), calcium hopantenate (HOPA) and citicoline, which are clinically used for improving cerebral insufficiency, were tested for their effects on gastric acid secretion in the perfused stomach of urethane-anesthetized rats. These drugs were administered intravenously or subcutaneously. They produced a significant increase in gastric acid secretion, although the extents in increase were different. The duration of action was 1-2 h. The potency order was baclofen greater than GABOB greater than or equal to citicoline greater than or equal to HOPA. The stimulatory response to each drug was completely inhibited by atropine. In contrast, the peripheral secretory activity of each drug was not demonstrated in the isolated lumen-perfused stomach of mice. These results indicate that all the tested drugs produce an increase in gastric acid secretion probably via central cholinergic activation and that gastric acid stimulation by compounds improving the cerebral insufficiency might be included in side effects in their clinical use.

A syndrome of hyperandrogenism, insulin resistance, and acanthosis nigricans associated with polycystic ovary syndrome: clinical and laboratory features.

We describe an adolescent Japanese girl with acanthosis nigricans and irregular anovulatory menstruation following menarche. Serum LH levels were elevated, whereas serum FSH levels were within normal range. An exaggerated response to LHRH was observed. Further, serum androstenedione levels were markedly elevated. Ultrasonogram revealed bilateral polycystic changes of ovaries. She had a mild degree of insulin resistance. Insulin binding studies using erythrocytes demonstrated a decreased binding capacity of insulin. From the above findings, this patient presents the syndrome consisting of hyperandrogenism, insulin resistance, and acanthosis nigricans and also has clinical and biochemical features compatible with polycystic ovary syndrome.

Development of artificial placenta: oxygen metabolism of isolated goat fetuses with umbilical arteriovenous extracorporeal membrane oxygenation.

To assess the oxygen utilization of extrauterine goat fetuses with arteriovenous extracorporeal membrane oxygenation via umbilical vessels, oxygen delivery (OD) and oxygen consumption (OC) were investigated in 19 long-term incubations (incubation periods 58-236 h). The OCs did not change (6.4 +/- 1.3 ml/min/kg, mean +/- SD, n = 227) when the ODs were more than 10 ml/min/kg, but were significantly suppressed when the ODs were less than 10 ml/min/kg. It was suggested that the oxygen metabolism of extrauterine goat fetuses is equivalent to that of fetuses in utero.

Enhancement of the mutagenicity of Trp-P-1, Trp-P-2 and benzo[a]pyrene by bupleuri radix extract.

Boiling-water extract of Korean-Saiko (Bupleuri Radix, from South Korea, Bupleurum falcatum L.) enhanced the mutagenic activity of Trp-P-1, Trp-P-2 and benzo[a]pyrene with S9mix. The boiling-water extract was fractionated with ether and then n-BuOH. Both the ether and the n-BuOH fractions also enhanced mutagenicity of Trp-P-1, respectively. The n-BuOH fraction was separated into seven fractions by silica gel chromatography and the chloroform eluate had the strongest enhancing effect on the mutagenic activity of Trp-P-1 with S9mix. The chloroform eluate fraction was further separated into five spots by thin-layer chromatography. Two of the spots had the strongest enhancing effect on the mutagenic activity of Trp-P-1. Since saikosaponin is a well known component in Bupleuri Radix, the effects of its existence were tested and saikosaponins a and c were found. The enhancement activity of saikosaponin a was very weak. The effective components are now being studied.

[Phagocytic activity of leaves of Epimedium species on mouse reticuloendotherial system].

Effects of a 70% methanolic extract (E-S) from the dried leaves of Epimedium sagittatum on a phagocytic activity of reticuloendothelial system were studied by the carbon clearance method in mice. The clearance-rate of carbon significantly increased 1 h after the oral administration of E-S (200 or 500 mg/kg, one time/d for 5d). E-S activated the phagocytosis of carbon by Kupffer cells in the liver. Icariin and epimedin C isolated from E-S also activated the phagocytosis. These results suggest that E-S promotes the phagocytic activity of the reticuloendothelial system in mice and has a stimulatory effect on macrophage.

Centrally mediated inhibitory effect of 5-[2-(diethylamino)ethyl]amino-5,11-dihydro[1]benzoxepino[3,4- b]pyridine trihydrochloride (KW-5805) on gastric acid secretion in rats.

KW-5805, 5-[2-(diethylamino)ethyl]amino-5,11-dihydro[1]benzoxepino[3,4- b]pyridine trihydrochloride, is a new tricyclic compound with antiulcer activities. Its effect on stimulated gastric acid secretion was investigated in the perfused stomach of anesthetized rats. KW-5805 at 0.3-10 mg/kg, i.v., dose-dependently inhibited gastric acid secretion stimulated by 2-deoxy-D-glucose (2-DG). On the other hand, the compound at 10-20 mg/kg, i.v., exerted a moderate decrease in gastric acid secretion stimulated by bethanechol; and at 10 mg/kg, i.v., it produced no change in gastric acid secretion evoked peripherally by vagal electrical stimulation. When applied intracerebroventricularly at 1-5 micrograms/rat, this compound dose-relatedly reduced gastric acid secretion stimulated by 2-DG. Three main metabolites (KF-10504, KF-9530 and KF-10847) of KW-5805 at 1 mg/kg, i.v., caused no significant decrease in gastric acid secretion stimulated by 2-DG. Doxepin, a tricyclic compound, definitely depressed the 2-DG stimulated gastric acid secretion at 1 mg/kg, i.v. It is suggested that intravenous administration of KW-5805 inhibits gastric acid secretion stimulated by 2-DG, mainly via centrally mediated mechanisms, and that biotransformation of KW-5805 to the metabolites contributes little to the development of the antisecretory effect.

Estradiol stimulates the binding of epidermal growth factor in cultured rat granulosa cells.

The effect of estradiol on the binding of epidermal growth factor (EGF) was investigated in a rat granulosa cell culture system. Cultured granulosa cells without estradiol possess an EGF receptor with a Kd value of 7.4 x 10(-10) M and binding sites of 1.45 x 10(3) molecules per cell. The addition of estradiol to the culture media resulted in an increase in the binding of EGF in a dose-dependent manner at concentration between 10(-9) M and 10(-7) M. Estradiol at 10(-7) M produced a 2.5-fold increase in the number of binding sites without affecting the binding affinity. An increase in the binding was observed as early as after 6 h incubation. The addition of testosterone alone had no effect on the EGF binding. However, in the presence of follicle stimulating hormone, it increased the binding dose-dependently. The stimulatory effect of testosterone was completely abolished by an aromatase inhibitor, thus suggesting that estradiol converted from testosterone could be a contributing factor in the mechanism of testosterone action. These observations imply that estradiol may play a role in regulating growth and the differentiation of granulosa cells by modulating the EGF binding.

Contrast medium extravasation during cerebral angiography for ruptured intracranial aneurysm--clinical analysis of 26 cases.

Since 1976, the authors performed angiography on 837 patients with ruptured intracranial aneurysm, among which extravasation (EV) of contrast medium occurred in 26. Eleven patients underwent radical surgery within 3 hours after the EV, and four recovered well. The other 15 patients were conservatively treated, and all died. A large quantity of EV occurred when consciousness was severely disturbed and angiography was performed within 3 hours of the last attack. Most EV resulted in poor outcome. However, some patients with mildly disturbed consciousness before angiography, even with severe temporary disturbance of consciousness after the EV, recovered well after emergency radical surgery.
