[Toxicological studies on (2''R)-4'-O-tetrahydropyranyladriamycin, a new antitumor antibiotic. Its acute toxicity in rats].

(2''R)-4'-O-Tetrahydropyranyladriamycin (THP), a new antitumor antibiotic of anthracycline derivative, was given to Jcl-SD strain rats through intravenous (i.v.), intraperitoneal (i.p.), subcutaneous (s.c.) or oral (p.o.) administration routes and the animals were observed in respect of mortality, clinical signs and body weight for 21 days. Autopsy was done and histopathology on the tissues showing macroscopic abnormality was performed. The results were summarized as follows. Values of LD50 were 18.09 mg/kg i.v., 22.58 mg/kg i.p., 25.39 mg/kg s.c. and above 1,013 mg/kg p.o. for males and 18.07 mg/kg i.v., 20.30 mg/kg i.p., 21.76 mg/kg s.c. and above 1,013 mg/kg p.o. for females. No significant difference was found in LD50 values of different sexes. When higher than lethal dose levels of THP was given to animals, their clinical signs grew worse and weight loss occurred in about 5 days after the administration of the drug. Thereafter, deaths were observed. Macroscopic and microscopic observations on dead and survived rats revealed atrophy of spleen and thymus, whity clouding of spleen capsule, hemorrhage in mucosa of glandular stomach and congestion and hemorrhage in testes. These results suggest that THP shows weaker acute toxicity to rats than doxorubicin does, but the toxic effect of THP is approximately the same as that of other anthracycline derivatives.

The distance separating Cys-10 from the high-affinity metal binding site in actin.

The fluorescence resonance energy transfer between 5-[2-[iodoacetyl)amino)ethyl]aminonaphthalene-1-sulphonic acid (1,5-IAEDANS) attached to Cys-10 residue and Co2+ bound to the high affinity metal site was measured. The resonance energy transfer efficiency was 8 +/- 2%, which corresponds to a distance of 2.1 nm using the absorption spectrum of Co-EDTA and 3.0 nm using the more relevant absorption spectrum of Co2+ bound to carboxypeptidase as a model spectrum of Co2+ bound to actin, respectively.

Selective labelling of Cys-10 on actin.

Conditions are described for the selective modification of Cys-10 on actin achieved following the blocking of the more reactive Cys-374. Labelling of Cys-10 did not affect the formation of actin filaments. This residue should be capable of serving as a site for fluorescence donors or acceptors and thus will be a useful locus to probe F-actin structure.

[Clinical evaluation of norfloxacin in gonococcal urethritis].

Norfloxacin, a new quinolinecarboxylic acid derivative, was administered to 30 male patients with gonococcal urethritis at a daily dose of 600 mg for 7-21 days. The clinical response was evaluated after administration of 7 days as excellent; Negative culture of N. gonorrhoeae. WBC less than 3/hpf in first voided urine sediment, good; Negative culture, WBC greater than or equal to 3/hpf, and poor; Positive culture. The result was excellent in 14 cases and good in 16 cases. No subjective side effects were observed. The minimum inhibitory concentration (MIC) distribution against the clinically isolated 30 strains of N. gonorrhoeae ranged from 0.0096 micrograms/ml to 0.34 micrograms/ml. Seven of thirty strains were resistant to ABPC. The MIC of these 7 strains ranged from 0.018 micrograms/ml to 0.18 micrograms/ml. Seven cases with ABPC resistant strains had a similar clinical response to other cases. Twelve patients (40%) developed post gonococcal urethritis for 7-14 days after treatment. Clinical observation of this series suggests that a 7 day therapy of Norfloxacin for the patients with gonococcal urethritis is sufficiently effective and that treatment should be changed to other antibacterial agents in the case of post gonococcal urethritis, since continuous administration for more than 7 days of Norfloxacin is not so effective.

The distances separating Tyr-69 from the high-affinity nucleotide and metal binding sites in actin.

The nucleotide binding site in actin was occupied with the fluorescent analogue formycin A 5' triphosphate which acted as a fluorescent donor for the acceptor chromophore dansyl chloride attached to Tyr-69. The distance separating the two chromophores was calculated to be 2.1 nm from the fluorescence energy transfer measurements. Similar measurements were made of the distances separating dansyl chloride, acting as donor, on Tyr-69 from Co2+ occupying the metal binding site. A distance of 2.1 nm was similarly obtained.

Fluorescence energy transfer between points in G-actin: the nucleotide-binding site, the metal-binding site and Cys-373 residue.

Fluorescence energy transfers were studied in order to investigate the spatial relationships between the nucleotide-binding site, the metal-binding site and the Cys-373 residue in the G-actin molecule. When 1-N6-ethenoadenosine-5'-triphosphate (epsilon-ATP) in the nucleotide-binding site and Co2+ or Ni2+ in the metal-binding site were used as fluorescence donor and acceptor, respectively, the fluorescence intensity of epsilon-ATP was perfectly quenched by Ni2+ or Co2+. This indicated that the nucleotide-binding site is very close to the metal-binding site; the distance should be less than 10 A. When N-iodoacetyl-N'-(5-sulfo-1-naphthyl)ethylenediamine (IAEDANS) bound to Cys-373 residue and Co2+ in the metal-binding site were used as a fluorescence donor and an acceptor, respectively, the transfer efficiency was equal to 5 +/- 1%. The corresponding distance was calculated to be 23-32 A, assuming a random orientation factor K2 = 2/3.

Plasminogen activator activity of cultured endothelial cells derived from canine coronary vessel and human umbilical artery and vein.

The present study was undertaken to determine which cells participate in plasminogen activator (PA) release from the vascular wall. Canine coronary vessel was confirmed to release PA when various agents were administered in perfusion experiments. Endothelial cells from the coronary vessel were then isolated and cultured for 2 days. PA activity was observed in lysates of the cultured cells, and the medium used for the cultivation was found to contain little PA activity. This suggests that the endothelial cells participate in PA release from the vascular wall. In addition, the PA synthesis was also studied in cultured endothelial cells from human umbilical artery and vein. Both contained little PA activity, indicating that vascular endothelial cells may vary in PA synthesis and release according to the vessel.

Chemical modification of tropomyosin with NBD-chloride.

Muscle tropomyosin was modified with 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole (NBD-chloride) at several different pH values. NBD-chloride reacts specifically with SH residue at neutral pH but it reacts with both SH residue and amino residues at alkaline pH. The polymerizability of tropomyosin at low ionic strength and the binding property of tropomyosin to F-actin were not affected by the modification of SH residues but they were lost rapidly by the modification of amino groups, in accordance with the previous report [Johnson, P. & Smillie, L.B. (1977) Biochemistry 16, 2264-2269]. By the addition of heavy meromyosin, labeled tropomyosin which could not bind to F-actin recovered the binding ability to F-actin and it could regulate the superprecipitation of actomyosin in the presence of troponin. Further modification of amino groups (labeling ratios more than 5) led to loss of the regulating ability completely.

Comparative changes in plasma and RBC alpha-tocopherol after administration of dl-alpha-tocopheryl acetate and d-alpha-tocopherol.

Changes in plasma and RBC alpha-tocopherol after intravenous and oral administration of dl-alpha-tocopheryl acetate were investigated using vitamin E-deficient rats and were compared with those following the administration of d-alpha-tocopherol, which is the most active and natural form of tocopherol. Intravenous administration: By the administration of dl-tocopheryl acetate, the elevated plasma tocopherol levels were higher at 6 h than those elevated by d-tocopherol, but the RBC tocopherol was lower 6 h after the infusion of dl-tocopheryl acetate than after d-tocopherol infusion. The tocopheryl acetate incorporated into the RBCs remained for 24 h after the administration of tocopheryl acetate, the acetate then becoming undetectable in plasma. Oral administration: The tocopherol elevated in plasma and RBC was only the alpha-form and not tocopheryl acetate. In both plasma and RBCs, tocopherol levels were higher after the administration of d-tocopherol than after dl-tocopheryl acetate administration, and more was found in RBCs.

[Tumor markers of prostate cancer--evaluation of serum PAP and PA].

The clinical usefulness of PAP and PA as a tumor marker for the prostate cancer were discussed. The materials for this study were 1385 cases which contained 158 cases with prostatic carcinoma. The positive rate of serum PAP and PA were 77.7% and 94.1% in untreated prostatic carcinoma and 15.1% and 70.0% in benign prostatic hypertrophy using 3.0 ng/ml as an upper limit of normal controls of serum PAP and PA. The cut off level in serum PA should be discussed more. PA was not superior to PAP as a tumor marker in the series, but our results have suggested the simultaneous assay of serum PAP and PA is valuable in detection and following-up of prostate cancer.

[Diagnosis and treatment of renal angiomyolipoma].

We have hitherto reported 6 cases of renal angiomyolipoma. Recently, we encountered two more such cases. Case 1 is a 34-year-old woman with fever as the chief complaint. DIP revealed a tumor mass in the right upper pelvic pole. This mass showed a strong echo level on ECHO and adipose tissue of low density on CT scan. Therefore, the patient was diagnosed as having renal angiomyolipoma. Since liposarcoma was not ruled out by the examination of frozen sections during operation, nephrectomy was performed. Case 2 is a 40-year-old woman. Diagnosed as having bilateral renal angiomyolipoma, she underwent right nephrectomy 14 years ago. Two years ago, she had heavy hematuria, and had embolization of the left renal artery. She has had no bleeding since the embolization. We are of the view that ECHO and CT can are very useful for diagnosis of renal angiomyolipoma, and embolization for heavy hematuria, a complication, should be performed first of all.

Fluorescence energy transfer measurements between the nucleotide binding site and Cys-373 in actin and their application to the kinetics of actin polymerization.

Intramonomer fluorescence energy transfer between the donor epsilon-ATP bound to the nucleotide-binding site and the acceptor 7-chloro-4-nitrobenzo-2-oxa-1,3-diazole bound to Cys-373 in G-actin was measured by steady-state fluorimetry. Assuming for the orientation factor its dynamic limit K2 = 2/3, the donor and acceptor distance in a G-actin molecule was calculated to be about 3 nm. The intermonomer energy transfer in F-actin occurring between the donor bound to an actin monomer and the acceptor bound to the nearest-neighbour actin monomer was also measured and the distance was calculated to be about 4 nm. The kinetics of the actin polymerization process was studied by following the decrease in fluorescence intensity upon addition of salts to G-actin solution. The initial velocity of the fluorescence intensity change was proportional to the square of the initial G-actin concentration. The temperature dependence of the velocity was proportional to the square of the initial G-actin concentration. The temperature dependence of the velocity was proportional to exp(-10/RT). These results indicated that the initial fluorescence intensity change corresponds to monomer-dimer transformation and its activation enthalpy was 10 kcal/mol.

Fluorescence energy transfers between points in acto-subfragment-1 rigor complex.

Fluorescence energy transfer was measured by time-resolved and steady-state fluorimetry in order to investigate the spatial relationships between the nucleotide binding site of actin, the Cys-373 residue of actin, and the SH1 of myosin subfragment-1 in the rigor complex of acto-subfragment-1. N-Iodoacetyl-N'-(5-sulfo-1-naphthyl)ethylenediamine (IAEDANS) bound to the Cys-373 of actin or the fluorescent ADP analogue 1-N6-ethenoadenosine-5'-diphosphate (epsilon-ADP) bound to F-actin was used as a donor and 4-(N-(iodoacetoxy)ethyl-N-methyl)amino-7-nitrobenz-2-oxa-1,3-diazo le (IANBD) or 5-iodoacetamidofluorescein (IAF) bound to SH1 of myosin subfragment-1 was used as an acceptor. Assuming the random orientation factor, K2, to be 2/3, the distance between Cys-373 residue of actin and SH1 of myosin subfragment-1 was calculated to be about 50 A, in agreement with the values previously reported, 60 A (Takashi, R. (1969) Biochemistry 18, 5164-69) and 50 A (Trayer, H.R. and Trayer, I.P. (1983) Eur. J. Biochem. 135, 47-59). The distance between the nucleotide binding site of actin and SH1 of myosin subfragment-1 was calculated to be about 70 A or greater.

Prostacyclin release from the coronary vascular wall by vasoactive substances.

Which vasoactive substances that are synthesized in vivo could induce the release of a sufficient amount of prostacyclin (PGI2) to inhibit platelet aggregation from the vascular wall was investigated in the isolated dog heart perfused by a modified method of Langendorff. Infusion of 5 microM bradykinin or 25 u/ml crude thrombin into the heart for 30 sec resulted in the transient appearance of inhibitory activity of platelet aggregation. The inhibitory activity was stable at alkaline pH but unstable at acidic pH and thermolabile. The appearance of the inhibitory activity was prevented by treatment of the coronary vessel with 30 microM indomethacin or 1 mM tranylcypromine. These results indicated that the inhibitory activity was caused by PGI2. When 25 microM acetylcholine, 25 microM noradrenaline, 25 microM isoproterenol, 10 microM adenosine triphosphate (ATP), 5 microM adenosine, 1 microM angiotensin II, 25 microM histamine or 1 microM serotonin was infused for 30 sec, no inhibitory activity of platelet aggregation was observed. Bradykinin (5 X 10(-9) approximately 5 X 10(-6) M) and purified thrombin (1 X 10(-9) approximately 1 X 10(-7) M) induced a dose-dependent release of PGI2 which was assayed using a radioimmunoassay for 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha).
