Immunologic abnormality in NZB/NZW F1 mice. Thymus-independent occurrence of B cell abnormality and requirement for T cells in the development of autoimmune disease, as evidenced by an analysis of the athymic nude individuals.

Both NZB nu/+ and NZW nu/+ mice were microbially clean by cesarean section. The (NZB x NZW)F1 hybrid (NZB/W) nu/nu mice and nu/+ littermates were then generated by mating of NZB nu/+ with NZW nu/+mice under specific pathogen-free conditions. The female NZB/W F1 nu/nu mice did not develop autoimmune kidney disease, whereas all of nu/+ female littermates mice exhibited proteinuria and died of renal failure with a 50% survival time of 35 wk. Namely, nude mice had no signs of proteinuria up to the time of their death caused by other diseases rather than glomerulonephritis, and their mean survival time was greater than 45 wk. Nude mice had also no anti-ssDNA antibody in their serum. However, splenic B cells of NZB/W nude mice exhibited hyper-responsiveness to both LPS and B151-TRF2, a T cell-derived polyclonal B cell-stimulation factor, and produced large numbers of Ig-secreting cells and anti-TNP plaque-forming cells as well as anti-ssDNA antibody comparable to the nu/+ littermate mice. Interestingly, thymus-engrafted NZB/W nude mice developed autoimmune disease exemplified by the induction of anti-ssDNA antibody and proteinuria at approximately the same time as their nu/+ littermates. These results indicate that the B cell hyper-responsiveness found in NZB/W mice is apparently determined by the T cell-independent process, and T cells are obligatorily required for the development of autoimmune disease in NZB/W mice.

Scanning electron microscopy of skin surface and the internal structure of corneocyte in normal human skin. An application of the osmium-dimethyl sulfoxide-osmium method.

A horny layer of normal human skin prepared according to the newly developed osmium-dimethyl sulfoxide-osmium method was examined using scanning electron microscopy. On the skin surface, cytomembranes of the uppermost corneocytes frequently had unilateral, very slightly flat-elevated, zonal areas along the junctions between the corneocytes. The uppermost corneocytes peeled off along the junctions, leaving the remnants of their cell bodies in the junctional areas. In the cracked surface, cytomembranes of the corneocytes protruded from the plane of their cytoplasmic surface. In the lateral junction between the corneocytes, the cytomembranes of the corneocytes were in tight contact with each other, while occasionally the marginal bands had become detached from the cytomembranes. In the vertical connection, cleavages formed between the cytomembranes of the corneocytes. There were thick woollen thread-like structures about 10-30 nm thick in the cytoplasm of the corneocytes. They formed fine irregular meshworks, with their tips projecting digitally: Transmission electron microscopy revealed these structures as most likely being keratin bundles transformed during processing.

An immunomodulating anti-rheumatic drug, lobenzarit disodium (CCA): inhibition of polyclonal B-cell activation and prevention of autoimmune disease in MRL/Mp-lpr/lpr mice.

In this study, we examined the effect of an immunoregulatory antirheumatic agent, lobenzarit disodium (CCA), on spontaneously developing glomerulonephritis in MRL/Mp-lpr/lpr (MRL/l) mice. Starting from 6 weeks of age, mice were given CCA orally 5 days a week at a dose of 2 or 10 mg/kg. A control group was given the same volume of distilled water. The CCA treatment suppressed the excretion of protein in the urine. At 40 weeks of age, the incidence of proteinuria was 10/10 in the controls, 6/10 in the 2-mg/kg treatment group, and 5/10 in the 10 mg/kg group. The life span was prolonged dose dependently. The 50% survival time was 33 weeks for the controls, 35.5 weeks for the 2-mg/kg group, and 41 weeks for the 10-mg/kg group. The serum levels of anti-ssDNA antibody, anti-TNP antibody, and rheumatoid factor (RF) of the Ig G isotype and immune complex were reduced compared with control group. But the antibodies of Ig M isotype were not reduced. The serum Ig G1, Ig G2, and Ig G3 were significantly lower in the CCA-treated mice than in the controls. But again the serum level of Ig M was unchanged. These effects of CCA may be based on the suppression of lymphadenopathy. CCA may correct abnormal B-cell growth and differentiation factor release by the MRL/l abnormal T cells. These results show that CCA inhibits the development of lupus nephritis in MRL/l mice through the amelioration of the abnormal immune response, polyclonal B-cell activation.

Effect of immunostimulants and antitumor agents on tumor necrosis factor (TNF) production.

OK-432, a lyophilized preparation of Streptococcus pyogenes, showed a priming activity for TNF production in mice, associated with an increase of spleen weight. PSK, a protein-bound polysaccharide preparation from Coriolus versicolor, did not show such activity. Both OK-432 and PSK potentiated the TNF production in mice primed with Corynebacterium parvum (CP) and challenged with Escherichia coli endotoxin (LPS). Cytotoxic antitumor agents of 5-fluorouracil (5-FU), cyclophosphamide (CY) and bleomycin (BLM) suppressed TNF production in mice primed with CP and challenged with LPS. TNF production suppressed by 5-FU, CY and BLM was partially restored by the combined treatment with OK-432 or PSK. These results suggest that the administration of cytotoxic antitumor agents suppresses the intrinsic TNF production in cancer patients, and the combined use of immunostimulants such as OK-432 and PSK is advantageous in restoring TNF production suppressed by cytotoxic antitumor agents.

Pseudotail associated with spinal dysraphism.

A 5-year-old girl had a caudal appendage and her left buttock was larger than the righ buttock. X-ray examination revealed spina bifida and bony defect of sacrum; computed tomography demonstrated the extension of the tumor from subcutaneous tissue to the spinal canal. Histologically, the pseudotail contained lobulated fatty tissue which was consistent with lipoma. It emphasizes the fact that even lesions that are not situated in the median line should be carefully explored before excision.

Perineural xanthoma.

A 50-year-old man suffering from cholestatic hepatitis and diabetes mellitus with hyperlipoproteinaemia had small, painful, slightly elevated, reddish, firm indurated plaques on his soles. Histologically, the lesions were composed of a centrally located cutaneous nerve surrounded by concentric layers of xanthoma cells. Electron microscopy showed the cutaneous nerves to be unmyelinated, their axons were vacuolated and contained dense bodies. The xanthoma cells had the same ultrastructural features as those observed in usual xanthomatous lesions. We suggest that this entity be named perineural xanthoma.

Superficial spreading capillary hemangioma. A peculiar type of capillary hemangioma.

A 57-year-old Japanese woman had a history of several years of a slowly growing, dark red plaque on the sole of her foot without any subjective symptoms. Histopathologically, the lesion was a capillary hemangioma which showed a lobular structure composed of newly formed capillaries. Each lobule was separated by thin elongated rete ridges. The lesions was definitely circumscribed in papillary and subpapillary layers and appeared to extend laterally.

Posttraumatic occurrence of infantile digital fibromatosis. A histologic and electron microscopic study.

We encountered a patient with infantile digital fibromatosis. The representative lesion was a firm, indurated nodule, which developed in the scar of a previous injury on the skin of the left thigh of an 11-year-old girl. Electron microscopy revealed tumor cells that were strongly reminiscent of myofibroblasts. Histopathologically, the cytoplasmic inclusion bodies in the tumor cells stained positively or negatively with hematoxylin-eosin. Electron microscopically, they were either well-defined dense bodies that often showed a doughnutlike appearance or ill-demarcated dense bodies. Spotty calcified foci were seen in the central area of the tumor mass.

Regenerated cutaneous nerves in human epidermal and subepidermal regions. An electron microscopy study.

To elucidate the regeneration of cutaneous nerves in human skin, biopsy specimens were taken from the regenerated skin of extremities of five adult men with third degree burns that had occurred about 3 weeks previously. Numerous subepidermal nerves could be detected in the newly formed granulation tissue, independent of the blood vessels. These nerves were axon-Schwann's cell complexes. Some axons were packed with small clear vesicles 40-60 nm in diameter or large dense-cored vesicles 70-100 nm in diameter and large dense bodies. Schwann's cell components showed a relatively abundant cytoplasm and many microfilaments. Intraepidermal nerves were frequently encountered in the interspace between the basal lamina and the cytomembrane of basal keratinocytes in most cases or in the intercellular spaces between basal keratinocytes; however, they were never seen in and beyond spinous layers. They were seen as axon-Schwann's cell complexes or naked axons. In one case a Schwann's cell containing axons migrated into the epidermis. These findings suggest that cutaneous nerves may show hyperregeneration in very early stages of wound healing.
