Fibrous hamartoma of infancy. A study of eight cases with immunohistochemical and electron microscopical findings.

Eight cases of fibrous hamartomas of infancy are presented. Actin positivity and desmin negativity in collagen forming spindle-shaped cells and electron microscopical findings point to the myofibroblastic nature of this entity. "Dark cells" which spread regularly in all but one of our tumors were immunohistochemically determined to be a mixture of B and T cell lymphocytes. Their selective location and possible role are discussed.

Papillary carcinoma of thyroid with exuberant nodular fasciitis-like stroma.

We describe two cases of papillary carcinoma of the thyroid containing prominent nodular, fasciitis-like stroma. In one of the cases infiltration into the adjacent parathyroid gland and metastases to two cervical lymph nodes occurred. In the lymph nodes and the parathyroid gland the carcinoma grew without any fasciitis-like stroma. This unusual change in tumour stroma seems to be reactive in nature and confined only to the thyroid and adjacent soft tissues.

Myxoid change within cellular blue naevi: a diagnostic pitfall.

We describe four cases of a distinctive myxoid change located in the centre of cellular blue naevi. Because all the reported lesions were large and were present on pressure-exposed locations, the myxoid change is thought to be caused by pressure together with direct repeated minor tissue trauma.

Embryonal form of extraskeletal myxoid chondrosarcoma with intermediate filament positive hyaline-like globules.

Reported is a case of an embryonal form of extraskeletal myxoid chondrosarcoma. The tumor cells contained PAS-positive, eccentrically located intracytoplasmatic, hyaline-like globules. These globules seemed to be a hallmark of this tumor, and they reacted strongly with cytokeratin and S-100 protein antibodies. The tumor was negative in reactions with desmin and GFAP antibody.

Myoid differentiation in vascular transformation of lymph node sinuses due to venous obstruction. Immunohistochemical and ultrastructural studies.

Five cases of vascular transformation of lymph node sinuses due to venous obstruction are described in this paper. Evidence was found in four cases to actin immunoreactivity, and involvement of myofibroblasts was electronmicroscopically observed in one case. Differential diagnosis of vascular transformation of lymph node sinuses is discussed. The lesion is similar to KAPOSI's sarcoma-like subcutaneous processes and to chemodectoma-like bodies in the lung.

Collagenous spherulosis. A comment on its histogenesis.

Collagenous spherulosis is a benign breast lesion involving lobular acini and ductules consisting of eosinophilic spherules measuring up to 100 mu in diameter. It is a myoepithelial product. We described similar lesions in salivary gland tumors and a benign lymphoepithelial lesion of the parotid gland.

Epimyoepithelial carcinoma of parotid gland.

Epimyoepithelial carcinoma (EMC) of salivary gland origin, primarily in the parotid gland, is a tumor in its own right and of low malignancy, usually occurring in women. Our patient is alive with her second recurrence, 16 years after the first surgical intervention. Immunohistochemical results suggest active involvement of myoepithelial and epithelial components of intercalated ducts in the histogenesis of this particular tumor.

Cleaved and multilobated cell immunocytoma of low grade malignancy with amyloid. An immunohistochemical and electron microscopical study.

An unusual variant of immunocytoma of low grade malignancy, cytologically resembling follicular cleaved-cell malignant lymphoma is described. Presence of heavy deposits of amyloid, intracytoplasmic monoclonal IgM-kappa immunoglobulin, cells with Dutcher inclusions, electron-microscopic features and negativity of T-cell markers led to diagnosis of immunocytoma. After cytostatic chemotherapy the lymphoma changed its morphology into typical lymphoplasmacytoid immunocytoma.

Reduced thrombus formation in vivo after administration of pentoxifylline (Trental).

The hamster cheek pouch model of experimental thrombosis in which thrombi are induced in the microvasculature by iontophoretical administration of adenosine diphosphate (ADP) was used to test the antithrombotic potential of pentoxifylline (Trental). Single intraperitoneal injections of 5, 10 and 20 mg/kg pentoxifylline reduced thrombus formation by 20 to 50% from 30 to 105 min following drug administration. The effect of a single application of 10 mg/kg was exceeded significantly (p less than 0.05) by the higher rate of inhibition after repeated injections of the same dose given three times daily. This suggests a residual antithrombotic effect from the preceding administrations.

[Immunohistochemical detection of triiodothyronine in thyroid tumors using T3-01 monoclonal antibody].

Cells of the spleen of BALB/c mice immunized with a conjugate of triiodothyronine and bovine immunoglobulin were fused with cells of the murine myeloma line F O. Selection and cloning were used to look up the T3-01 hybridoma producing a monoclonal antibody of the IgG1 subclass which responded in a radioimmunoassay specifically to triiodothyronine. The T3-01 monoclonal antibody was used for the immunohistochemical demonstration of triiodothyronine on 19 tumour derivatives.

Inhibition of sorbitol formation in human erythrocytes by calcium dobesilate.

The effects of calcium dobesilate (Doxium, a drug already known to prevent or reverse the retinal microvascular lesions of diabetes mellitus) on sorbitol formation in human erythrocytes was studied, and compared with the effects of tetramethylene glutaric acid (TMGA), a specific aldose reductase inhibitor. Both compounds inhibited sorbitol formation. In a 29 mmol/l glucose medium, calcium dobesilate in concentrations of 2 and 10 mmol/l caused, 50 and 100% inhibition of sorbitol formation, respectively, whereas TMGA completely inhibited sorbitol formation at a concentration of 0.1 mmol/l. These results suggest that calcium dobesilate may exert, at least in part, its beneficial effect on diabetic microangiopathy by inhibiting the formation of sorbitol. The inhibition of sorbitol synthesis may be mediated by an inhibitory action of calcium dobesilate on aldose reductase.

[Early repositioning of fractures of the forearm, wrist, leg and navicular bone using an extension device attached to a bed].

The author informs on the results of early reposition of fractures on an extension device on an emergency bed of the author's design which meets the demands laid on urgent, sparing and high quality reposition already in the out-patient department, which is an advantage in particular in out-patient departments which are far from the hospital. Another advantage is, as apparent from results that reposition were successful in 273 (98.2%) of the patients despite the fact that they were made without an X-ray intensifier of the picture with a television screen. In all children and adults with fractures of the forearm, wrist, leg and navicular bone after clinical examination of the patient the X-ray pictures were made and checked only after application of a plaster bandage. The advantages of early reposition on the extension device should stimulate their use in all ambulances and in-patient departments where patients with above fractures are treated.

Effect of calcium dobesilate on platelet function.

Platelet-active drugs are of potential benefit in prevention of diabetic microangiopathy as well as other thromboembolic complications. The present investigation examines the effect of an angio-protective agent, calcium dobesilate (Doxium) on platelets and suggests its mechanism of action. Calcium dobesilate reduces aggregation and the release reaction induced by thrombin and collagen in rabbit platelets. Calcium dobesilate also increases platelet cAMP levels in vitro and ex-vivo. The experimental results indicate that the inhibitory effect of calcium dobesilate on platelet function is mediated through the cyclic AMP pathway, probably through activation of adenyl cyclase.

Effect of calcium dobesilate and its interaction with aspirin on thrombus formation in vivo.

The effect of calcium dobesilate (calcium dihydroxy-2,5 benzenesulphonate) on thrombus formation in vivo was evaluated. Experimental thrombi were induced in a venule of a hamster cheek pouch following iontophoresis of adenosine diphosphate (ADP). Calcium dobesilate administered intraperitoneally as a single injection inhibited thrombus formation in a time- and dose-dependent manner. Furthermore, the combination of calcium dobesilate and aspirin resulted in the potentiation of the inhibition of thrombus formation.
