Renal cell carcinoma having heterogeneous histological appearance and homogeneous enzymatic property.

A case of renal cell carcinoma (RCC) presenting various histologic growth patterns, including papillary, tubular and solid arrangements, and an area resembling sarcoma are described. In order to identify RCC of high versatility of renal carcinosarcoma, electrophoretic studies of tissue alkaline phosphatase (Al-P) and gamma-glutamyl transpeptidase (gamma-GTP), as well as histochemical studies of these enzymes, were conducted. The Al-P was proved to be bond type alone and the gamma-GTP corresponded with the novel isozyme associated with RCC tissue. Moreover, these two enzymes were found to be distributed in every part of different histologic appearance. It was concluded from these results that the histologic pleomorphism of the case herein reported was due to morphologic transformation of the neoplastic cells sharing essentially identical enzymatic nature.

Lactate dehydrogenase in human renal carcinoma tissues.

Lactate dehydrogenase (LDH) activity and isoenzyme were determined in 27 human renal carcinoma (RC) tissues. Although LDH-1 and LDH-2 were predominant in the normal kidney, a completely inverted isoenzyme pattern was observed in 16 carcinomas. The others, however, showed either a normal or an incompletely inverted pattern. The LDH activity in the tumours with a completely inverted pattern was significantly higher than the activity of incompletely inverted or normal kidney pattern. It was also found that the isoenzyme pattern was more closely correlated with cell type than the grade of malignancy of the tumour, 14 out of the 16 cases of the completely inverted pattern being of the clear cell type, and 9 out of the 11 cases of incompletely inverted or normal kidney pattern being of either granular cell or mixed cell type. These results seemed to well substantiate an opinion that in clear cells anaerobic glycolysis provides a major energy source, whereas granular cells seek energy source primarily in TCA cycle and subsequent oxidative phosphorylation.

Linkage disequilibrium in isolated populations of Drosophila melanogaster.

In order to test the interaction effect of overdominance and random genetic drift on the formation of linkage disequilibria under the condition of multiplicative gene action, linkage disequilibria between isozyme genes, inter se, and between polymorphic inversions and isozyme genes were tested for the second chromosomes of Drosophila melanogaster sampled from two isolated Pacific islands and one locality of the northern district of the mainlands of Japan. The effective sizes of these populations were known to be approximately 3,000 to 6,000 on the basis of the allelism rate of lethal chromosomes and their frequencies. The following results were obtained: (1) No linkage disequilibrium considered to be induced by epistasis, including the interaction between overdominance and random genetic drift, was detected. (2) Nonrandom association between polymorphic inversions and isozyme genes that are included in the inversions or located in the adjacent region outside the inversions was detected. (3) On the basis of the comparison of chi (2) values, indicating the magnitudes of linkage disequilibrium, between the present isolated populations and the Raleigh, N.C., population (Mukai et al. 1974, 1977), the characteristics of the isolated populations and the nature of these linkage disequilibria are discussed.

Non-hodgkin malignant lymphoma with special reference to desmosome-like junctions.

Seventy-nine cases of non-Hodgkin malignant lymphoma were light and electron microscopically evaluated with special reference to intercellular correlationship comparing with non-neoplastic lymph nodal cells and cultured B cells. Reticulum cells and histiocytes are regarded as an independent cell unit. Reticulum cell sarcoma shows a fibrosarcomatous proliferation with abundant reticulin fibers, while histiocytic sarcoma is confined to a pleomorphic neoplasm with valid phagocytization. Lymphoid cells are divided into large, intermediate and small lymphocytes. Under inflammation and cultivation they, especially intermediate and large, preferentially proliferated clustering in a reticular fashion, which are designated as reticular lymphocytes, in contrast with nonreticular lymphocytes individually separated. The desmosome-like junctions are found among reticular lymphocytes not only in but also outside germinal centers as well as in vessels and among cultured B cells. Lymphocytic sarcomas, either nodular or diffuse, are classified into reticular and nonreticular types in association with their cellular sizes. The desmosome-like junctions are predisposedly disclosed in reticular lymphoma irrespective of proliferating patterns, nodular or diffuse, and of membrane character, B or T. And the junctions are not regarded as a hallmark for follicular or nodular lymphomas of germinal center cell origin. ACTA PATHOL. JPN. 30: 195 approximately 217, 1980.

Watery diarrhea, hypokalemia and achlorhydria syndrome. Morphological and immunohistological study.

A case of WDHA syndrome accompanied by a pancreatic tumor in a 44-year-old Japanese male is presented, the 6th case in Japan. Clinically, the patient suffered from unremitting watery diarrhea, hypokalemia and achlorhydria with marked anemia and jaundice. The patient died of emaciation, dehydration and bronchopenumonia, and an autopsy was performed. Autopsy examination revealed a hen's egg-sized tumor in the tail of the pancreas with metastases in liver, lungs and lymph nodes. In addition, bronchopneumonia and diabetic nephrosclerosis were present. Histologically, the tumor had the characteristics of an islet cell tumor, and histochemically the tumor cells were positive to Grimelius' stain which revealed non-B-islet cell features. Electron-microscopically, the tumor cells had electron dense round membrane-bounded granules resembling non-B-granules of pancreatic islet cells. With the immunoperoxidase procedure (PAP method), tumor cells nearly almost reacted to anti-vasoactive intestinal polypeptide (VIP) serum, which suggested that the tumor of the present case had the capability to produce VIP.

The effect of antivitamin B6 administration on gamma-aminobutyric acid metabolism in retina and electroretinogram.

The effect of several antivitamin B6 on gamma-aminobutyric acid (GABA) metabolism was studied in the rat retina. The rat electroretinogram (ERG) was also recorded after administration of these drugs. Aminooxyacetic acid (AOAA) and hydrazine administration increased the GABA content and inhibited the GABA degrading enzyme, GABA transaminase in retina. In addition, there drugs elongated the peak latency of the oscillatory potential in the rat ERG. In contrast, 4-deoxypyridoxine (DOP) or isonicotinic acid hydrazide (INAH) administration decreased the GABA content and inhibited the GABA synthesizing enzyme, glutamic acid decarboxylase in retina, and administration of these drugs together with AOAA lessened the degrees of elevation of GABA content and of the elongation of the peak latency produced as compared with AOAA alone, though neither of the former drugs had a significant effect on ERG. The retinal GABA seems to play an important role in relation to the oscillatory potential of ERG.
