Evaluation of in-vitro-cultured Plasmodium falciparum as antigen for malaria serology.

A handicap in developing seroepidemiology for malaria in India is the lack of homologous antigens. Aotus monkeys, in which human plasmodia can be propagated, are not easily available in India. Other alternatives include the use of infected human blood or the in-vitro culture of Plasmodium falciparum, using the candle-jar technique of Trager & Jensen (1976). With certain modifications, the in-vitro culture technique has been established at the St John's Medical College (SJMC) to obtain a regular and high yield of P. falciparum to use as a source of antigen in serological tests. Antigens prepared from in-vitro-cultivated P. falciparum have been tested against positive and negative reference sera by the indirect haemagglutination (IHA) and immunofluorescent antibody (IFA) tests. These reference sera were also tested with the IHA test by using P. falciparum-sensitized human "O' erythrocytes received from the Center for Disease Control (CDC), Atlanta. IHA test results with both types of antigens were comparable (Spearman rank correlation coefficient, rS=0.8440, P less than 0.01). The antigen from in-vitro culture was also found suitable for use in the IFA test. The IFA titres obtained were comparable to those found at the CDC (Spearman rank correlation coefficient, rS=0.7940, P less than 0.01). These results indicate that P. falciparum parasites prepared from in-vitro continuous culture can be used as a source of antigen for use in the IHA and IFA tests for malaria.

Studies on the effects of glucagon on ouabain-induced cardiac disorders using the PISA method.

This paper presents the application of a newly developed noninvasive system (PISA; Phase-Invariant Signature Algorithm) in the early detection and quantification of the effects of glucagon on the ouaban-induced electrophysiological disturbances in the heart of the dog. Three doses (20, 30, and 40 migrograms/kg) of ouabain were administered intravenously to each of six anesthetized dogs and broadband ECGs were recorded on FM tape for 100 min. The records were analyzed for PISA signatures and indexes at 20, 40, 60, 80, and 100 min after ouabain administration. There were dose-dependent increases in the PISA indexes. Ouabain (20 micrograms/kg) did not produce any observable changes in the conventional ECG, although it produced significant increases in the PISA indexes. In a second group sox dogs, glucagon (50 micrograms/kg) was administered intravenously 20 min after ouabain (40 micrograms/kg) administration and the ECGs, both conventional and for PISA analysis, were recorded for a further period of 80 min. Glucagon reversed the effects of ouabain on the PISA indexes. These indicate that the PISA method has the capability of early detection and quantitication of drug-induced cardiac disorder.

XXY Klinefelter's syndrome with bilateral cryptochidism, obesity, multiple capillary hemangiomas and telangiectasia.

A case of XXY Klinefelter's syndrome associated with bilateral cryptochidism, -ultiple capillary hemangiomas and vesical telangiectasia is reported. Testicular biopsy revealed Leydig cell hyperplasia with hyalinization of the seminiferous tubular membrane. The patient presented with recurrent hematuria. Excretory urography, urethroscopy and renal angiography were normal. Cystoscopy revealed multiple telangiectasia, which was responsible for the hematuria. To our knowledge this is the first report of multiple capillary hemangiomas and vesical telangiectasia associated with calssical Klinefelter's syndrome.

A preliminary study in the detection of cardiac disorders via phase-invariant signature algorithm of ECG.

The present paper presents a simple and unambiguous computer-assisted method for the detection of cardiac disorders by processing the electrocardiogram. A new technique "Phase Invariant Signature Algorithm (PISA)", has been described, which is useful in the detection of cardiac abnormalities by investigating certain statistical properties of the measured waveform of ECG in a "phase-locked" fashion. This method of detection of abnormalities in the heart was used in the changes in the hypoxic action potential and in the ECG due to ionic changes and ischemia in the heart of cats and dogs. The signature of the normal ECG or action potential was straight-line. Any change in the waveform of ECG or action potential was detected as spikes in the signature. This method of detection of cardiac abnormalities does not require a priori reference to any standard ECG. The results indicate that this new method is capable of detecting cardiac disorders at an early stage and hence a higher sensitivity than the presently available type of ECG analysis. This method is in the developmental stage and further studies are being carried out.
