Serum glycolytic enzymes in breast carcinoma.

Serum levels of lactate dehydrogenase and its isoenzymes, phosphohexos isomerase, aldolase and hexokinase were determined in 152 cases of breast carcinoma, in search of proper diagnostic and prognostic parameters. The enzyme levels showed a relation to the clinical stage of the tumor and were, on an average, higher in advanced stages of disease. Phosphohexose isomerase and lactate dehydrogenase-isoenzyme levels were of best diagnostic significance even at an early stage of the disease. The enzyme levels correlated well with the prognosis of the disease. In group three, the prognosis was very poor and the patients responded only slightly to the treatment, and only up to the third month; after that enzyme levels again increased.

Efficient prediction of chemical carcinogenicity by microsomal degranulation.

Microsomes prepared at low 'g' force (10 000) have been used to evaluate the carcinogenicity of 25 coded and 14 noncoded compounds to validate the microsomal degranulation technique for its use as one of the tests in a battery of 5 to 6 tests. The method correctly predicted 35 compounds out of 39 randomly selected carcinogens and non-carcinogens showing a higher predictive efficiency in comparison to that achieved by using rough endoplasmic reticulum prepared at high 'g' forces (105 000 X g or above). The use of this technique is advocated on a large scale for the screening of environmental carcinogens.

PISA--a new noninvasive method for early detection and quantification of ischemia in hypertensive patients.

Hypertension if untreated, is known to produce cardiac damage. Cardiac complications result from the effects of hypertension on the myocardium and/or to accelerated atherosclerotic changes in the coronary arteries. Presently available diagnostic tools are able to detect cardiac complications only when they are relatively at an advanced stage. Early detection of cardiac complications would have the advantage of early treatment, with potential for reversal. PISA (Phase-Invariant Signature Algorithm) - a new noninvasive method has been shown by us to have high sensitivity and specificity for diagnosis of ischemic heart disease. This method was therefore used in 25 healthy individuals, 20 patients with documented ischemic heart disease and 50 hypertensive patients (duration 1-6 years) with no end-organ disease in order to determine if these patients have myocardial ischemia and/or infarction. These patients were on antihypertensive medication. Healthy individuals as expected, had phase-invariant PISA signature and low PISA index. All 20 patients with documented ischemic heart disease had phase variant signature and had high PISA index in two or more leads. The sensitivity of this method for detection of ischemic heart disease was found to be 100%. Twenty-nine of the 50 hypertensive patients were found to have phase-variant (abnormal) PISA signatures and their PISA index was beyond the upper limit of normal PISA range (mean + 3S.D. of healthy individual). This study suggests that these 29 hypertensive patients possibly have ischemic heart disease. This PISA study indicates that 8 patients have anterior wall, 10 inferior wall, and 11 both anterior and inferior wall ischemia and/or infarction. These twenty-nine patients have varying degree of ischemia and/or infarction.

Seroepidemiology of malaria in Karnataka State: longitudinal study of a population from an area with high incidence at Kolar, South India.

Serologic methods have considerable value in epidemiologic surveillance of malaria. Antigens prepared from P falciparum cultured in vitro have been found to be sensitive and to a certain extent specific in the IFA test to detect malarial antibodies in the sera from a population continuously exposed to P falciparum and Plasmodium vivax infections [1]. However, it is necessary to assess the serologic data against the results of conventional examination of peripheral blood smears in monitoring the effect of antimalaria measures on malaria transmission. With this objective, we studied a randomly selected population from GPHU, which is located in an area in the Kolar District of the state of Karnataka where malaria is known to be endemic. During the transmission period of 1980 (from July to November), the annual parasitic incidence was 202 per 1,000, with 78% due to P vivax and 22% to P falciparum. The antimalaria measures were subsequently intensified by the GPHU staff, which lead to a decrease in the incidence of malaria in 1981 (60 per 1,000, with 91% due to P vivax and 9% to P falciparum; data obtained from GPHU records). Serum samples were collected after each transmission period and tested by IFA using a local isolate of P falciparum as antigen. There was a marked decrease in the number of seropositive samples in the second batch among children less than 10 years of age and a threefold decrease in malarial antibody titers up to the age of 15 years.(ABSTRACT TRUNCATED AT 250 WORDS)

Evaluation of a crude soluble antigen from in-vitro culture of Plasmodium falciparum for ELISA.

The present study was to evaluate the soluble antigen prepared from the in-vitro cultured P. falciparum (FCR3) as an alternative source of antigen to those obtained from in-vivo models for enzyme linked immunosorbent assay (ELISA) in serology of malaria. Results obtained on known positive and negative reference sera revealed good correlation between the ELISA and the indirect fluorescent antibody (IFA) technique (rs = 0.797; p less than 0.001). However such close correlation was not observed on six local sera from patients whose peripheral blood smear showed ring stage of P. falciparum (rs = 0.43; p greater than 0.05), though all the six sera were positive by the IFA and ELISA tests. Test was repeated to establish its reproducibility. The results indicate that antigen prepared from in vitro culture and stored in liquid nitrogen was found sensitive in ELISA for serology of malaria.
