Lung cancer proliferation correlates with [F-18]fluorodeoxyglucose uptake by positron emission tomography.

Tumor proliferation has prognostic value in resected early-stage non-small cell lung cancer (NSCLC). We evaluated whether [F-18]fluorodeoxyglucose (FDG) uptake of NSCLC correlates with tumor proliferation and, thus, could noninvasively grade NSCLCs (refining patient prognosis and therapy). Thirty-nine patients with potentially resectable NSCLC underwent whole-body FDG positron emission tomography (PET) 45 min after i.v. injection of 10 mCi of FDG. Tumor FDG uptake was quantitated with the maximum pixel standardized uptake value (maxSUV). The lesion diameter from computed tomography was used to correct the maxSUV for partial volume effects using recovery coefficients determined for the General Electric Advance PET scanner. Thirty-eight patients underwent complete surgical staging (bronchoscopy and mediastinoscopy, with or without thoracotomy). One stage IV patient by PET underwent bronchoscopic biopsy only. Immunohistochemistry for Ki-67 (proliferation index marker) was performed on all of the 39 NSCLC specimens (35 resections, 1 percutaneous, and 3 surgical biopsies). The specimens were reviewed for cellular differentiation (poor, moderate, well) and tumor type. Lesions ranged from 0.7 to 6.1 cm. The correlation found between uncorrected maxSUV and lesion size (Rho, 0.56; P = 0.0006) disappeared when applying the recovery coefficients (Rho, -0.035; P = 0.83). Ki-67 expression (percentage of positive cells) correlated strongly with FDG uptake (corrected maxSUV: Rho, 0.73; P < 0.0001). The correlation was stronger for stage I lesions (11 stage IA, 15 stage IB): Rho, 0.79; P < 0.0001) and strongest in stage IB (Rho, 0.83; P = 0.0019). A significant association (P < 0.0001) between tumor differentiation and corrected SUV was noted. FDG PET may be used to noninvasively assess NSCLC proliferation in vivo, identifying rapidly growing NSCLCs with poor prognosis that could benefit from preoperative chemotherapy.

[The chromosome-specific PCR marker's screening and identification of barley 6H chromosome].

Two barley 6H chromosome specific RAPD markers were obtained by screening DNA of barley Hordeum vulgare (Betzes) and wheat-barley 6H addition line with 200 primers, then the RAPD markers were changed into specific PCR markers. Checking different plant materials by the PCR markers, it revealed that there was a specific band in those materials containing 6H chromosome such as Betzes, Igri, CS6H, and there was no specific band if the material did not contain 6H chromosome, such as Triticum aestivum, Secale cereale, Agropyron intermedium, Haynaldia villosa, Thinopyrum elongatum. Therefore, those PCR markers specific to chromosome 6H of barley are established. Southern hybridization indicated that the two cloned DNA fragments belong to barley genomic specific high-copy repeat sequence and low-copy sequence in wheat and barley genomes respectively.

Specific regulatory inhibition of transfected HepG(2) with HCV 5'NCR by antisense phosphorothioate oligodeoxynucleotides.

OBJECTIVE: To screen efficient and specific new drugs against hepatitis C virus (HCV) and find the best target sequence of antisense oligodeoxynucleotides (ASODNs) targeting at HCV gene. METHODS: Fifteen S-ASODNs targeting at the 5'NCR and start AUG of HCV RNA were designed and synthesized according to the predicted RNA secondary structure of the HCV 5'NCR and the adjacent AUG region. HepG(2) cells were co-transfected with pHCV-neo4 and S-ASODN. The inhibitory effects of S-ASODNs on gene expression controlled by HCV 5'NCR were determined by the assay of luciferase activity. RESULTS: Five S-ASODNs, i.e. HCV65, HCV279, HCV363, HCV349 and HCV352, showed sequence-specific and dose-dependent inhibitory activities with an inhibition rate of more than 80% at a concentration of 100 nmol/L. According to 50% inhibitory concentrations, the inhibitory activity of HCV363 was the best. On the other hand, ASODNs had only little non-specific effect by negative control trials. In addition, the results also indicated that there were some coordinate effects between ASODNs at different targets. CONCLUSION: Stem-loop 2a and 3d of HCV 5'NCR and the start AUG of polyprotein precursor are candidate targets of S-ASODNs.

[Purification and characterization of recombinant Aeromonas punctata prolyl endopeptidase].

The study of down-stream techniques of recombinant Aeromonas punctata prolyl endopeptidase (apPEP) was presented here. High cell-density fermentation of E. coli BL21/pKKH-PEP in NBS BioFlo 3000 5 L fermentor was achieved, the final cell density was 22.5 g (DCW)/L after 14 h cultivation, the yield of apPEP expressed in soluble protein was 3.0 g per litter broth. After sonication, the supernatant of free cell extract was purified by ammonium sulfate fractionation, High performance Q sepharose FF, Phenyl sepharose 6 FF, the purity of apPEP reached 96%, enzyme specific activity was 65.5 u/mg, apPEP yield reached 0.86 g/L broth. Total recovery of enzyme protein was 8.2%, actviity recovery was 24.4%. The molecular weight of apPEP was 76,464 +/- 30 Da measured by MS, N terminus amino acids sequence consistent with that deduced from DNA sequence. pI 6.0, which was similar with PEP from Aeromonas hydrophila.

The association of BMI and WHR on blood pressure levels and prevalence of hypertension in middle-aged and elderly people in rural China.

The purpose of this study was to examine the effects of BMI (Body Mass Index) and WHR (Waist Hip Ratio) on average blood pressure and the prevalence of hypertension in middle-aged and elderly population in rural China. A total of 12955 subjects including 6276 males and 6688 females over 40 years of age were surveyed. Height, weight, waist circumference, hip circumference and blood pressure were measured. The association of BMI and WHR on average blood pressure levels and prevalence of hypertension were analyzed by dividing BMI and WHR into tertiles. The average blood pressure levels and the prevalence of hypertension in males and females increased significantly with the increase of BMI or WHR (P < 0.01), as well as with the increase of both of them (P < 0.01). The average blood pressure and the prevalence of hypertension stopped increasing when WHR was > or = 0.76, suggesting that WHR > or = 0.80 could be used as a cut-off value for the prediction of hypertension risk for both males and females. Therefore hypertension could be effectively prevented and controlled by controlling BMI and WHR.

[A system of microcomputer analysis for biological oxygen consumption and its application].

This paper introduces a microcomputer system of data analysis and methods of measuring oxygen consumption for biological materials. The system overcame some disadvantages of operating inconvenience and difficulties of collective and analytic data by traditional means. The system possesses functions of automatic enactment, collection, save/take and analysis for many experimental data. It can be used to measure oxygen consumption of tissues, cells and mitochondria.

Potentiation of docetaxel antitumor activity by batimastat against mouse forestomach carcinoma.

Docetaxel is a chemical compound belonging to the taxoid class of anticancer agents. Batimastat (BB-94) is the first matrix metalloproteinase inhibitor entering clinical trials. To improve the treatment of tumors, we studied the combined effects of docetaxel and batimastat on mouse forestomach carcinoma (MFC), and compared them with doxorubicin. In vitro growth curve analysis, MTT assay, and clonogenic assay were used to determine the cytotoxic effect of docetaxel or/and BB-94 on MFC. They showed that docetaxel, but not BB-94, had a significant cytotoxicity and that the effect of docetaxel was not enhanced by BB-94. In an early stage MFC tumor model, an obvious antitumor effect of docetaxel or doxorubicin given iv at maximum tolerated dose (MTD) was observed. Tumor growth inhibition was greater for docetaxel + BB-94 (96.0%) than for doxorubicin + BB-94 (88.0%), docetaxel (89.0%), doxorubicin (68.0%), and BB-94 (33.0%). Docetaxel showed activity against advanced stage MFC tumor in a dose-dependent manner and was more effective at MTD than doxorubicin, with 4/5 regression, 46.5 days tumor growth delay, and 2.8 log10 tumor-cell kill. Our results suggest that docetaxel is an effective new cytotoxic drug against MFC tumor and that BB-94 enhances the antitumor activity of docetaxel in the dose and schedule used.

[E-cadherin expression and its clinical significance in cervical cancer].

OBJECTIVE: To study the clinical significance of E-cadherin (E-CD) expression in cervical cancer. METHODS: Specimens of 60 cervical cancer, 10 normal cervical epithelia and 12 cervical intraepithelial neoplasia(CIN) were examined for E-CD expression by immunohistochemical staining. RESULTS: E-CD was expressed in all of 10 specimens of normal cervical epithelia. Positive staining was seen in 66.7% of CIN, 46.7% of cervical cancer. In cervical cancer, the frequency of abnormal E-CD expression was correlated to pathologic grade, lymph node metastasis and clinical stage (P < 0.05). The higher the pathologic grade of tumor, the lower was the expression of E-CD. Negative E-CD expression was significantly more frequent in the advanced stage of cervical cancer with lymph node metastasis. CONCLUSION: Decreased expression of E-CD is a useful parameter of cervical malignancy.

[Study on parents phenotypes in women with polycystic ovary syndrome].

OBJECTIVE: To explore the parents phenotypes in women with polycystic ovary syndrome (PCOS). METHODS: Clinical data were collected by questionnaires from 139 parents of women with PCOS and from 137 parents of controls with normal menses. It was compared for the distribution of mother's menses irregularity, father's premature balding and parents hypertension by chi-square tests among parents of the PCOS group and of the controls. A multiple Logistic regression model was applied for more detailed analyses. RESULTS: The prevalences of mother's menses irregularity, the incidence of father's premature balding and father's hypertension was significantly higher in PCOS group (38.1%, 19.4% and 30.9%, respectively) than in the controls (3.6%, 5.1% and 16.1%, respectively P < 0.01). The prevalence of mother's hypertension was higher in the PCOS group (23.0%) than in the controls (13.9%), but the difference was not significant (P = 0.06). When mother's menses irregularity, father's premature balding and father's hypertension were entered the Logistic regression model, all the odds ratio (OR) values were still much higher than 1 (P < 0.005). The partial regression coefficients of the three predictors changed little when they were entered the model in turn. The partial regression coefficients for father's age was very small when it entered the model as a possible confounding factor(P > 0.05). The differences between maximum -2log likelihoods used to evaluate the role responsible for the occurrence of PCOS in their daughters indicated that: mother's menses irregularity > father's premature balding > father's hypertension. CONCLUSION: In addition to mother's menses irregularity and father's premature balding, father's hypertension may also be an independent phenotype in families of women with PCOS.

[Congression and regression tree analyses on the outcome of ovulation induction in women with polycystic ovary syndrome].

OBJECTIVE: To investigate the risk factors for failure of ovulation induction in polycystic ovary syndrome (PCOS) patients and establish congression and regression tree (CART) model. METHODS: Sex hormones were measured in 103 PCOS patients and 31 healthy women by radioimmunoassay, oral glucose tolerance test (OGTT) and insulin release test were done as well. Three ovulation induction protocols including clomiphene (CC)/chorionic gonadotrophin (hCG), CC/menotrophins (hMG)/hCG and gonadotrophin releasing hormone agonist(GnRH-a)/hMG/hCG were used for PCOS related infertility. Transvaginal ultrasonography was used for monitoring. The correlation between hormone levels and clinical outcomes was analyzed by CART method. RESULTS: The ovulation rate in patients with 2-hour blood insulin level < or = 68.9 mIU/L during OGTT was 86.9%, which was significantly higher than those with > 68.9 mIU/L (61.9%, P < 0.05). In the model, the insulin and glucose level of 0 min, 60 min during OGTT acted obviously, but estradiol and basal T level did not, the protocol of ovulation induction was not able to enter the model. CONCLUSION: Two hour insulin level during OGTT is the most important factor affecting the outcome of ovulation induction, while the others parameters of glucose and insulin level have useful action too.

[Comparison on effects between concentrated-dose and non-concentrated-dose pralidoxime chloride on respiratory muscle paralysis in acute organophosphorus pesticide poisoning].

OBJECTIVE: To compare the effects between concentrated-dose and non-concentrated-dose pralidoxime chloride on respiratory muscle paralysis(RMP) in acute organophosphorus pesticide poisoning(AOPP). METHODS: 76 cases of RMP due to AOPP were involved in a retrospective study. All the patients were poisoned through ingestion. According to the administered dose of pralidoxime chloride within the first three days after the onset, the patients were divided into two groups: Group A consisted 30 patients; they were given pralidoxime chloride 10.0-25.8 grams (with an average of 11.4 grams) everyday in the first three days after the onset. Group B included 46 patients; the dose of pralidoxime chloride used daily in the first three days ranged from 3.3 to 8.6 grams (average 6.3). RESULTS: The curative rate and the death rate in group A were 73.3% and 26.7% respectively, while those in group B were 21.7% and 78.3% respectively. The curative rate in group A was significantly higher than that in group B (chi 2 = 19.83, P < 0.005). CONCLUSION: Concentrated-dose pralidoxime chloride was more effective than non-concentrated-dose for patients with RMP due to AOPP and this kind of administration could remarkably improve the prognosis in such patients.

[Dynamics of community structure in successional process of needle and broad-leaved mixed forest in Heishiding of Guangdong].

This paper dealt with the dynamics of community structure in a ten-year succession of needle and broad-leaved mixed forest in Heishiding of Guangdong Province. The vertical community structure tended to be complex, while the horizontal one increased by a wide range. There was no big change in species composition, but the dominance of conifers Pinus massoniana and Cunninghamia lanceolata was substituted by the broad-leaved species, such as Schima superba, Symplocos adenophylla and Schefflera octophylla. The whole community changed from needle and boad-leaved mixed forest into broad-leaved and needle mixed forest. The species diversity and evenness of tree layer were increased, the ecological dominance was decreased, and the community became more complicated.

[Molecular ecology of Cryptocarya chinensis populations in different communities].

The amplified fragment length polymorphism (AFLP) method was used to analyze two populations of Cryptocarya chinensis, the succession climax species in lower subtropical broadleaved forest. The across two communities of evergreen broadleaved forest and mixed coniferous-broadleaved forest. The analysis of molecular variance (AMOVA) indicated that the genetic variation was mostly due to the difference within individuals of the population (89.55%), with high significant genetic variation (10.45%) due to the different communities. The microenvironment of different communities and population biological characteristics resulted in the genetic divergence.

[Molecular markers and their application in plant population research].

This review surveys kinds of markers such as allozyme, AFLP, RAPD, SSR, RFLP and their application in molecular ecology of plant populations. The advantages and disadvantages of these molecular markers in plant population research are discussed based on the comparison of the methods and the experience in research work. It is considered emphatically that the adoption of the molecular markers is dependent on the various research purposes and different quality and quantity of the molecular information are acquired from various molecular marker methods.

[Effects of ovulation induction therapies on endometrial integrin alpha v beta 3 expression in patients with polycystic ovarian syndrome related infertility].

OBJECTIVE: To study the endometrial integrins alpha v beta 3 expressions during mid-luteal(ML) phase in polycystic ovarian syndrome (PCOS) related infertile patients receiving ovulation induction therapies. METHODS: Immunohistochemical methods using monoclonal antibody against integrins alpha v beta 3 were performed on endometrium of ML phase obtained from 22 normal subjects and 40 PCOS related anovulatory patients who underwent ovulation induction therapy by one of the following regimens: clomiphene citrate (CC) + human chorionicgonadotropin (hCG), CC + human menopausal gonadotropin (hMG) + hCG, or gonadotropin-releasing hormone agonist + hMG + hCG. RESULTS: The expressions of endometrial integrin alpha v beta 3 during ML phase in GnRHa + hMG + hMG + hCG cycles were similar with those in normal natural cycles, but were weaker in CC + hCG, CC + hMG + hCG cycles as compared with controls. CONCLUSION: CC + hCG, CC + hMG + hCG regimens may induce decline of endometrial receptivity in ML phase.

[Relationship between serum epidermal growth factor and uterine leiomyoma].

OBJECTIVE: To study the effects of epidermal growth factor (EGF) on uterine leiomyoma and the relationship of EGF and estrogen, progesterone. METHODS: Fourty patients with uterine leiomyoma and twenty normal women were studied. Serum EGF level was determined by radioimumunoassay, estradiol (E2) and progesterone (P) levels were measured by enzyme marked monoclonal antibody assay. RESULTS: Serum EGF, E2 and P levels in observed group were significantly higher than those in control group during secretive phase (P < 0.005). The serum EGF level in observed group during prolifrative phase was significantly higher than that of secretive phase (P < 0.05). The serum EGF level increased while uterus enlarged; EGF wasn't related to E2(r = 0.25, P > 0.05) and was related to P (r = 0.71, P < 0.005). The EGF level was the highest when E2 and P both increased. CONCLUSION: EGF has the effect on growth of uterine leiomyoma. It is possible that EGF secretion can be stimulated by E2 and the effect of P is stronger than E2.

[Expression of bcl-2 protein in adenomyosis].

OBJECTIVE: To investigate the expression of B cell lymphoma/leukemia-2 (Bcl-2) protein in adenomyosis and its clinical implication. METHODS: Endometrium specimens were obtained from 16 normal uterus and 32 with adenomyosis. bcl-2 expression in these paraffin embraced specimens was examined by immunohistochemical technique with specific monoclonal antibodies. RESULTS: bcl-2 expression in normal endometrium showed a significant cyclic variation. bcl-2 expression of glandular cells in both eutopic and ectopic endometrium of adenomyosis also showed a significant cyclic variation. bcl-2 expression in endometrium of adenomyosis was stronger than that in normal endometrium, and bcl-2 expression in ectopic endometrium was stronger than that in eutopic endometrium. CONCLUSION: The decrease of cellular apoptosis resulted from the high bcl-2 expression may play an important role in the pathogenesis of adenomyosis.
