Angiotensin II binding sites in the anteroventral-third ventricle (AV3V) area and related structures of the rat brain.

Angiotensin II binding sites were localized in the rat brain by incubation of sagittal sections with 125I-[Sar1]-angiotensin II, followed by [3H]Ultrofilm autoradiography. Binding sites were localized in the subfornical organ and the entire anteroventral-third ventricle (AV3V) area, including the subependymal area of the anterior third ventricle, the median preoptic nucleus and the organon vasculosum laminae terminalis. Binding sites were also present in the paraventricular and periventricular nuclei and in the median eminence. These findings support the hypothesis of an extensive involvement of angiotensin in the neuronal circuits connecting the subfornical organ, the AV3V area, the paraventricular nucleus and the median eminence, and in the regulation of fluid balance, blood pressure and pituitary secretion.

Localization of atrial natriuretic peptide, ANP-(99-126) binding sites in the rat thymus and spleen with quantitative autoradiography.

Binding sites for atrial natriuretic peptide, ANP-(99-126) were studied in lymphoid organs of the rat with quantitative autoradiography. Tissue sections were incubated in the presence of 0.13 nM 125I-ANP-(99-126) followed by autoradiography using [3H]-Ultrofilm, and the results were analyzed by computerized densitometry and comparison to 125I-standards. Specific ANP binding sites were localized in the medulla and the cortex of the rat thymus and in the white pulp of the rat spleen, with apparent binding sites concentrations of 93, 65, and 126 fmol/mg protein, respectively. The presence of ANP binding sites in areas related to the maturation and function of lymphocytes, and to the production of thymic hormones, suggests the possibility of a role of circulating ANP in the modulation of the immune response.

Decreased number and affinity of rat atrial natriuretic peptide (6-33) binding sites in the subfornical organ of spontaneously hypertensive rats.

Binding sites for rat atrial natriuretic peptide (6-33) were quantified by incubation of brain sections with (3-[125I]iodotyrosyl28) rat atrial natriuretic peptide (6-33), followed by autoradiography with computerized microdensitometry. Spontaneously hypertensive rats present lower numbers and lower affinity of binding sites than normotensive controls, Wistar-Kyoto rats, in the subfornical organ (binding capacity 61.7 +/- 8.9 and 124.3 +/- 10.7 fmol/mg protein; affinity constant 4.25 +/- 0.55 and 11.10 +/- 1.67 X 10(9) M-1, respectively). In the choroid plexus, hypertensive rats have lower numbers of sites than normotensive rats (binding capacity 72.7 +/- 10.5 and 173.6 +/- 22.8 fmol/mg protein, respectively), but there was no difference in the binding affinity (affinity constant 6.28 +/- 0.82 and 7.60 +/- 2.06 X 10(9) M-1, respectively). Our results suggest that discretely localized brain binding sites for rat atrial natriuretic peptide (6-33) may have a physiological function in genetically hypertensive rats.

Increased atrial natriuretic peptide (6-33) binding sites in the subfornical organ of water deprived and Brattleboro rats.

Binding sites for rat atrial natriuretic peptide (6-33) (ANP) were quantitated in the subfornical organ of chronically dehydrated homozygous Brattleboro rats unable to synthesize vasopressin; heterozygous Brattleboro rats, their controls, Long Evans rats and Long Evans rats after 4 days of water deprivation. Brain sections were incubated in the presence of 125I-ANP and the results analyzed by autoradiography coupled to computerized microdensitometry and comparison to 125I-standards. Brattleboro rats and water deprived Long Evans rats presented a higher number of ANP binding sites than their normally hydrated controls. Our results suggest a role of ANP binding sites in the subfornical organ in the central regulation of fluid balance and vasopressin secretion.

[Cancer chemotherapy of unresectable primary gastric cancer].

The criteria for cancer chemotherapy of gastric cancers proposed by the Japanese Research Society for Gastric Cancer is introduced with several effective cases. Three subtypes of judgment method--namely, measurable lesions, non-measurable but evaluable by X-ray or endoscopy, and diffuse infiltrating lesions are applied according to the shape and effect of primary foci. Measurable lesions in a case such as aPR should be recorded in metric notation, using a ruler or calipers on X-ray films. bPR is a case when chemotherapy of primary foci is markedly effective with a flattening of the randwall or polypoid part and a shrinkage of cancerous ulceration, but the size of a primary lesion is not decreased. cPR is a case when an expansion of affected site by more than 100% is proved by gastric radiogram or endoscopic image on primary focus in Borrman type 4, a synonym of gastric scirrhous clinically.

[Combination chemotherapy with cis-platinum, adriamycin and mitomycin C (PAM) in the treatment of non-small cell carcinoma of the lung].

Twenty-three patients with inoperable non-small cell lung cancer were treated with a combination chemotherapy of CDDP 100 mg/m2, ADM 30 mg/m2 and MMC 8 mg/m2 (PAM). Ten cases were adenocarcinoma, 9 cases were squamous cell carcinoma and 4 cases were large cell carcinoma. In 21 evaluable cases, partial response was obtained in 47.6%. (The response rates were 40.0% in patients with adenocarcinoma, 50.0% in those with squamous cell carcinoma and 66.7% in those with large cell carcinoma.) Leukocytopenia of less than 4,000/mm3 occurred in 100% of cases, thrombocytopenia of less than 100,000/mm3 occurred in 81.0%, and anemia(fall in hemoglobin over 2.0 g/dl) occurred in 66.7%. A transient elevation of Cr (over 1.5 mg/dl) and/or BUN (over 30 mg/dl) was observed in 23.8%. Nausea and vomiting occurred in almost all patients. No death occurred due to toxicity of PAM. These results demonstrate that PAM is an effective combination chemotherapy in patients with non-small cell carcinoma of the lung.

[Present diagnostic status of scirrhous type gastric cancer--with special reference to the endoscopic diagnosis].

First, we presented an actual diagnostic situation in nowadays for gastric cancer of Borrmann 4, which is virtually the same as scirrhous gastric cancer. Among 12 patients treated by the author, all of whom were discovered late, only 3 underwent surgery. In fact, with inoperable cases in Borrmann 4, even those endoscopically found to show insufficient stretching of the gastric wall, thickening and tortuosity of folds, uneven gastric mucosa, redness and white coating, there may be negative in gastric biopsy. However, the significance of an endoscopic examination for diagnosis of scirrhous cancer is in obtaining proof of the carcinoma (especially when still operable) by gastric biopsy. Thus, one must strive not to overlook slight redness, white coating which means small erosions, but to go over gastric biopsy again and again. Next, with carcinoma presenting a leather bottle (linitis plastica type) of the stomach itself, the II c portion of the stomach consisted of fundic glands (undifferentiated carcinoma) shall become the primary focus supporting Nakamura's theory. One case of diffuse invasive cancer, mistakenly diagnosed as a II c case, and two cases of regional type, one similar to II c and the other a Borrmann 2 carcinoma of advanced carcinoma showing strongly fibrous scirrhous tendency toward infiltration, were jointly monitored.

[Combined therapy of polyamine antimetabolite and nitrosourea in human gastric cancer].

Antitumor therapy using the polyamine antimetabolites, alpha-difluoromethylornithine (DFMO) and methylglyoxal-bis-guanylhydrazone (MGBG), combined with ACNU was studied in human gastric cancer xenotransplanted into nude mice. DFMO 1,000 mg/kg (in two divided doses) and MGBG 50 mg/kg were given i.p. for 6 successive days from the time when the xenotransplanted tumor weighed about 100 mg, and ACNU 20 mg/kg was given i.p. every other day from the same time. Antitumor efficacy was assessed by the time course of tumor weight as well as of DNA biosynthesis and polyamine levels in tumor tissue. Tumor weight was estimated using Battelle's Columbus Institute protocol and DNA biosynthesis was assayed biochemically by 3H-TdR injection at a prescribed interval after termination of therapy. Furthermore, tumoral polyamine levels were assayed by HPLC. This three-drug regimen showed a favorable antitumor effect, compared to those of the other two therapies with DFMO plus MGBG as well as ACNU only. These data suggest that this combined regimen may have a synergistic efficacy judging from the action mechanisms of these three drugs.

Increased number of angiotensin II receptors in the subfornical organ of spontaneously hypertensive rats.

Angiotensin II receptors were studied in the brain of spontaneously hypertensive rats (SHR) and their normotensive controls, Wistar-Kyoto rats (WKY) by quantitative autoradiographic methods coupled to computerized microdensitometry and comparison with 125I-standards, after incubation of brain sections with 125I-Sar1-angiotensin II. Binding sites for angiotensin II were highly localized in the subfornical organ. The number of binding sites was higher in the subfornical organ of young (4-week-old) and adult (14-week-old) SHR when compared with WKY rats (young rats: 735 +/- 91 and 444 +/- 85 fmol/mg protein, SHR and WKY, respectively; adult rats: 1390 +/- 64 and 848 +/- 136 fmol/mg protein, SHR and WKY, respectively). Our results suggest that subfornical organ angiotensin II receptors are involved in genetic hypertension in the rat. An increased number of subfornical organ angiotensin II receptors substantiates the idea of an overactive central angiotensin II system in spontaneous hypertension.

[A case of remarkable regression of lung adenocarcinoma with PSK immunotherapy].

An asymptomatic 84-year-old man was admitted to our hospital for a further examination of an abnormal shadow in the right lower lung (S9) on chest X-ray film. Sputum and specimens of transbronchial brushing showed cytological characteristics of well differentiated adenocarcinoma. Considering his age, his family did not agree to any surgical operation, irradiation or intensive chemotherapy. Therefore, using PSK: 3.0 g/day, immunotherapy was started on October 26, 1984. However, the abnormal density continued to increase in size and was observed radiographically to infiltrate to the neighboring segment (S6) over the following four months. From March 1985, the tumor regressed gradually and had almost disappeared on chest X-ray films by December 1985. Since then, this remission has been maintained by administration of PSK alone (3.0 g/day).
