Induction of a high spin form of microsomal cytochrome P-448 in rat liver by 4-aminoazobenzene derivatives.

Male Sprague-Dawley rats were treated with 4-aminoazobenzene derivatives or other drug metabolizing enzyme inducers such as phenobarbital, 3-methylcholanthrene and isosafrole. The expression of hepatic microsomal cytochrome P-450 of the rats, principally that of a high spin form of cytochrome P-448 (cytochrome P-448H), was assessed by a bacterial mutation test and by immunological methods. The results of the mutation test with use of Salmonella typhimurium TA9 and 3 aromatic amine substrates showed that 2-methoxyl, 3-methoxyl and 2',3-dimethoxyl derivatives of AAB and methyl derivatives of AAB such as o-aminoazotoluene, N-methyl-4-aminoazobenzene and N,N-dimethyl-4-aminoazobenzene have a large capacity for the selective induction of cytochrome P-448H. Activity of the cytochrome increased by 6 hr after an azo dye treatment, reached a maximum after 24 hr, and then declined. In contrast, 4'-methoxy-AAB has a small, and AAB has no, capacity for the cytochrome induction. The aminoazo dye-induced enzymes differ in their substrate specificities from those induced with 3-methylcholanthrene or phenobarbital, and the induced enzyme was identified to be cytochrome P-448H, as determined by an enzyme-linked immunosorbent assay and immunoblotting with use of anti-cytochrome P-448 monoclonal antibodies. These observations indicate that several methoxyl and methyl derivatives of 4-aminoazobenzene are potent and selective inducers of cytochrome P-448H in the rat.

Metabolic pathway of 2-deoxy-2-fluoro-D-glucose studied by F-19 NMR.

The behavior of 2-deoxy-2-fluoro-D-glucose (FDG) in mouse has been studied by F-19 NMR method for long period. The F-19 NMR signals of FDG or its metabolites were observed in tissues without serious broadening. FDG was found to be accumulated in organs in the form of FDG or FDG-6-phosphate and 2-deoxy-2-fluoro-D-mannose (FDM) or FDM-6-phosphate, and the latter dominated the former in the heart sampled at 24 hr or later. The fluorine compounds were excreted in urine in both forms. The clearance was rapid from brain, liver, and blood, but was slow from heart.

Purification and characterization of hydroxycinnamoyl D-glucose. Quinate hydroxycinnamoyl transferase in the root of sweet potato, Ipomoea batatas Lam.

We have previously proposed a chlorogenic acid biosynthetic pathway which involves a transesterification reaction between hydroxycinnamoyl D-glucose and D-quinic acid. The proposed pathway was based on tracer experimental results (Kojima, M., and Uritani, I. (1972) Plant Cell Physiol. 13, 311-319). The enzyme that catalyzes the above reaction has been purified 160-fold from sweet potato root (Ipomoea batatas Lam.) and characterized. The purified enzyme yielded one band of 26,000 daltons on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and its molecular weight was estimated to be 25,000 by gel filtration chromatography. Therefore, the enzyme seems to consist of a single polypeptide of 25,000-26,000 daltons. The isoelectric point of the enzyme was 8.6. The optimum pH of the enzyme reaction was 6.0. The enzyme did not require any metal for activity and showed a broad substrate specificity toward hydroxycinnamoyl D-glucose as donors. The Km and Vmax values were 3.7 mM and 8.5 units/mg of protein for t-cinnamoyl D-glucose, 3.9 mM and 15.1 units/mg of protein for p-coumaroyl D-glucose, and 14.3 mM and 38.1 units/mg of protein for caffeoyl D-glucose. The enzyme showed a strict substrate specificity toward D-quinic acid-related compounds as acceptors; the Km and Vmax values were 16.7 mM and 15.1 units/mg of protein for D-quinic acid, 250 mM and 19.0 units/mg of protein for shikimic acid, and there was no activity with either L-malic acid or meso-tartaric acid. The enzyme activity changed in a manner suggesting its involvement in chlorogenic acid biosynthesis during incubation of sliced sweet potato root tissues.

Peptide C-terminal alpha-amidating enzyme purified to homogeneity from Xenopus laevis skin.

The C-terminal alpha-amide formation of the peptides is one of the most important events of prohormone processing. In this study, we have developed a simple and sensitive assay for monitoring alpha-amidating activity by using radioiodinated Ac-Tyr-Phe-Gly as a substrate. By utilizing this assay, an alpha-amidating enzyme was first purified to homogeneity from Xenopus laevis skin. The purified enzyme has a single polypeptide chain with an apparent molecular weight of 39,000 and its N-terminal sequence was determined as Ser-Leu-Ser-. The enzyme converts several synthetic peptides with C-terminal glycine to the corresponding des-glycine peptide alpha-amides. The enzyme activity, with an optimal pH 6-7, was dependent on the copper ion and ascorbate. In the presence of 0.25 mM ascorbate, the enzyme exhibited a Km of 0.35 microM and a Vmax of 1.9 nmol/microgram/h for Ac-Tyr-Phe-Gly.

Effects of calcium channel blockers on ouabain-induced oscillatory afterpotentials in organ-cultured young embryonic chick hearts.

Ouabain induces oscillatory afterpotentials (OAPs) in organ-cultured young (3 day old) embryonic chick hearts. Since increased [Ca]i resulting from an inhibition of the Na pump by ouabain triggers oscillatory movements of Ca2+ (i.e. OAPs) intracellularly, Ca2+ influx through the cell membrane, which tends to increase [Ca]i, may be important in developing the OAPs. Therefore, in the present experiments, effects of calcium channel blockers on ouabain-induced OAPs in organ-cultured 3 day old embryonic chick hearts were examined. Automaticity was suppressed by elevating [K]o to 6 mM. To induce the OAPs, the preparations were stimulated (0.5 Hz) in the presence of ouabain (2.5-6.3 microM). The calcium channel blockers (10 microM) depressed the OAPs in the following order of potency: bepridil greater than verapamil greater than nifedipine greater than diltiazem. This order of potency of the calcium channel blockers in depressing the OAPs was the same as that for drug penetration into the cells, but different from that for depressing slow action potentials: nifedipine greater than diltiazem greater than verapamil greater than bepridil (our previous findings). These results suggest that an intracellular site of action of the calcium channel blockers is important for depression of the OAPs, and suppression of the slow inward Ca2+ current cannot be the sole mechanism for suppression of the OAPs by these drugs.

Solar urticaria. The relationship of photoallergen and action spectrum.

We studied six cases of solar urticaria immunologically and photobiologically. Serum in five patients and plasma in one patient yielded an immediate reaction when injected into the patients' own skin after light irradiation. These photoallergens were gel filtrated, and the molecular weights were determined. The result was compared with the action spectrum obtained by a monochrometer. In three patients, with the action spectrum in short visible light ranges (400 to 500 nm), the molecular weight of the photoallergen was 25,000 to 45,000 daltons. In a patient with an action spectrum from ultraviolet-A to short visible light ranges (330 to 520 nm), there was another photoallergen molecular weight that was 300,000 to 1,000,000 daltons. In a patient with a wide action spectrum from ultraviolet-B to long visible light ranges (290 to 630 nm), photoallergens, which were believed to be multiple, were generated only from plasma.

Effects of tetraethylammonium on the action potential duration as modified by catecholamine-releasing action in guinea-pig papillary muscles.

The mechanism whereby tetraethylammonium ion (TEA) prolongs the action potential (AP) was examined by standard microelectrode techniques in papillary muscles isolated from nonreserpinized and reserpinized guinea-pig hearts. First, effects of 5 and 10 mmol/l TEA on steady-state AP parameters, i.e. the maximum rate of rise of AP (Vmax), AP amplitude, AP duration and resting potential, were examined at stimulation rates of 0.1 to 5 Hz. Second, effects of 5 and 10 mmol/l TEA on nonsteady-state AP parameters, i.e. the recovery processes of Vmax, AP duration and overshoot, were examined by introducing premature responses with various diastolic intervals between the basic stimulation rate of 0.1 Hz. TEA prolonged the AP more in the nonreserpinized preparations than in the reserpinized ones at all stimulation rates. In both preparations, this prolongation of the AP was greater at lower rates (0.1-1 Hz) than at higher rates (2-5 Hz), but the ratio of TEA-treated AP duration to control one was almost the same at all stimulation rates. TEA slowed the recovery process of AP duration in the nonreserpinized preparations, but not in the reserpinized ones. TEA had no effects on the recovery processes of Vmax and overshoot in both preparations. These results suggest that: TEA modifies its intrinsic prolonging action of the AP by releasing norepinephrine from sympathetic nerve terminals; TEA prolongs the AP by reducing the time-independent outward current rather than the time-dependent outward current; and a TEA-sensitive current does not effectively contribute to the total ionic current at the time of Vmax.

Amino acid sequence of the variable region of heavy chain in immunoglobulin (Mot) having unusual papain cleavage sites.

An IgGl(lambda) Mot myeloma protein showed a unique susceptibility toward papain digestion. The Fab fragment of Mot was more digestible with papain than the Fc fragment. This phenomenon was found to occur by unusual cleavage of the Fd fragment with papain. Determination of the complete primary structure of the V region of the H chain of Mot identified two papain cleavage sites in the second complementarity-determining region (CDR). Amino acid sequence of the cleavage sites was Ser(55)-Asp-Asp-Argdecrease-Thr-Thr-Tyr-Gly-Pro-Argdecrease- Ser-Gln- (decrease = cleavage site). In the vicinity of these cleavage sites, many hydrophilic and polar residues are present and the predicted secondary structure near these cleavage sites suggested that this region was exposed on the surface of the molecule, and that the unusual papain cleavage of the IgG Mot might be caused by a unique conformation of the molecule, making it highly susceptible to enzyme digestion.

Frequency-dependent effects of phenytoin on the maximum upstroke velocity of action potentials in guinea-pig papillary muscles.

Phenytoin, at 50 to 200 micrograms reduced the maximum upstroke velocity of action potentials (Vmax) with increases in frequency from 0.25 to 5 Hz and in the external potassium concentration [( K+]0) from 2.7 to 8.1 mM. The drug-induced shortening of action potential duration was evident at 0.25 to 2 Hz but little at 3 to 5 Hz. Time courses of recovery of Vmax was studied by applying premature responses between the conditioning responses at 1 Hz both in control and in drug-treated preparations. Concerning the time courses of the difference between the Vmax values before and after drug treatments at the same diastolic interval, with increases in drug concentrations the intercepts at APD90 were increased but the time constants were not changed or slightly decreased in 8.1 to 5.4 mM [K+]0, whereas they were increased in 2.7 mM [K+]0. To understand the kinetic behavior of this drug on sodium channels, rate constants for the interaction of phenytoin with three states of channels in terms of Hondeghem-Katzung model were estimated from the above experiments of Vmax. The model most consistent with the present experiments was that with an affinity for inactivated channels 20 times greater than that for resting channels and with a minor affinity for open channels. Phenytoin produced a delay in the time course of recovery of overshoot and action potential duration at 0 mV (APD0), suggesting an additional inhibition of the slow channel by this drug.

A case report of mantle zone lymphoma progressing into follicular lymphoma.

The authors report a case of mantle zone lymphoma (MZL) progressing into follicular lymphoma (FL) with macroglobulinemia (IgM, lambda). At a first lymph node biopsy, a diagnosis of MZL was made on the basis of the presence of vague neoplastic nodules, consisting of small lymphoid cells and small cleaved cells. At a second lymph node biopsy, nodules of follicular lymphoma were observed in the central zones of the vague neoplastic nodules. At a third biopsy, the central zones were found to expand, revealing FL dominant pattern. The progress of FL from MZL also was suggested by sIg, AIPase, anti-CALLA (common acute lymphoblastic leukemia antigen) antibody, and anti-DRC (dendritic reticulum cell) antibody findings. Consequently, this is suggested to be a neoplasm of B-lymphocytes differentiating from small lymphocytes in the primary follicle or the mantle zone of secondary follicle into germinal center cells. The anti-DRC antibody findings suggest the neoplastic cells of FL induce DRC as the neoplastic stroma.

[Pharmacological studies of MO-8282, a new antidepressant].

We investigated the pharmacological characteristics of MO-8282 as a novel antidepressant. MO-8282 inhibited the specific binding of 3H-clonidine to cerebro-cortical membrane fractions from rats about five times more potently than mianserin, and it competed with clonidine in the twitch response of the isolated guinea-pig ileum under field stimulation. The results indicated that MO-8282 possessed alpha 2-adrenergic receptor blocking activity. MO-8282 in a dose of 30 mg/kg (p.o.) showed no inhibition against the uptake of noradrenaline, dopamine and serotonin in the rat brain, whereas mianserin inhibited the uptake of serotonin specifically. MO-8282, similar to mianserin, had no effect on spontaneous release of 3H-noradrenaline and slightly stimulated the release of 3H-serotonin from the rat cerebrocortical synaptosome. The turnover rate of noradrenaline in rat brain was accelerated by administration of MO-8282 (30 mg/kg) for 15 days; however, that of dopamine and serotonin was not affected. The above findings indicate that MO-8282, unlike tricyclic antidepressants, mainly exerts alpha 2-adrenoceptor blocking action on the central noradrenergic system, similar to mianserin. In addition, the fact that MO-8282 unlike mianserin showed no inhibition against uptake of serotonin in brain suggests that the alpha 2-adrenoceptor blocking of MO-8282 is more specific and potent than that of mianserin.

Experimental study on the regulation of testicular function by the cremaster reflex in rats.

The present study was performed to clarify the role of the cremaster reflex in regulating the testicular function in rats. The impairment of cremaster reflex by bilateral dissection of the genitofemoral nerves resulted in a decrease of testicular weight and degenerative changes of the semeniferous tubules in the rats kept in a cold room (4 degrees C), but not in the rats kept at room temperature (22 degrees C). The results indicate that the cremaster reflex contributes much to the temperature regulation of the testicle located within the scrotum.

In vitro evaluation of SF-2103A, a novel carbapenem antibiotic, as a beta-lactamase inhibitor.

SF-2103A, a new carbapenem antibiotic, exhibited a broad antibacterial spectrum and a potent inhibitory activity against a wide range of beta-lactamases, in particular, against cephalosporinases, with lower I50 values than those displayed by sulbactam and clavulanic acid. Using a fixed combination and checkerboard titration, in vitro synergy against the majority of the beta-lactamase-producing strains tested was demonstrated between SF-2103A and various beta-lactam antibiotics, especially cefotaxime, ceftizoxime, and cefoperazone. The synergistic effect of SF-2103A was more pronounced than that of sulbactam. The in vitro synergy was also confirmed by bactericidal and bacteriolytic activities and morphological effects.
