The excretion of urinary steroids were investigated in rats in the course of mammary carcinogenesis by 7, 12-dimethylbenz [a] anthracene (DMBA). The carcinogen was fed to female Sprague-Dawley rats on days 50 and 64 after birth at a dose of 10 mg each. The excretion of urinary androgen, 11-deoxy-17-oxosteroid (11-DOOS), increased temporarily (at 80 days of age) in DMBA-treated rats as compared with non-treated controls. The excretion of adrenal corticosteroid, 17-deoxy-corticosteroid (17-DOCS), was lower than normal in the rats that failed to develop tumors within 200 days after receiving DMBA. The difference between the above non-responders and the tumor-developing responders (or the non-treated controls) was significant at 80 and 120 days of age, but not at 160 days of age. The final tumor weights at 160 days of age were also positively correlated to the excretion of 17-DOCS at 130 days of age. It was indicated that the overall process of DMBA carcinogenesis could be promoted by the endogenous adrenocortical steroids. The possible mechanism underlying the production of mammary cancer is discussed in the light of the metabolic intimacy between DMBA and the adrenal steroids.
Epidemiologic and hormonal aspects of cervical cancer were compared with those of breast cancer in order to explore the genesis of female cancers. When accessed by the urinary steroid analysis, cervical cancer was associated with such steroidal deviations as to reflect an arrest of adrenal maturation. Breast cancer was associated with hypercorticoidism and ovulation failure. Evidence was presented to indicate that the geneses of these 2 female cancers were each related to 2 specific disorders of adolescence (imbalances of adrenal-ovarian maturation) which were to be conditioned each by 2 specified combinations of climate, diet and marital life.
Morphological alterations of IB-RS-2 cells cultured in serum-free maintenance medium after inoculation of swine vesicular disease virus (SVDV) were studied electron-microscopically. Cells harvested 0 to 3 hours after inoculation showed no alterations. Cellular alterations were observed from 4 to 7 hours after inoculation. Many vacuoles appeared just beneath the cytoplasmic membrane and were separated by thin cytoplasm. Narrow pathways were sometimes seen in the degenerative cells. They occasionally ran from the vacuole just beneath the cytoplasmic membrane to the nuclear cistern. Ribosome-like granules were seen along the narrow pathway or diffusely in the cytoplasm, or accumulated to form an islet. Membrane-bound bodies were frequently noticed in the central region of the cell. Cytoplasmic blebs were sometimes seen projecting from the cell surface. Vacuoles, narrow pathways and cytoplasmic blebs were absent in cells cultured in the medium with serum. So serum might exert some effects on the cytoplasmic membrane or cytoskeletal framework. Crystalline arrays of SVDV were found in the cytoplasm of degenerative cells harvested from 4 to 7 hours after inoculation. They were near the ribosome-like granules and membrane-bound bodies, but had no relationship with vacuoles just beneath the cytoplasmic membrane and narrow pathway. Their size and morphology were the same as those seen in the cells cultured in the medium with serum.

The epoxide hydratase inhibitor, 1,1,1-trichloro-2,3-propene oxide (TCPO) in combination with benzo[a]pyrene (B[a]P) was injected s.c. in ddN mice. The formation of fibrosarcoma by B[a]P was slightly accelerated at low dose of TCPO, and remarkably inhibited at high dose of TCPO. The correlation of carcinogenesis with B[a]P metabolism was discussed.
Methylurea, a precursor of carcinogenic nitroso compound, was formed by incubating methylamine and carbamyl phosphate in neutral buffer. The formation was checked on a radiochromatogram, as well as by color reaction. The presence of methylamine and carbamyl phosphate in preserved, fermented foods provided a suitable condition for the formation of methylurea.

A total of 524 fecal samples were collected from healthy swine of 36 hog farms scattered all over Japan from which had been detected neutralizing antibody against swine vesicular disease (SVD) virus. A virus was isolated from 21 of them. It was neutralized by antiserum against SVD virus. Of the 21 samples, 19 were derived from six farms, in areas which remained to be free from SVD in the past years and two from a farm where SVD broke out 18 months before. The cross-neutralization test was carried out with seven strains of SVD virus isolated from healthy pigs (SVDV-H) and five strains of SVD virus isolated from diseased pigs (SVDV-D). There was, however, no significant difference in antigenicity between the two groups. Some strains of SVDV-H were antigenically close to the Faulkner strain of Coxsackie B5 virus, and others to the freshly isolated strain of this virus. Neutralizing antibody of low titer against SVD virus was detected from pigs kept in areas free from SVD. It was presumed to have been produced in these pigs involved in silent infection with this virus.
An infection experiment was carried out on pigs with swine vesicular disease virus isolated from healthy pigs (SVDV-H). Inoculation was done by two routes, intradermal in the coronary band of the foot and oral. Observation was made on the formation of vesicles and their spread, the virus contents of serum, swab of the oral cavity, and feces, the vicissitude of neutralizing antibody titers, and the distribution of virus in the body. From its results the pathogenicity of virus was judged. In the pigs inoculated intradermally there was a difference in the extension of the area involved in vesicular formation between any two strains of virus. That is, vesicular formation was restricted to the site of inoculation, involved the site of inoculation and the sole of the hoof, or spread over the oral and nasal regions. In every pig, however, vesicles developed only for 2 approximately 5 days after inoculation. After that, repair progressed rapidly. Some strains caused viremia, which was mild. The virus was detected from the site of vesicular formation, but not from any organ. Neutralizing antibody began to be detected 3 days after inoculation. Its titer reached a plateau about 10 days later. In the pigs inoculated perorally, no vesicles were formed. The virus was only detected from the tonsils and the intestinal contents. These findings made it clear that SVDV-H was less pathogenic than swine vesicular disease virus isolated from diseased pigs.
Crystal formation of swine vesicular disease virus (SVDV) in IB-RS-2 cells was studied by electron microscopy. Cells were harvested 0, 3, 3.5, 4, 4.5, 5, 6 and 7 hours after inoculation. Crystalline arrays of SVDV was first observed in the cytoplasm of a few cells 4.5 hours after inoculation. In the cytoplasm of many cells harvested at 5 hours, 1 to 3 crystalline arrays of SVDV were observed. After that, a small number of cells had crystalline arrays in the cytoplasm. The cells with crystalline arrays were rich in ribosome and polysome with dilated mitochondria and many tiny vesicles. An individual virus particle was ca. 18 nm in diameter, and the center-to-center space ca. 22 nm. Crystalline arrays varied in size depending on the plane of section.
L-Keflex is a newly manufactured cephalexin product in order to maintain effective blood level of the drug for a long period of time. The results of the fundamental and clinical studies are as follows: 1. Mean blood levels of the drug after its single oral dose of 1 g in fasting in 22 cases before operations were: 7.19 mcg/ml at 2 hours (peak), 4.35 mcg/ml at 4 hours, 4.21 mcg/ml at 6 hours, 2.47 mcg/ml at 8 hours and 1.81 mcg/ml at 12 hours, respectively. Existence of the drug in blood was observed for a long period of time. 2. The distribution into the tissues of L-Keflex was generally good. The tissue levels in 19 of 22 samples ranged from 1.30 to 18.0 mcg/g, but 3 samples did not detect the drug in the tissues. Tissue level/blood level ratios were 0.19 approximately 2.67. 3. Half of 30 cases with mild dental infections was treated with a daily dose of 1 g with clinical response of 60.0%, and the other half with 2 g was 78.6% in clinical response. The overall efficacy rate was 69.0%. 4. As a side effect of the drug, only one of 52 cases (fundamental 22 cases, clinical 30 cases) complained of edema on both eyelids. No. abnormality was observed in blood finding, hepatic and/or renal function. From the above results, it is considered that L-Keflex is an effective antibiotic product in infections in the field of oral surgery. Also, L-Keflex has an advantage in that its administration frequence (b.i.d) is less than that of regular cephalexin (q.i.d.).
