Isolations of a cartilage factor that inhibits tumor neovascularization.

A cartilage fraction isolated by guanidine extraction and purified by affinity chromatography inhibits tumor-induced vascular proliferation and consequently restricts tumor growth. This fraction contains several different proteins; the major one has a molecular weight of about 16,000. The fraction strongly inhibits protease activity.

Cortisone sensitive T-cells in Peyers' patches.

Pretreatment of donor lymphoid cells with cortisone has been shown to depress the T-cell subpopulation responsible for cellular proliferation in the GVH reaction. A quantitative assay as well as the histological criteria of the GVH reaction have been used in this study to demonstrate the presence of cortisone-sensitive T-cells within the Peyer's patches as well as in the spleen and mesenteric lymph nodes in the rat.

Prospective evaluation of periodic breast examination programs.

A mathematical model is developed to obtain prospective estimates of average tumor sizes and the expected proportion of positive regional lymph node cases for periodic breast cancer examination programs. Semiannual, annual, and biannual programs, with or without mammography, are evaluated and compared with results from the National Breast Poject. Calculations indicate that a semiannual clinical-mammographic program can be expected to result in about 37% fewer positive node cases than those observed in the National Breast Project and substantially small tumors. The expected reduction in positive node cases from a similar annual program is about 30%, and from a biannual program, even without mammography, about 20%. Calculations also indicate the important role of diligent patient self-examination in such programs, especially for the detection of fast-growing tumors.

Biosynthesis of deuterated riboflavin: structure determination by NMR and mass spectrometry.

The riboflavin-producing fungus Eremothecium ashbyii was cultured in various growth media containing high concentrations of deuteriuj, and the product was isolated. The structures of highly deuterated riboflavin, in which at least 13 of 15 nonexchangeable hydrogens were replaced by deuterium, and fully deuterated riboflavin, in which all 15 nonexchangeable sites contained deuterium, were established by NMR and mass spectrometry. The aromatic protons (C-5 and C-8) wer partially substituted in the highly deuterated molecule. Information regarding three areas of the biosynthetic pathway within the microorganism was obtained as a result of the formation of these compounds. Extensive solvent interaction, possibly due to passage of sugar through the transaldolase-transketolase pathway, occurs during formation of the ribityl chain. Limited solvent participation takes place during formation of 6,7-dimethyl-8-ribityllumazine, the immediate precursor of riboflavin. Deuteration of the riboflavin C-6 and C-7 methyl groups indicates significant solvent exchange during the final step of the biosynthetic process.

Drug abuse in junior high school populations.

Illegal drug use in a population of 3,807 junior high school students in a suburban area appeared strongly correlated with family instability, student personal problems, and low academic performance. Use of tranquilizers, amphetamines, and sedatives was often motivated by attempts at self-medication to reduce painful feelings. Over 14% of illegal users took an overdose with about half of them intentional. Illegal users and nonusers agreed that drug education programs and the threat of legal penalties had little influence in their decisions about drugs.

Ethyl apovincaminate therapy in neurovascular diseases.

Of a series of 100 patients 46 were given combined (i.m. and oral) treatment with ethyl apovincaminate (RGH-4405, Cavinton) in daily doses of 10-30 mg; 54 were put on oral Cavinton (30-45 mg daily). Significant and relatively quick improvement was obtained in reversible vascular diseases, such as hypertensive encephalopathy, intermittent vascular cerebral insufficiency, in the early stage or light cases of cerebral endarteriitis and cerebral arteriosclerosis. On Cavinton effect hypoxic character of the EEG improved, and so did performance in psychodiagnostic tests, alertness and memory in the first place, furthermore changes of the eyeground. The doses administered did not damage parenchymal organs, they did not cumulate and when used in combination incompatibility could not be observed.

[Formation and persistence of latexagglutination-inhibiting antibodies in rhesus monkeys infected with Coxsackie virus type B4 (author's transl)].

The formation and persistence of latexagglutination-inhibiting (LAH), neutralizing (N), and complement-fixing (CF) antibodies were in Rhesus monkeys afterr experimental infection with coxsackievirus type B4. In all 3 animals the LAH antibodies showed a significant decrease between the 49th and 56th day after infection. In 2 cases the CF antibodies declined to 25% of their maximum titer on day 84 after infection, and in 2 animals the N antibodies declined between the 70th and 84th day afte infection.

[Immunosuppressive role of the liver in the graft versus host reaction].

The ability of the liver to reduce the intensity of the graft versus host (GVH) reaction has been investigated in F1 hybrid rats implanted with parental lymph nodes. Intrahepatic and intrarenal tissue implantations were compared using classical GVH criteria. The intrahepatic implantation of lymph nodes suppress the mortality observed after intrarenal implantation. The results confirm the interest of portal drainage in organ transplantation and suggest a new site of implantation for lymphoid cells.

[Treatment of myasthenia gravis].

The authors discuss various therapeutical possibilities for myasthenia, such as drug therapy, immuno-suppression, radiological, and operative treatment. The indicatons of the different therapeutical methods are dealth with. Thymectomy was performed in 92 of our patients. Due to complex therapy the fate of patients with myasthenia is to be regarded as much more favourable than in the past.

The frequency of adverse drug reactions as dependent upon age, sex and duration of hospitalization.

In 1972, 171 (18, 7%) of 914 patients showed evidence of an adverse drug reaction (ADR) in a prospective study of the division of internal medicine in a regional teaching hospital (Zieglerspital, Bern). The following factors correlated positively with frequency of ADR: 1. The duration of hospital stay. 2. The age between 60 and 80 years. 3. Female sex. 4. Previous ADR. The fact that most of the ADR occur within the first three weeks of a hospital stay, combined with point 1, suggests early ADR having some predictive value for duration of hospital stay. Statistical data of type, severity, duration and pathogenesis of ADR are given. The drugs involved are mentioned.

Gene V protein of fd bacteriophage. Dimer formation and the role of tyrosyl groups in DNA binding.

Gene V protein exists principally as a dimer in neutral buffers which are 0.15 M in NaCl, NaF, or NaClO4. Higher concentrations of NaClO4 or NaCl disrupt the dimers, but higher concentrations of NaF do not. There is a single sulfhydryl group per gene V protein monomer in native monomers, dimers, and DNA-protein complexes. Disulfide formation leads to loss of protein solubility and DNA binding capacity. The fluorescence of tyrosyl groups is the same for monomers and dimers in NaCl and NaClO4 solutions, but it is extensively quenched on binding to poly(dT) and fd-DNA. Complexes of gene V protein and fd-DNA isolated from lysates of infected cells were found to contain 4.70- +/- 0.13 nucleotides per monomer of gene V protein whereas complexes formed in vitro contain 4.05 +/- 0.17 nucleotides/monomer. It is postulated that tyrosyl groups are not involved in the protein-protein interactions of the monomer-dimer equilibrium, that tyrosyl groups stack with DNA bases in the complexes, and that each subunit of gene V protein in the intracellular complexes with fd-DNA is replaced by exactly two subunits of major coat protein during final assembly of the virus.

Limited form of Wegener granulomatosis. Eye involvement as a major sign.

Bilateral uveitis was the initial manifestation of a limited form of Wegener granulomatosis in a 47-year-old man. Diagnosis was established by biopsy of a pulmonary nodule. Subsequently, evidence of lesions developed in the patient's central and peripheral nervous systems, but renal lesions were absent. Pulmonary lesions regressed on therapy with azathioprine and prednisone. Blindness ultimately resulted from retinal phlebitis, vitreous hemorrhages, and neovascular glaucoma.

[Persistence of latex-agglutination inhibiting and neutralizing antibodies against enterovirus in the healthy population (author's transl)].

Sera of 122 persons were examined by the neutralization and latex-agglutination inhibition tests for antibodies against Polio type I, Coxsackie virus type A 9 and B 4, as well as Echo virus type 9. With increasing age there was a titre difference between latex-agglutination inhibiting and neutralizing antibodies, due to the earlier fall in the former. These antibodies, contrary to neutralizing ones, were in the lower titre region. The results confirmed the usefulness of the latex-agglutination inhibition test in the diagnosis of acute enterovirus infections.

Isolation of 19alpha-H-lupeol from Maclura pomifera.

A reinvestigation of the constituents of the Osage orange (maclura pomifera) yielded, in addition to the previously reported triterpenses (lupeol, butyrospermol, and lupan-3beta,20-diol), the pigments osajin and pomiferin, and a previously unreported constituent. The structure of this new compound was investigated. On the basis of spectroscopic and chemical data, it appeared to be an epimer of lupeol and is referred to as 19alpha-H-lupeol.
