A screening method for new antimicrobial agents from streptomyces culture filtrates was developed. Three different types of test organisms were used for determining the antimicrobial activities of the culture filtrates: they were (a) an antibacterial activity test against Staphylococcus aureus, (b) an antimycoplasmal activity test against Acholeplasma laidlawii, and (c) a cytotoxicity test against HeLa-S3 cells. The active filtrates showing antimicrobial spectra (anticellogram) which did not match those of known antimicrobial agents were used for further purification to obtain new antimicrobial agents. Of these screening methods, antimycoplasmal activity against Acholeplasma laidawii was the most sensitive for discriminating new compounds contained in the filtrates of known antibiotics.
Carbohydrate metabolism was studied in eight patients with Down's syndrome, aged 8 to 18 years. Diabetic glucose tolerance was observed in only one patient with obesity. This impaired glucose tolerance was improved with the weight reduction due to diet restriction. The flat glucose tolerance curves with low peak values were also observed in the other two patients. In the remaining five patients, normal glucose tolerance was obtained. Insulin response and free fatty acid levels during oral glucose load were not characteristic except for the diabetic patient with obesity. The sera of Down's syndrome showed normal binding capacity for insulin. These results suggest that obesity might partly participate in the impaired glucose tolerance in Down's syndrome.

A simple proton magnetic resonance spectroscopic method is described for the identification and and confirmation of several stereoisomeric paris of C27 stanols as well as their keto and acetate derivatives related to cholesterol. The method, which involves the use of lanthanide shift reagents, is useful in distinguishing clearly between the isomeric pairs differing only in the geometry of a functional group and/or of the A/B-ring junction in the steroid skeleton.

The effects of peptidoglutaminase (PGln-ase) I and II on human pancreatic juice amylase purified as an isozyme were investigated. Several amylase isozymes were formed which corresponded to minor components of pancreatic amylase isozymes, indicating that appearance of amylase isozymes are due to enzymic deamidation. A similar result was observed when the purified amylase isozyme was incubated with the supernatant of human pancreatic juice whose amylase was previously removed by adsorption onto raw corn starch. These findings are discussed in connection with amylase isozymes in the sera of the patients suffering from pancreatic inflammation.

A case of pancreatic heterotopia in a lymph node is described. Small pieces of pancreatic tissue were found incidentally in four lymph nodes located around the common hepatic artery in a 65-year-old man. Both exo- and endocrine elements of the pancreas, together with ductal structures were found to constitute the heterotopic tissue. No authentic case of heterotopic pancreas in lymph nodes has been previously reported in the literature.
The lymphoma cells from a patient with leukemia lymphoblastic sarcoma (Kiel classification) were observed by scanning and transmission electron microscopy. These cells were also examined by E, EA, EAC rosette-formation tests and by the indirect immunofluorescence technique for surface immunoglobulins. The malignant cells showed failure of rosette-formation or absence of surface immunoglobulins. Scanning electron microscopy revealed that many uniform protrusions were present on the cell surfaces. These surface protrusions were different from those seen on E-or EAC-rosette-forming cells. Ultrastructurally, the malignant cells were characterized by long profiles of rough surfaced endoplasmic reticulum with regular, narrow cisternae which radiated from Golgi area to the periphery of cytoplasm. These appearances differed from those observed in T-or B-lymphoma cells.
