Regulatory factors of lymphocyte-lymphocyte interaction. II. Characterization of human mitogenic factor derived from the culture supernatant of a mouse-human hybridoma.

Cell hybridization of phytohemagglutinin (PHA)-stimulated human peripheral blood lymphocyte (PBL) with murine lymphoma (EL-4) provided three hybridomas (MHH-16, MHH-20, and MHH-22) which spontaneously produced human mitogenic factor (MF). MHH-16 was serially subcloned by limiting dilution procedures, which resulted in maintaining two subclones producing human MF spontaneously for more than one year (PQL-3 and PQL-5 subcloned lines). Human MF MHH-MF) derived from supernatants of PQL-5 line cultures had a molecular weight (m.w.) of about 26,000-30,000 daltons (the major peak) with a minor peak with an m.w. of 15,000 daltons on Sephadex G-100 chromatography, and at a high concentration of NaCl (1 M), the activity of the 26,000-30,000-m.w. fraction became weak and that of the 15,000-m.w. fraction became predominant. MHH-MF had an isoelectric point of Ph 5.0-6.5. On DEAE-cellulose chromatography, MHH-MF was eluted at a fairly low salt concentration (sodium phosphate buffer 0.02 M, pH 8.0, NaCl 10 mM). After periodate treatment of this MHH-MF, the mitogenic activity almost disappeared. MHH-MF was relatively unstable to heating at 56 c for 20 min. In the presence of tunicamycin (0.3 microgram/ml), an inhibitor of N-linked glycosylation the synthesized MHH-MF showed a decrease in m.w. as follows: the major peak shifted from 26,000-30,000 to 23,000 daltons and the minor peak from 15,000 to 10,000 daltons on Sephadex G-100 chromatography. In internal labeling experiments with [3H] leucine, the 3H-labeled MF was partially purified, with mitogenic activity as a guide. This 3H-labeled MHH-MF fraction could be absorbed by PHA blasts but not by normal PBL. On SDS-PAGE under reducing conditions, only the radioactive peak of the 15,000-dalton fraction was recovered. MHH-MF obtained from the hybridoma culture supernatants may be a dimer of the 15,000-dalton fraction and a glycoprotein.

[Cystic meningioma associated with intratumoral hemorrhage in a child--report of a case and review of the literature].

The case reported here was a 6-year-old girl who complained of sudden headache and nausea followed by vomiting and visited our clinic. Physical examination revealed mental confusion, right abducent palsy and papilledema. Computed tomography demonstrated a round high-density area surrounded by a low-density zone indicating cystic nature and including high-density part. The lesion was located in the right parietal lobe attaching to the flax. Right frontoparietal craniotomy disclosed a brain tumor containing a large cyst in which bleeding from the tumor was confirmed. This mass lesion was totally resected out. Histo-pathological diagnosis was meningioma to contained cystic formation and hemorrhage from the tumor. Therefore, further discussion on the pathogenesis of cyst formation and bleeding from this tumor was presented.

Regulatory factors of lymphocyte-lymphocyte interaction. I. Con A-induced mitogenic factor acts on the late G1 stage of T-cell proliferation.

DNA synthesis in murine lymphocytes was augmented by a soluble factor in the supernatant of serum-free cultures of syngeneic spleen cells activated with concanavalin A (Con A). This so-called mitogenic factor (MF), which is probably identical with interleukin II, partially purified by DEAE-cellulose and Sephadex G-75 chromatography, is a fairly homogeneous molecule of 17-25 X 10(3) daltons. By using partially purified MF, the role of MF in lymphocyte proliferation was investigated. Pretreatment of lymphocytes with Con A for the first 3 hr of culture, which does not in itself induce cell proliferation, markedly augmented the effect of MF. The presence of MF, however, is necessary only in a restricted stage(s) of lymphocyte proliferation. The addition and removal of MF at various times during culture showed that MF exerts its effect on a process which occurs 3-6 hr before the beginning of DNA synthesis. These results strongly suggest that MF regulates the proliferation of mitogen-stimulated lymphocytes by acting on a restricted stage(s) of the cell cycle.

[Biological activity of prostaglandin E1 derivative, ONO-802 on smooth muscle (author's transl)].

The effects of prostaglandin (PG) E1 derivative, 16,16-dimethyl-trans-delta-2PGE1 methyl ester (ONO-802) on the rat stomach fundal strip, uterus and ileum were studied. Its substrate specificity for 15-hydroxy prostaglandin dehydrogenase (PGDH) was also examined. Contractions were recorded isometrically by superfusion method. PGE1, E2, and F2 alpha caused maximal contraction of the fundal strip immediately after application and relaxation to the original state within 10 min. Contractile pattern of the fundal strip produced by ONO-802 was quite specific and different from those by classic PGs. Contraction was increased slowly to its maximum in 10 to 20 min after application of ONO-802 and thereafter returned to the original state in 20 to 40 min. Contractive response of the rat ileum to ONO-802 was a gradual increase of the tonus and restoration in more than 120 min after reaching its maximal tonus. The rat uterus responded to 1 microgram of ONO-802 with double maximal contraction amplitude of spontaneous activity, with increased contraction amplitude restored in 90 min. In a combination of ONO-802 with PGE1, E2 and F2 alpha, the contractile response of the fundal strip to ONO-802 was masked by the actions of classic PGs and additional potentiation was not observed. Contractive activity of ONO-802 was not reduced by placental PGDH.

Vagus cardioinhibitory fibers in rats.

In chloralose-urethane anesthetized rats, compound spike potentials provoked in the cervical vagus nerve with electrical stimulation of the central cut end were found to consist of three major groups, A, delta-B and C. A remarkable cardioinhibition was observed on repetitive stimulation of the vagus nerve with an intensity which gave rise to the delta-B spike potential group. However, when the stimulus intensity was increased further beyond the level where the delta-B potential group had reached the maximum, the potency of cardioinhibition continued to be reinforced with the development of the C potential group. Selective activation of C fibers by anodal block of conductions along A and delta-B fibers was still associated with a considerable degree of cardioinhibition. These findings indicate that activities of both C fibers and delta-B fibers contribute to vagal cardioinhibition in rats and the view that the vagal cardioinhibition is mediated by B fibers, although valid in cats, can not be held applicable to all species of mammalians.
