Small dose of 2-deoxy-D-glucose completely excludes the overgrowth of fibroblasts.

The present work has been undertaken to assess the effects of a small dose of 2-deoxy-D-glucose on proliferation of fibroblasts in culture by morphological criteria. Fibroblasts in culture medium with 2-deoxy-D-glucose proliferated only to a slight degree, but cell confluence was not achieved. They became swollen and the cytoplasm was increasingly occupied by lucent area. These changes became prominent on the 10th day of incubation. When cell mixtures of fibroblasts and TYK-nu (cell line of undifferentiated adenocarcinoma of the ovary) or TYK-nu only were incubated under the influence of 2-deoxy-D-glucose, the degenerative changes aforementioned were observed only in fibroblasts, while no significant changes were ascertained in the cell line of the cancer by morphological criteria. These results suggest that a small dose of 2-deoxy-D-glucose shows considerable promise in specific inhibition of fibroblastic proliferation during the process of establishing the cell line of the cancer.

Effects of mechanical trauma of blood cells on dynamic viscoelasticity of blood during clotting.

Effects of mechanical trauma of platelets and red blood cells on dynamic viscoelasticity of blood during clotting were examined. Two different methods were attempted to give the mechanical damage to blood cells. In one method, a flow apparatus consisting of a thin teflon tube and two sample reservoirs connecting to both ends of the tube was employed. After a blood sample repeated the shuttle flow many times through the tube, the dynamic rigidity modulus (G') and loss modulus (G") of the blood sample during clotting were measured. The mechanical trauma of platelets brought about the increase of G' and G" for clots of PRP and whole blood, in addition to shorter clotting time. Slight hemolysis of red blood cells (less than 0.5% of total hemolysis) only yielded shorter clotting time for whole blood. In other method, packed red blood cells were hemolyzed by exposing the ultrasonic wave (the amount of hemolysis was about 5% of total hemolysis). Addition of supernatant of hemolyzed red blood cells to PRP caused the platelet aggregation. Furthermore, a marked hemolysis brought about higher values of G' and G" for blood clot, in addition to shorter clotting time. These results suggest that the release of constituents from damaged red blood cells and alteration of cell surface due to mechanical damage would accelerate the coagulation sequence.

[Effect of a traditional herbal medicine, tokishakuyakusan, on the fetal development of the spontaneously hypertensive rat].

Forty-three pregnant spontaneously hypertensive rats were administered 1.5% saline and divided into 2 groups: The experimental group of 21 rats and the control group of 22 rats. The animals in the experimental group were administered daily 500 mg/kg of Tokishakuyakusan. No drug was administered to the animals in the control group. Systolic blood pressure measured from 4 to 18 days of pregnancy was similar in both the experimental and control groups. Mean (+/- S.E.) levels of blood viscosity in the experimental group measured on the 19th day of pregnancy were 13.6 +/- 1.6 centipoise (cp) at a shear rate of 0.5/sec and 3.5 +/- 0.1 cp at a shear rate of 230/sec and were significantly lower than those of the control group (19.1 +/- 1.6 cp at 0.5/sec, p less than 0.02; 4.4 +/- 0.2 cp at 230/sec, p less than 0.001). There were observed no significant differences in hematocrit, erythrocyte deformability and fibrinogen levels between two groups. There was observed no significant difference between the mean number in the litter in the 2 groups. The mean weight of the fetuses in the experimental group delivered on the 19th day of pregnancy was 2.4 +/- 0.1 g, being significantly heavier than that of the control group (2.0 +/- 0.0 g) (p less than 0.01). These results indicated that Tokishakuyakusan accelerated development of fetuses of hypertensive rats by utero-placental circulatory improvement which was caused by decreased maternal blood viscosity.

Erythrocyte filterability and fetal development in normal pregnancy.

The relationship between maternal erythrocyte filterability determined in late pregnancy and infant birth weight was studied in uncomplicated pregnancy. There was a significant positive correlation (r = 0.7671, p less than 0.005). Maternal erythrocyte filterability is suggested to be an indicator of flow state of the placental microcirculation, which affects fetal development.

Results of HMG (Humegon)-HCG therapy in 6096 treatment cycles of 2166 Japanese women with anovulatory infertility.

The effects of HMG (Humegon)-HCG therapy in 6096 cycles in 2166 Japanese women with anovulatory infertility were examined. The rates of ovulation, pregnancy, the ovarian hyperstimulation syndrome, multiple pregnancy, abortion, and malformations in the newborn were recorded, and the possible factors of multiple pregnancies were analyzed. Ovulation occurred in 73.2% of the cases and 64.5% of the treatment cycles. Pregnancy occurred in 23.0% of the cases and 8.6% of the cycles. Ovarian hyperstimulation syndrome with grade I of WHO definition or more was observed in 10.3% of the cases and 5.3% of the cycles. The incidence of the ovarian hyperstimulation syndrome was high in amenorrheic patients, who respond to progestin with bleeding. The multiple pregnancy rate was 20.5%, of which 13.0% was twins and 7.5% triplets or more. The abortion rate was 22.0%, and the abortion rate in multiple pregnancy was significantly higher (P less than 0.05) than that in singleton pregnancy. The external malformation rate was 1.68% in the 594 newborn who could be examined. No significant differences were found in maternal factors, the treatment schedule, or the ovarian response to treatment in singleton and multiple pregnancy groups. This survey revealed that the efficacy and the incidence of adverse effects of Humegon-HCG therapy in a large number of Japanese women were not different from those in Caucasians except for a lower rate of multiple pregnancy, and no special causative factors for multiple pregnancy were found.

A new rheological method to measure fluidity change of blood during coagulation : application to in vitro evaluation of anticoagulability of artificial materials.

In order to attempt in vitro evaluation of antithrombogenecity for materials of artificial blood vessel tube, a new type of rheometer was developed. The rheometer originally consists of a cylindrical tube suspended from a torsion wire and filled with blood. The tube is excited in torsional oscillation and subsequent damped oscillation is observed. The apparatus can sensitively follow the change of fluidity during coagulation of blood. The damped oscillation curves during coagulation for fibrinogen - thrombin solution and blood put in a cylindrical tube made of the artificial material were measured. For fibrinogen - thrombin solution with lower fibrinogen and thrombin concentrations, the values of logarithmic damping factor (LDF) during coagulation increased and then decreased through a maximum. For blood and fibrinogen-thrombin solution with the higher concentrations of fibrinogen and thrombin, LDF monotonously decreased with the progress of coagulation. With a glass tube, the decrease of LDF for whole blood taken without anticoagulant rapidly occurred within about 15 min after sampling, while, with expanded polytetrafluoroethylene (EPTFE; Goar tex) and polydimethylsiloxane (Silastic) tubes, the decrease of LDF proceeds over 40-60 min. The present method is probably available for in vitro evaluation of anticoagulability or antithrombogenecity of artificial materials.

Idiopathic thrombocythemia and pregnancy: report of a case.

Pregnancy and delivery in patients with idiopathic thrombocythemia have not been reported elsewhere, to the knowledge of the authors. Such a case is presented herein. The platelet count decreased with progression of pregnancy, falling to a normal level near term. It was, however, again increased in the puerperium, reaching a level of 1570 X 10(3) per mm3 eight months after delivery. Aspirin was continuously administered throughout pregnancy to prevent thrombus formation, but no adverse effects on the delivery or the infant were observed.

Erythrocyte filterability and adenosine triphosphate levels in normal pregnancy and puerperium.

The filterability of erythrocytes was determined by a filtration method using erythrocytes suspended either in phosphate-buffered saline or in native plasma. Erythrocytes from pregnant women in the second and third trimester were significantly less filterable than those from non-pregnant women. On 0 to 1 day after delivery, erythrocyte filterability was further reduced, being significantly lower than that in the third trimester of pregnancy. Erythrocyte adenosine triphosphate (ATP) levels during pregnancy were not significantly different but in early puerperal period it was significantly lower than that in non-pregnant women. One month after delivery the filterability and ATP content of erythrocytes returned to the non-pregnant levels. The size and hemoglobin concentration of erythrocytes were unchanged during pregnancy and puerperium.

Blood viscosity and erythrocyte deformability in gestational diabetes.

Hemorheological properties were investigated in 15 gestational diabetics and compared with those in 20 normal pregnant patients at 37 weeks gestation. Whole blood viscosity in gestational diabetes was increased significantly. Erythrocyte deformability measured by a filtration method was decreased significantly in gestational diabetes. A hypothesis that placental hypertrophy observed in gestational diabetes might be produced as a compensatory reaction to hypoxia caused by intervillous microcirculatory disturbance due to the abnormal hemorheology was proposed.

Methods to study rheological properties of blood during clotting.

In vitro rheological studies on clotting of fibrinogen solution, plasma and blood are reviewed. Multiple viscoelastorecorder (MVER) and viscoelastorecorder (VER) were developed for determining the dynamic rigidity modulus (G') and loss modulus (G") during clotting. The effect of some parameters on clotting and the properties of fibrin clot are discussed. The parameters include measuring frequency, measuring shear strain and surface coating of measuring cell. The change of dynamic rigidity during clotting was compared between normal plasma and plasma from patient with Glanzmann's disease. By detecting the stress at the higher harmonic frequency, the non-linear terms of G' and G" were determined.

Dynamic evaluation of aggregation and agglutination of red blood cells.

Viscoelastic evaluation of aggregation and agglutination of red blood cells was attempted. A concentric double cylinder viscoelastometer was used for determining the dynamic rigidity modulus and loss modulus of blood sample. The dynamic rigidity modulus of horse blood were measured over a wide range of hematocrit. The relation between the viscoelastic behavior and the erythrocyte sedimentation was examined. The change in the amount of surface charge of enzyme treated red blood cells was qualitatively estimated from the measurements of dynamic viscoelasticity of red blood cells suspension with added poly-L-lysine. The dynamic rigidity modulus of red blood cells agglutinated by adding lectin (concanavalin A) were also measured.

[Hemodilution and anemia in pregnancy and fetal development].

The relation between hemodilution or anemia of pregnancy and fetal development was studied in 350 pregnant women who were delivered of full term singleton infants and were without any complications except iron deficiency anemia. The mean birth weight of the infants whose mother's hematocrit levels were 28.0-31.9% in 28-32 weeks' gestation was significantly higher than those of infants whose mother's hematocrits were 32.0-35.9% and 36.0-39.9%. There was also observed the same relation between birth weights and mother's hematocrits in 36-37 weeks' gestation. The incidence of small for dates infants was not significantly different between pregnant with different hematocrits. The iron deficiency anemia of pregnancy had no adverse effects on fetal development. It was suggested that hemodilution of pregnancy had an important physiological role in improving placental microcirculation to accelerate fetal development.

[Erythrocyte deformability and adenosine triphosphate (ATP) levels in normal pregnancy and puerperium].

Erythrocyte deformability and erythrocyte adenosine triphosphate (ATP) levels were measured in normal pregnancy and puerperium. Erythrocyte deformability was determined by measuring filtered volumes of red cell-plasma or buffer suspension per second. Erythrocytes from pregnant women at 16-27 weeks, 28-31 weeks and 36-41 weeks of gestation were found to be significantly less deformable than erythrocytes from non pregnant women. On 0-1 day after delivery, erythrocyte deformability was significantly reduced, compared with that at 36-41 weeks of gestation. Erythrocyte ATP levels at 16-27 weeks, 28-31 weeks and 36-41 weeks of gestation were significantly lower than that in non pregnant women. In puerperium, erythrocyte ATP levels dropped significantly compared with those in late gestational period. From these results, it was suggested that the depressed erythrocyte deformabilities were accompanied by a lowering of erythrocyte ATP levels in pregnancy and puerperium.

Relationship between focal penicillin spikes and cortical spindles in the cerveau isolé cat.

Using the unanesthetized, cerveau isolé preparation in the cat, the association between artificially induced penicillin (PCN) spikes and spontaneously occurring electrocorticographic (ECoG) spindles was investigated. Spikes were elicited by surface application of small pledgets of PCN. After the application of PCN, there was a decrease in spindle amplitude but no change in frequency, duration, or spindle wave frequency in the area of the focus. Examination of the times of occurrence of the spikes and spindles disclosed that in the majority of cases, within a few minutes of the initiation of the foci, there was very high simultaneity, usually 100% between the occurrences of these two events. Examination of the times of occurrence of the spikes within the ECoG spindles failed to disclose any compelling evidence which would favor either the hypothesis that spikes "trigger" spindles or the hypothesis that spindles predispose to focal spikes. Thus, whether spikes trigger spindles, or spikes simply occur in a nonspecific manner during the occurrence of the spindle, or whether it is a combination of both these explanations, must remain an open question on the basis of the data available.

Rheological behaviors of bovine blood forming artificial rouleaux.

A purpose of the present study is to make an artificial rouleau of bovine red blood cells which is not capable of rouleau formation under physiological condition. Rheological behaviors of bovine blood forming artificial rouleaux were examined. The modification of cell surface by enzyme trypsin induced rouleau formation, whereas the modification of cell surface by neuraminidase did not cause any aggregate formation. The drastic elevation of the fibrinogen content in bovine red blood cells suspension also brought about the formation of rouleau. The value of dynamic rigidity modulus G' of bovine red blood cells in saline solution containing high concentration of fibrinogen is somewhat smaller than that of trypsin treated bovine red blood cells in plasma. The value of G' of trypsin treated bovine red blood cells in plasma first increased to a maximum value and then decreased with the time. It is supposed that the removal of macro-molecules from the cell surface facilitates the mutual approach of cells and causes the formation of rouleau which seems to be the same as that of human and horse bloods.
