Degradation of human normal immunoglobulin preparations and the level of measles virus antibodies.

Seventy-four batches of normal human immunoglobulin of placental origin (NHIp) and 25 batches of the preparation derived from the serum (NHIs) were tested. The batches were divided into groups (A-E) according to production date and presence of sediment. The degree of degradation of NHI preparations was determined measuring the components soluble in trichloroacetic acid (TCASC), performing molecular filtration or carrying out electrophoresis in polyacrylamide gel. The level of antibodies to measles virus was determined with the enzymatic immunoadsorption test (ELISA) and haemagglutination inhibition test (HIT). It was found that NHI preparations were undergoing significant degradation before their expiration date. The content of TCASC ranged from 1.83% for NHIs preparations recently produced (group E) to 10.57-10.63% respectively in NHIp preparations with sediment (groups B and A). Molecular filtration made possible isolation of a polymer, a monomer, and degradation products. The level of antibodies against measles virus was the lower (7,400) the greater was the degree of degradation of NHIp preparations in the relation of NHI preparations recently produced (21,500).

Cytomegalovirus in the mononucleosis syndrome in children.

In 7 children aged 18 months to 7 years isolated from a group of 128 children with infectious mononucleosis the cytomegalovirus infection was found. Infection was diagnosed by determination of antibodies against immediate early and late CMV antigen by means of the ELISA test. Besides that, antibodies were determined against the capsid antigen and early antigen of EB virus by the method of indirect immunofluorescence. In four children only cytomegalovirus infection was found and three had a mixed infection with both viruses and the diagnosis in these cases was: infectious mononucleosis (due to EBV) with coexistent or following CMV infection. In the sera of two children with cytomegalovirus mononucleosis changes were observed in the antibodies against EBV which is explained as a result of interactions between CMV and EBV in the organism of the host.

Complement and circulating immune complexes in children with mumps meningitis.

Haemolytic activity of complement (CH50) and occurrence of circulating immune, complexes (CIC) were studied in serum samples from children with mumps meningitis taken at regular intervals. The decrease of CH50 was observed in 46 out of 77 samples (60%) in the Ist examination, between 1-6th day from the appearance of symptoms, in 43/67 samples (64%) in the IInd examination, between 12--16th day of disease, and in 7/22 samples (32%) in the IIIrd examination, between 12--14th month of observation since the appearance of symptoms. Typical changes of complement level during the disease were of such profile that the most pronounced decrease was in the Ist and IInd examination with the normalization in the IIIrd examination. The presence of CIC was found in 21%, 15% and 14% of samples in the Ist, IInd, and IIIrd examination respectively. Serum sample form the child on 5th day of disease, with high concentration of CIC (482 micrograms/ml), was subjected to sepharose 6B gel filtration on the calibrated column. The molecular weight of thus analyzed immune complexes was as follows: 2.5 X 10(6) and 5.0 X 10(5) daltons. CIC contained anti-mumps antibodies, determined by Elisa, in IgG, IgA and IgM classes.

Cytomegalovirus antibodies in renal transplant recipients.

The relationship between CMV infections and the development of various viral antibodies were studied in successively taken sera from 5 renal kidney recipients. In all of the cases tested the infections were connected with the development of antibodies to immediately early antigens and then specific IgM and antibodies to various CMV early antigens. The increase of antibodies measured by ELISA, CF and immunofluorescence for late CMV antigens were observed in samples taken from two patients. Based on serological findings the relation between CMV infections and quantity of blood received before transplantation was established.
