The concomitant injection of isoproterenol with house dust allergen inhibited the immediate skin allergic reaction in asthmatic patients. Pretreatment with dibutyryl cyclic AMP showed an inhibitory effect on the whealing formation to the allergen. Those results showed the inhibitory effect of isoproterenol on the skin reaction through increasing intracellular cyclic AMP levels.

Though gamma-Oryzanol has been applicated as the autonomic agent in various fields for a few decades, recently its effect on lipid metabolism comes to be much more interested. We studied on the effect of gamma-Oryzanol on climacteric disturbance, and besides on the difference of lipid peroxides level which has an important and indispensable connection with mary syndromes involving aging. We obtained the following results from 40 subjects with climacteric disturbance after administration of gamma-Oryzanol Fine Particule 1.5 g (gamma-Oryzanol 300 mg included) for 4 to 8 weeks. 1. 90% of the cases improved generally; Excellent 16 (40%), Good 14 (35%), Effective 6 (15%), No change 4 (10%). 2. Effect on climacteric disturbance was judged by the Kupperman method and the Ueda method; generally recovered 85%, 80% each. 3. Significantly reduced total cholesterol, triglyceride and increased HDL-cholesterol were noted in cases with hyperlipidemia (TC greater than or equal to 220 mg/dl, TG greater than or equal to 140 mg/dl, HDL-chol. less than or equal to 45 mg/dl). Therefore, atherogenic index was excellently recovered. 4. In cases with climacteric disturbance lipid peroxides level was high, and they were significantly recovered by administration of gamma-Oryzanol. 5. During this study, no side effects have been recognized and no particular change in function test was found. Thus, we confirmed the efficacy and utility of gamma-Oryzanol on many complaints in climacteric disturbance and also on lipid metabolism.
The structure of the branching site of poly(ADP-ribose) was determined as O-alpha-D-ribofuranosyl-(1"' leads to 2")-O-alpha-D-ribofuranosyl-(1" leads to 2')-adenosine-5',5",5"'-tris(phosphate) by gas chromatography, mass spectrometry, and 1H-NMR measurements. Thus the structures of all the ribose-ribose linkages known in poly(ADP-ribose) are uniformly alpha(1 leads to 2)glycosidic bond. This indicates that branching ADP-ribosylation and elongating ADP-ribosylation of poly(ADP-ribose) synthesis are catalyzed by similar alpha(1 leads to 2)-specific ADP-ribosyl transferases or the same enzyme. Poly(ADP-ribose) glycohydrolase, which specifically hydrolyzes the ribose-ribose bonds of poly(ADP-ribose), also cleaves the ribose-ribose-ribose bonds at the site of branching.

In an attempt to devise a screening test for aldosterone-producing adenoma (APA) among hypertensive patients, the serum sodium and potassium levels, plasma renin activity (PRA), plasma aldosterone concentration, and aldosterone-PRA ratio were measured in 348 patients with hypertension. Nine patients with a substantially elevated aldosterone-PRA ratio were selected and hospitalized for further investigations. All nine patients were then recognized by scintigraphy with labeled cholesterol, venography, and surgical excision as having APA. The serum concentration of potassium was subnormal in three of nine patients with APA. In patients with APA, administration of diuretics and salt restriction significantly elevated PRA. However, even under notable diurnal and day-to-day variations of plasma aldosterone concentrations, the aldosterone-PRA ratio was always elevated inappropriately (more than 400) in patients with APA. In contrast, after administration of diuretics, both the PRA and aldosterone levels increased significantly in patients with essential hypertension, but the aldosterone-PRA ratio was less than 200. Since the renin-angiotensin system seems to be a major factor controlling aldosterone secretion in normal subjects, it is suggested that an elevation of aldosterone-PRA ratio more than 400 is a useful screening tool for the prediction of APA among hypertensive patients.

A new platelet aggregation inhibitor compound, 5-(2-chlorobenzyl-4,5,6,7-tetrahydrothieno[3,2-C]pyridine hydrochloride (ticlopidine), was examined for its inhibitory effects on blood-borne metastasis using three different rodent tumors (B16 melanoma, Lewis lung carcinoma, and rat ascites hepatoma, AH130). Ticlopidine was administered p.o. to the rodents. It inhibited the aggregation of platelets induced by adenosine diphosphate, thrombin, crude extract of AH130, and viable AH130 and B16 melanoma cells and also resulted in a significant decrease of pulmonary metastasis induced by i.v. injection of B16 melanoma and AH130. Spontaneous pulmonary metastasis of Lewis lung carcinoma was also inhibited by p.o. administration of ticlopidine. This new compound may be a useful agent for inhibiting platelet aggregation caused by various agents and for suppressing hematogenous pulmonary metastasis.
In an attempt to determine the effects of antihypertensive treatment on proteinuria and renal function, these factors were studied in 155 patients with essential hypertension, before and during antihypertensive therapy. Slight renal impairment tended to increase with the progress of hypertension, as evidenced by a slight but significant increase in the levels of blood urea nitrogen (BUN) and serum creatinine, and by a progressive decrease in creatinine clearance. A decrease in proteinuria appeared as early as one month after initiation of treatment and continued for at least two years during treatment. Apparently the proteinuria was caused by increased glomerular pressure and by vascular damage in the glomerular vessels. In 7 patients, normalization of blood pressure resulted in an elevation of BUN and creatinine concentrations, although initial renal function did not differ significantly from that in other groups who did not show such derangement during treatment. Thus, a reduction of proteinuria seems to be a good criterion for predicting beneficial results from antihypertensive treatment.
