Dynamic aspects of the contractile system in Physarum plasmodium. III. Cyclic contraction-relaxation of the plasmodial fragment in accordance with the generation-degeneration of cytoplasmic actomyosin fibrils.

Plasmodial fragments of Physarum polycephalum, excised from anterior regions of a thin-spread plasmodium, contracted-relaxed cyclicly with a period of 3-5 min. The area of the fragments decreased approximately 10% during contraction. In most cases, there was little endoplasmic streaming which indicates that contractions were synchronized throughout the fragment. By both polarized light and fluorescence microscopy, the organization and distribution of the cytoplasmic actomyosin fibrils in the fragments changed in synchrony with the contraction cycle. The fibrils formed during the contraction phase, and finally became a highly organized framework consisting of a three-dimensional network of numerous fibrils with many converging points (the nodes). During relaxation, the fibrils degenerated and disappeared almost completely, though some very weak fibrils remained near the nodes and the periphery. The results obtained by fluorometry of the fragments, stained with rhodamine-phalloidin, suggested that the G-F transformation of actin is not the main underlying process of the fibrillar formation.

Synthesis of hydrocarbons under presumed prebiotic conditions using high-frequency discharge.

Various hydrocarbons were synthesized by high-frequency discharge in a primordial terrestrial model atmosphere. The products were extracted by benzene or methanol and analyzed by GC-MS. The mean carbon chain length of the hydrocarbons formed by the discharge through pure CH4 gas was less than 6. Benzene was also obtained. Some isomers were obtained for each of the hydrocarbons containing a given number of carbons. When a small amount of C2H2 was added to the CH4, longer chain compounds were formed, as compared with discharge in CH4 only. However, when the amount of C2H2 was increased, unextractable high molecular weight compounds were produced. The amounts of products decreased as the mixing ratio of CO2 to CH4 increased. No hydrocarbons were detected when the ratio of CO2/CH4 exceeded 1.

Enzymatic synthesis and some properties of a model primitive tRNA.

A model primitive tRNA with the nucleotide sequence GGCCAAAAAAAGGCCp was synthesized using T4 RNA ligase. The nucleotide sequence of this newly synthesized oligonucleotide was confirmed by ladder analysis of several enzymatic digestion products. The secondary structure of the oligonucleotide was examined by comparison of the products of its digestion by single- and double-strand-specific nucleases with those of the digestion of the intermediate oligonucleotide GGCCAAAAAAAOH. The results indicated that the two GGCC segments of the 5' and 3' ends of the model tRNA may form base pairs in solution. The same conclusion was derived from the result of affinity-column chromatography of the model oligonucleotide. When 32pGGCCAAAAAAAGGCCOH was passed through a poly(U)-agarose column, about 70% of the applied sample bound to the poly(U)-agarose. In contrast, when the model oligonucleotide was passed through a poly(C)-agarose column, only 15% of the sample bound to the poly(C)-agarose. These results indicate that the newly synthesized oligonucleotide adopts a hairpin structure in solution. Two aspects of a potential biological activity of the synthetic model tRNA were examined. It was found that the oligonucleotide can bind to poly(U)-programmed 30S ribosomes and is recognized by Q beta replicase as a template for RNA synthesis.

Effects of furosemide versus isolated ultrafiltration on extravascular lung water in oleic acid-induced pulmonary edema.

We studied the effects of no treatment, furosemide treatment, and isolated ultrafiltration on extravascular lung water (ETVL) in mongrel dogs in whom pulmonary edema was induced with oleic acid. In all treatment groups, ETVL was significantly elevated 90 min after oleic acid infusion. At 270 min, we found no difference between nontreatment and furosemide. There was, however, a significant difference between no treatment and ultrafiltration but not between furosemide and ultrafiltration. In spite of observations which suggest that ultrafiltration is of benefit in reducing ETVL, we could not demonstrate superiority of one therapy over another.

The effects of d-propranolol and captopril on post-ischaemic acute renal failure in rats.

The effect of the combined therapy of d-propranolol and captopril was evaluated in post-ischaemic acute renal failure in rats. The glomerular filtration rate measured 24 hours after ischaemic insult in the animals receiving no drugs was 54 +/- 11 microliter/min/100g body weight while in animals treated with the combination of d-propranolol and captopril it was 305 +/- 35 microliter/min/100g body weight (p less than 0.05). The precise mechanism of protection afforded by the combination therapy is not clear, but this approach could be useful in protecting the kidney from ischaemic damage.

Differential coupling efficiency of chemically activated amino acid to tRNA.

Interaction based on possible chemical affinity of an amino acid for tRNA was examined as a model for the aminoacylation of primitive tRNA without aid of an enzyme system. Two types of reaction were carried out and compared. One was the acyl linkage of amino acid to the 5'-terminal phosphate of a tRNA activated as an imidazolide. The other was the incorporation of an amino acid activated as an imidazolide into 2'(3')-hydroxyl groups of intact tRNA. Both types of reaction indicated that none of the amino acids tested had any selectivity for the tRNAs examined. However, the rates of reaction with a given tRNA were different among amino acids. In the second type of reaction, amino acids were found mainly at loop-out regions of tRNA, but not at either its 5'- or 3'-terminal sites.

Formation of amino acids from models of Titan and more oxidized atmospheres.

Protein and non-protein amino acids were synthesized following hydrolysis of products obtained by high frequency discharge techniques applied to model atmospheres consisting of N2 as a nitrogen source together with CH4 and/or CO2 as a carbon source. Highest yields were obtained in the absence of CO2 and from mixtures rich in CH4. Amino acids would indeed be expected on the frozen surface of Titan with its CH4-N2 atmosphere.

Catalytic mechanism of histone in peptide formation from phenylalanyl adenylate.

Catalytic action of histone on peptide formation from phenylalanyl adenylate was studied. A peptide bond formation from phenylalanyl adenylate releases two protons. The proton release was increased in the presence of histone at a higher concentration. The catalytic action seems to be due to basic circumstance induced by histone, since (1) basic condition enhanced the proton release; (2) basic amino acid did not enhance the proton release.

[Experimental studies on tissue distribution of 14C-labelled antimicrobial agent in the otorhinolaryngological field--application of macro-autoradiography].

The distribution of antimicrobial agents in infected target tissues would be essential for their application to treat the sites of infection. Taking accounts of the view point, we had reported the distribution of 3 nalidixic acid analogs in tissues which are related to otorhinolaryngology by macro-autoradiography and radiometry. We investigated here in the distribution of a nalidixic acid analog OPC-7241. This agent is 9-fluro-5-methyl-8-(4-methyl-1-piperaginyl)-6, 7-dihydro-1-oxo-1H, 5H-benzo [ij]quinoligine-2-carboxylic acid supplied by Otsuka Pharmaceutical Co., Ltd. Autoradiography was performed 30 minutes after intravenous administration of 14C-OPC-7241 in 2.96 MBq(80.0 microCi)/12.5mg/kg to New Zieland White rabbit. Radiometry was performed 30 minutes, 1 hour and 3 hours after intravenous administration of 14C-OPC-7241 in 740 kBq (20.0 microCi)/10.0 mg/kg to NZW rabbits. Such care was taken in filling the carboxymethyl cellulose paste into the paranasal cavity, nasal cavity, oral cavity and external ear canal not to damage mechanically. Horizontal frozen sections parallel to mandibular basis were cut in 50 microns thickness in a cryostadt. Blackening of 14C-OPC-7241 was most significant in soft palate and its expanse which is apparently the same gland-like tissues as soft palate when stained with hematoxylin and eosin. Significant radioactivity was recognized in tonsils, the cartilage of external ear canal, septal cartilage, periodontal membrane, dental pulp, muscle and concha nasalis ventralis. The levels was low in ethmoid cells, mucosa of tympanic cavity, cochlea, maxillary sinus and bone. Quantitatively, the levels of 14C-OPC-7241 radioactivity were high in soft palate, mandibular gland and tonsils. Radioactivity was significant in tongue and concha nasalis ventralis. The levels were low in turbinates, ethmoid cells, maxillary sinus, septal cartilage, septal mucosa, cochlea, brain, optic nerve and lens. Thus radiometric results agreed with the above autoradiographic findings. Macro-autoradiography can be one of useful means for the evaluation chemotherapeutic agents for possible clinical application.

Effect of polynucleotides and a basic protein on the condensation of phenylalanyl adenylate.

The effect of polyuridylate and of histone on the oligomerization of phenylalanyl adenylate was tested. Polyuridylate which pairs with adenine under suitable conditions showed no effect. Histone increased the rate of the polymerization whereas a neutral protein, albumin, had no effect. Simultaneous presence of nucleotides diminished the effect of histone. The implication of these results on the prebiotic polypeptide formation is discussed.
