Europium-152 depth profile of a stone bridge pillar exposed to the Hiroshima atomic bomb: 152Eu activities for analysis of the neutron spectrum.

The 152Eu activity depth profile of a granite pillar of the Motoyasu bridge located 132 m from the Hiroshima atomic bomb hypocenter was assessed. The pillars each measured 82 cm in depth, 82 cm in width and 193 cm in height. One of the pillars was bored and 6.8-cm-diameter core samples were removed and cut into 2-cm-thick disks. Two gamma rays of 152Eu, 122 keV and 344 keV, in each disk were measured using a low background, gamma-ray spectrometer, and the activity distribution was determined as a function of depth in the granite. A concentration of stable Eu in the granite was determined by activation analysis. The specific radioactivity of 152Eu and 154Eu at the pillar surface was determined to have been 117 and 24 Bq per mg Eu, respectively, at the time of detonation. The value of 152Eu agrees within 20% of that calculated by Loewe. The depth profile of 152Eu in granite demonstrates a distinct difference from the estimates made only by thermal neutrons. Present data provide valuable information for the analysis of the neutron spectrum of the Hiroshima atomic bomb and its intensity.

[Increase of blood flow in human tumor tissue under angiotensin II-induced hypertension].

On the basis of the functional characteristics of microcirculation in tumor tissue, the clinical results of angiotensin II-induced hypertension chemotherapy (IHC) have been reported. The present paper deals with the results of measurement of blood flow in human tumors transplanted into athymic rodents using the electrolytic hydrogen clearance method. When the mean arterial blood pressure was elevated to 140-150 mmHg by the continuous infusion of angiotensin II, the increase of blood flow was 30.4 +/- 4.7 ml/min/100g in H-111 human gastric carcinoma (the mean increase rate being 14.4 times that in the normotensive state), 15.6 +/- 2.6(X 3.0) in NS-8 gastric carcinoma and 11.0 +/- 2.9 (X 2.9) in TE-8 esophageal carcinoma, respectively. These results confirmed that blood flow remarkably increased not only in animal tumors but also in human tumors under the induced hypertension caused by angiotensin II. On dynamic CT under hypertension, enhancement of the delivery of contrast medium to the tumor area was also shown clinically, according to the increase in CT numbers in the same ROIs in a normotensive state.

[Pharmacokinetic and clinical studies on cefotaxime in plastic and reconstructive surgery].

Pharmacokinetic and clinical studies on cefotaxime (CTX) in plastic and reconstructive surgery field were carried out. The results obtained are summarized as follows. Two grams of CTX was administered intravenously by single bolus injection to each of 4 patients with burn blisters and mean levels of CTX concentration in blister exudates were investigated. Thirty minutes after administration, the mean level of CTX in the exudates was 3.90 micrograms/ml and it reached a peak of 9.81 micrograms/ml in 2 hours. The clinical efficacy rate for 30 patients with burn infections and postoperative infections was 73.3%, and the efficacy rate for 29 patients in prophylactic use was 72.4%. A side effect (eruption) and abnormal laboratory findings were observed in each one case out of 59. From the above results, CTX may be considered to be useful in plastic and reconstructive surgical treatments.

Purified protein kinase C phosphorylates microtubule-associated protein 2.

We have investigated actions of purified protein kinase C on microtubule- and microfilament-related proteins. Among the cytoskeletal proteins examined, microtubule-associated protein 2 (MAP2) was found to serve as a good substrate. Other cytoskeletal proteins, tubulin, fodrin, cofilin, tropomyosin, and 53,000-Da protein, were very poorly phosphorylated. The amino acid residues of MAP2 that were phosphorylated by the protein kinase C were almost exclusively serine. The peptide mapping analysis indicated that protein kinase C and cAMP-dependent protein kinase phosphorylate MAP2 differently. The ability of MAP2 to interact with actin was markedly reduced by this protein kinase C-mediated phosphorylation. These data raise the possibility that phosphorylation of MAP2 by activated protein kinase C may be involved in cell-surface signal transduction.

Binding of microtubule-associated protein 2 and tau to the intermediate filament reassembled from neurofilament 70-kDa subunit protein. Its regulation by calmodulin.

Two major brain microtubule-associated proteins (MAPs), MAP2 and tau, were found to bind to the intermediate filaments reassembled from neurofilament 70-kDa subunit protein (= 70-kDa filaments). The binding was saturable. The apparent dissociation constant (KD) for the binding of MAP2 to the 70-kDa filaments was estimated to be 4.8 X 10(-7) M, and the maximum binding reached 1 mol of MAP2/approximately 30 mol of 70-kDa protein. The apparent KD for the tau binding was 1.6 X 10(-6) M, and the maximum binding was 1 mol of tau/approximately 3 mol of 70-kDa protein. It was also found that MAP2 and tau did not compete with each other for binding to the 70-kDa filaments. Most interestingly, calmodulin, a ubiquitous Ca2+-binding protein in eukaryotic cells, was found to inhibit the binding of MAP2 and tau to the 70-kDa filaments. The inhibition by calmodulin was regulated by changes in Ca2+ concentration around 10(-6) M, and was canceled by trifluoperazine, a calmodulin inhibitor.

Sperm glycosidase as a plausible mediator of sperm binding to the vitelline envelope in Ascidians.

In the Ascidian, Ciona intestinalis, sperm alpha-L-fucosidase is concluded to be a recognition protein for the sperm receptor in the vitelline envelope. The spermatozoa bind to the vitelline envelope probably by forming rather stable substrate (alpha-L-fucoside)-enzyme complex at many spots. Experimental evidence indicates a good correspondence in the properties between receptor binding activity and alpha-L-fucosidase activity such as the effects of saccharides, of pH and of monoclonal antibodies raised against sperm alpha-L-fucosidase, and locus in the sperm. A general mechanism involving an enzyme as a recognition protein for the cell to cell binding is also discussed in relation to the molecular evolution of the recognition proteins.

The effect of drugs which affect the arachidonate cascade in burned rabbits.

A full skin thickness burn covering 35 per cent of the total body surface area was inflected on the back of rabbits. The animals were divided into seven groups and the effectiveness of OP-41483, aspirin (ASA), OKY-046 and CV-3988 was studied. All rabbits died between 8 and 24 h after injury in the no therapy group. Renal function tests in this group showed a decrease in Cler and an increase in FENa and CH2O. Such changes were improved slightly with infusion therapy. On the contrary, with OP-41483, OKY-046 and ASA, distinct improvements were observed (in order of extent of improvement), but with CV-3988 the improvements were rather small. As a result these drugs may also be effective in preventing organ failure after burn injury, but as such changes presumably occur immediately after the burn it is considered that a fast-acting agent such as OP-41483 is the most effective form of treatment.

[Clinical study on angiotensin-induced hypertension chemotherapy (IHC)].

It has been found in experiments using rats that there is a lack of autoregulation of blood flow in tumor vessels and a selective increase of blood flow under angiotensin II-induced hypertension when the arterial blood pressure dose not exceed 150 mmHg, while there is no increase in normal tissues (Suzuki et al., JNCI:1981). On the basis of the functional difference of microcirculation, IHC has been developed clinically since 1978. In the procedure of treatment, the mean blood pressure of the patients was maintained at 140-150 mmHg when anti-cancer drugs were administered along with the continuous intravenous infusion of angiotensin II. In a randomized controlled study on gastric carcinoma treated with an AFM regimen, the response rate was 42.5% (8/21) in IHC vs. 10.5% (2/19) in non-IHC (p less than 0.05). The "initial response time" (15.5 vs. 28.8 days) and the "effective tumor reduction time" (36.7 vs. 57.5 days) were significantly shorter (p less than 0.01) in the IHC group. Frequency and grade of side effects were not different statistically. In an open trial, the overall response rate was 39.6% (54/134) and each response was closely related to the difference of drug sensitivity of tumor types. For example, it was 90.0% (9/10) in cancers of the head & neck, 66.7% (4/6) in the breast, 42.8% (12/28) in the stomach, 46.2% (6/13) in the pancreas, 23.1% (3/13) in the colon and 23.8% (5/21) in the lung. The effect on metastatic lymphnodes was 79.4% (27/34), which was higher than that of primary (48.1% : 26/54) and other organ metastases (34.6% : 18/52). Finally, this paper dealt with the problem of clinical evaluation of tumor lesions with a lot of fibrous granulation tissue and coagulative necrosis, and of the investigation of differential imaging.

The effect of a prostaglandin I2 analogue (OP-41483) on renal function in burned rabbits.

It is known that the number of platelets decreases after burn injury. However, the mechanism of such a decrease and its influence on systemic organs have not been fully elucidated. In this study, we evaluated the efficacy of OP-41483 (a derivative of the prostaglandin I2 (PGI2) in burned rabbits with special reference to its association with renal function. Full thickness skin loss burns, covering 35 per cent of the total body surface area, were experimentally produced on the back of rabbits. Among the following groups of rabbits (six rabbits each), the time courses of renal function and platelet function were compared; Group I (no treatment), Group II (fluid therapy only), Group III (treated with OP-41483 + fluid therapy; IIIa: 50 ng/kg/min, IIIb: 75 ng/kg/min, IIIc: 100 ng/kg/min). The results show that in Group I, all rabbits died after 8 h. Renal function tests in this group showed a decrease in creatinine clearance and increases in FENa and CH2O. Such changes were improved in Groups IIIb, IIIa and II (in order of the degree of improvement). Group IIIc showed no improvement. As for the platelets the decrease in platelets was the smallest in Group IIIb, where platelets showed no decrease until 8 h after injury and the fibrinogen concentration increased. No improvement was found in Group IIIc by our tests, presumably owing to an overdose of OP-41483. The optimal dose of OP-41483 for improving the renal function of burned rabbits was 75 ng/kg/min in our study.

Relationship between hypoglycemic symptoms and blood glucose levels due to self-monitoring in summer camp for diabetic children in Japan.

In a summer camp for 47 diabetic children in Kinki district, Japan, in 1984, the relationship between hypoglycemic symptoms and blood glucose levels by self-monitoring was analyzed. During the 7-day camp, self-monitoring of blood glucose (SMBG) was carried out 599 times in total, 12.7 times per camper. SMBG due to hypoglycemic complaints amounted to 371. 154 measurements out of 371 indicated blood glucose levels under 80 mg/dl, but 78 monitorings were found to be over 200 mg/dl. Fatigue or weakness were the most frequent hypoglycemic symptoms, as was hunger sensation, each reaching approximately 40% in frequency. In most complaints of tremor, the blood glucose level was critically low. Prompt measurement of blood glucose is indeed necessary to properly treat diabetic children with 'hypoglycemic' symptoms.

Dosimetry comparison and characterisation of an Al K ultrasoft x-ray beam from an MRC cold-cathode source.

Ultrasoft x-rays of 0.3-5 keV have provided a unique tool for the investigation of intracellular mechanisms of radiation action in biological organisms, including mammalian cells. However, their use presents unique practical problems in dosimetry and experimental design. Detailed interpretation of the biological results requires reliable dosimetry and well characterised monoenergetic beams. This paper presents a comparison between two fundamentally different dosimetric techniques, namely the ionisation current in an extrapolation chamber and photon counts in a proportional counter. Agreement within 7% was obtained when these two methods were applied to an Al K x-ray beam (1.5 keV) from an MRC cold-cathode transmission target discharge tube as previously used in many biological experiments. Photographic film was calibrated as a relative dosimetric technique and used for investigation of the intensity uniformity of the radiation field. These techniques provide a comprehensive characterisation of the beam in the position of the biological cells, including photon flux (or absorbed dose rate), spectral purity (showing much less than 1% bremsstrahlung relative to characteristic Al x-rays) and uniformity over the irradiation area (within about 5% for mammalian cell irradiations).

Substoichiometric determination of cobalt in crud.

A substoichiometric extraction method with nitroso-R salt (NRS) has been studied for the determination of trace Co in crud. The Co-NRS complex is extracted substoichiometrically into Capriquat-CHCl3 at pH 6.5-9.0 in 20 min of shaking time. The analytical results obtained by the method are within 3% of relative error in the determination range of 5 to 50 micrograms. The proposed method is simple and has sensitivity of 0.5 micrograms, though Fe(II), Ni(II) and Cu(II) seriously interfere. The results applied for the determination of trace Co in crud are described.

Elimination of silica gel from gangliosides by using a reversed-phase column after preparative thin-layer chromatography.

A simple and effective procedure has been developed for eliminating silica gel from gangliosides after preparative thin-layer chromatography. Gangliosides were extracted from the scraped silica gel with chloroform-methanol-water 10:10:3 (by volume) and dried. The residue was suspended in 0.1 M NaCl and centrifuged. After filtration through a sintered glass funnel, the supernatant was applied to a reversed-phase column. The column was washed with 0.1 M NaCl, water, and methanol-water 1:1 (v/v). Gangliosides were eluted from the cartridge with methanol with yields of 92% and 90% in terms of dry weight and sialic acid content, respectively. Almost all of the silica gel contamination was eliminated from gangliosides by this procedure.

Diabetic osteopenia in central Japan.

A large scale study of diabetic osteopenia was conducted in central Japan. The degree of bone mass loss was measured in 1267 diabetic patients and 646 control subjects. Based on six indices obtained by microdensitometry, the bone mass of 21.1% of the diabetic patients was found to be markedly decreased and that of 8.2% was found to be severely decreased, while only 7.9% of the control group showed bone mass loss. The prevalence of diabetic osteopenia in female patients was significantly higher than in males, but a positive correlation between the degree of bone mass loss and age was observed in both the male and female patients. Patients requiring oral agents or insulin had significantly more bone mass loss than those on dietary therapy alone.
