Differential ion dependence of frog olfactory responses to various odorants.

1. Dependence of the fron olfactory bulbar responses on NaCl concentration greatly varied from odorant to odorant. The responses to odorants such as 1-carvone and isoamyl acetate were essentially unchanged by removal of NaCl, while those to odorant such as citral and beta-ionone were greatly decreased by removal of NaCl. 2. The NaCl requirement for the responses to certain odorants was greatly decreased by an increase in pH or temperature of the stimulating solution. 3. It was concluded that changes in ion permeability at the apical membranes of olfactory cells including olfactory ciliary membranes are not involved in generation of the in vivo olfactory responses to certain odorants.

Construction and nucleotide sequence analysis of an infectious DNA clone of the autonomous parvovirus, mink enteritis virus.

We have cloned the replicative form (RF-) DNA of mink enteritis virus (MEV), constructed an infectious recombinant plasmid containing MEV DNA and determined the nucleotide sequence of the cloned MEV DNA. RF-DNAs were detected and infectious virus was generated when the recombinant plasmid containing the entire MEV genome was introduced into feline kidney cell cultures. The MEV genome was 5094 nucleotides (nt) in length; the 3' end of the virion strand contained a 205 nt palindromic sequence and the 5' end a 62 nt palindromic sequence that could assume Y- and U-shaped configurations, respectively. The 5' end of the virion strand had a direct repeat of 61 nt at the carboxyl terminus of the capsid protein gene. The organization of the MEV genome is similar to those of canine parvovirus (CPV) and feline panleukopenia virus (FPLV); there are two large open reading frames (ORFs), one in the 3' half and the other in the 5' half of the genome, with coding capacities of 668 and 722 amino acid residues, respectively. Both are in the same reading frame and no significant ORFs are apparent in the virion strand (negative-sense strand). Possible functional promoter motifs are located at map unit (m.u.) 4.5 and m.u. 40, and a possible functional poly(A) signal is located at m.u. 96. The nucleotide and amino acid sequence homology with CPV and FPLV is greater than 98%, consistent with the hypothesis that MEV and CPV are host-range variants of FPLV.

Is the predisposition to neuroleptic malignant syndrome genetically transmitted?

A Japanese family consisting of a mother and her two daughters, all experienced NMS on therapeutic or subtherapeutic doses of neuroleptics. The mother and the elder daughter had past histories of severe neuroleptic-induced extrapyramidal symptoms. The predisposition to this syndrome may be genetically transmitted. We suggest that neuroleptic treatment should be introduced with extreme caution to a patient with a family history of NMS.

[A long survival case of small cell lung cancer synchronized with renal cancer].

A long survival case of small cell lung cancer synchronized with renal cancer was reported. The patient was a 73-year-old male, complaining of cough and fever up. The chest roentgenogram showed a tumor mass in the right lower lung field. The specimen obtained from transbronchial lung biopsy of right S8b was diagnosed as small cell carcinoma of lung. In the check of the metastasis to other organs, abdominal CT scanning and the echogram demonstrated a solitary mass in the left kidney. We supposed a possibility of primary renal cancer rather than the metastasis from the lung because of being solitary mass, no existence of the metastasis except the kidney, and from the finding of the renal angiography. The patient underwent left nephrectomy for the renal cancer, and also underwent right lower lobectomy for the lung cancer after neo-adjuvant chemotherapy using cisplatinum and carboquone. Pathologically, the renal lesion was diagnosed as typical clear cell carcinoma of the kidney. He has survived for more than 4 years.

Determination of a new calcium antagonist, sesamodil fumarate (SD-3211), and its metabolite in plasma by liquid chromatography with electrochemical detection.

A sensitive and selective high-performance liquid chromatographic method is described for the determination of a novel calcium antagonist, (+)-(R)-3,4-dihydro-2-[5-methoxy-2-[3-[N-methyl-N-[2-[(3,4- methylenedioxy)phenoxy]ethyl]amino]propoxy]phenyl]-4-methyl-3-oxo-2H- 1,4-benzothiazine hydrogen fumarate (sesamodil fumarate; JAN, SD-3211, I), and its N-desmethylated metabolite (II) in plasma. Compounds I and II and an internal standard were isolated from plasma by solid-phase and liquid-liquid extraction. The extract was chromatographed on a reversed-phase C18 column, and the compounds of interest were detected by dual coulometric electrodes operated in an oxidative screen mode. The limit of determination for both I and II was at least 0.4 ng/ml in plasma. The utility of the assay was demonstrated by determining plasma levels of I and II in five dogs administered an oral dose of 60 mg of the drug.

Trial production and application of an optical system for measuring the movements of organs composed of soft tissue.

A new optical system to measure movements of less accessible organs composed of soft tissue is proposed. The sensor unit is composed of a phototransistor and two light-emitting diodes (LEDs). The LEDs act as light sources and are driven by two phase signals with a phase shift of 90 degrees. The distance between the sensor and the measuring object can be obtained as a function of the phase angle of reflected light detected by the phototransistor. This system can be easily constructed and simultaneously records data from multiple points in real time. The relative precision was 0.5 mm and the resolution was 0.05 mm. Output characteristics of this system were not significantly affected by the reflection factor of the object. Tongue movements in speech production were observed by use of this system.

Neuroendocrine differentiation in a case of acinic cell carcinoma of the parotid gland.

We report a case of acinic cell carcinoma of the parotid gland with neuroendocrine differentiation. Light microscopically, the tumor appeared as clear cell-type acinic cell carcinoma. In addition, the tumor showed neurosecretory features such as Grimelius positivity and the presence of neurosecretory granules by electron microscopy. We suggest that a tumor cell arising from a stem cell can show simultaneous differentiation to both neuroendocrine and acinic cells.

Identification of virus-specific antigens in cultured cells infected with Marek's disease virus serotype 2 and CVI-988 strain.

For the identification of serotype-specific antigens of Marek's disease virus (MDV) serotype 1 (MDV1) or serotype 2 (MDV2), a total of 24 hybridoma clones, secreting monoclonal antibodies (MAbs) against CVI-988 (MDV1) or HPRS-24 (MDV2) strain, were established and characterized by immunofluorescence assay, virus neutralization and immunoprecipitation analysis. Based upon the molecular weights (mol. wt.) of the immunoprecipitated polypeptides, the MAbs were subdivided into 7 groups. Among them, two groups of MAbs reacted with antigens that have not been reported, were identified. MAbs belonging to the first group reacted with CVI-988- and MDV2-specific antigens with mol. wt. ranging from 29 K to 34 K (29/34 K). This antigen was not found in cells infected with Md/5 and JM strains of MDV1, and the results of kinetic analysis of antigen expression showed this antigen appeared to be related to late membrane antigens. MAbs belonging to the second group immunoprecipitated MDV2-specific antigens with mol. wt. of 37 K, 33 K and 31 K from HPRS-24-infected cells or with those of 37 K, 34 K and 31 K from SB-1(MDV2)-infected cells, and these antigens appeared to be related to early antigens. MAbs belonging to the other 5 groups included those which recognized similar antigens reported previously or the antigens characterized insufficiently in this study.

[Observation of oral actions using digital image processing system].

UNLABELLED: A new digital image processing system to observe oral actions is proposed. The system provides analyses of motion pictures along with other physiological signals. The major components are a video tape recorder, a digital image processor, a percept scope, a CCD camera, an A/D converter and a personal computer. Five reference points were marked on the lip and eyeglasses of 9 adult subjects. Lip movements were recorded and analyzed using the system when uttering five vowels and [ka, sa, ta, ha, ra, ma, pa, ba[. RESULTS: 1. Positions of the lip when uttering five vowels were clearly classified. 2. Active articulatory movements of the lip were not recognized when uttering consonants [k, s, t, h, r[. It seemed lip movements were dependent on tongue and mandibular movements. Downward and rearward movements of the upper lip, and upward and forward movements of the lower lip were observed when uttering consonants [m, p, b[.

[Clinical significance of Su-polysaccharide (Su-PS) skin test in patients with primary lung cancer treated with chemotherapy and administration of OK-432, with special reference to the therapeutic effect and prognosis].

In order to evaluate the significance as a parameter for monitoring immunological status and for predicting therapeutic effect and prognosis, Su-PS skin test was performed in 68 cases of primary lung cancer treated with chemotherapy and administration of OK-432. In 16 of 68 cases, lymphocyte subsets of peripheral blood using Leu monoclonal antibodies were analyzed to investigate the immunological status. Su-PS skin test was performed at the time of before treatment, 1 week after administration of OK-432, 4 weeks after chemotherapy and intervals during treatment. The reactions to Su-PS skin test in these patients were divided into 3 groups according to the maximum size during the treatment as follows; less than 20 mm, 20 mm to 50 mm and over 50 mm. As compared with the reaction to Su-PS skin test and the survival, there was a significant difference between each group. The analysis of the lymphocyte subsets also indicated that immunological status tended to be activated more in strong responders to the Su-PS skin test than in weak responders. Therefore, Su-PS skin test is considered to be an useful immunological parameter of cancer patients under treatment.

[Investigation of UFT and radiation combination therapy of head and neck cancer--mainly in laryngeal cancer].

We studied the effects of simultaneous combination therapy with UFT and radiation for 69 head and neck cases with the same protocol in 6 universities in Kanagawa. Completed cases (cases in which therapeutic effects could be evaluated) totaled 67 patients who could receive more than 40 Gy as a given radiation dose and more than 8.4 g as a total UFT dose. Responder rate at 90th day after completion of radiation in 67 completed cases (CR 57 cases + PR 6 cases/67 cases) was as favorable as 94%. A comparison between 52 cases of the most frequent laryngeal cancer and historical control 113 cases received single radiation therapy by means of Kaplan-Meier method resulted significantly (P less than 0.05) favorable in UFT combination group as for disease free rate in all cases, disease free rate in Stage IV cases, disease free rate in glottic cancer, and survival rate and disease free rate in supraglottic cancer. No severe adverse reactions were observed in all cases. This combination therapy with radiation and UFT was judged effective for head and neck cancer, especially laryngeal cancer.

Characterization of replicative form DNA of the autonomous parvovirus mink enteritis virus.

Characterization of replicative form (RF) DNA of mink enteritis virus (MEV) was carried out. Most of the RF DNA were bound to terminal protein but some were free from the protein. The protein-free RF DNA increased about 7 times from 30 to 50 hr post-infection, while the DNA with protein increased less. The molecules of the replicative intermediate which were partially single-stranded DNA and bound to terminal protein were present. Two terminal conformations, "extended" and "turnaround," were observed in both ends of both terminal protein-bound and protein-free RF DNA. The 5' end labeling revealed that 5' ends of protein-free RF DNA were not blocked to phosphorylation by an amino acid or an oligopeptide which attaches to 5' ends of proteolytically deproteinized RF DNA. Restriction analysis of incomplete RF DNA which was partially double-stranded DNA showed that extended conformation was dominant in such incomplete RF molecules.
