Infection of human epithelial cell lines by Epstein-Barr virus. Interaction between epithelial cell and B lymphocyte.

The pathogenesis of nasopharyngeal carcinoma caused by the Epstein-Barr virus (EBV) has been studied experimentally by infecting monolayer cells derived from human normal nasopharynx with the B-lymphotropic EBV. Direct EBV infection of the cell sheets derived from the human nasopharynx resulted in the appearance of EBV-associated nuclear antigen-positive floating cells, suggesting that the monolayer cells have been transformed. Since these cells seemed to be of the B-type lymphocyte, it may be assumed that there must have been B lymphocytes among the cells of the monolayer which have undergone transformation by the virus. EBV infection of fused human epithelial cells (Ad-AH and D98-AH cells) and B-lymphoblastoid cells (BJAB cells) showed that the EBV receptors had been maintained for no longer than 24 h even when heterokaryons and hybrid cells were formed.

Ambulatory long-term vasodilator therapy for chronic refractory heart failure: hemodynamic evaluation and clinical response.

We evaluated long-term combined vasodilator therapy (hydralazine or ecarazine + isosorbide dinitrate) in 29 patients with chronic congestive heart failure resistant to the optimal conventional therapy. There were 24 men and 5 women, aged 28 to 76 years (mean 52 y/o). The etiology of heart failure was congestive cardiomyopathy in 24 patients, ischemic cardiomyopathy in 4 patients and advanced mitral regurgitation due to calcified mitral annulus in 1 patient. There were 21 patients in NYHA class III and 8 patients in NYHA class IV. All patients continued their previous therapeutic regimen during the period of this study. Hemodynamic measurements were performed with a triple lumen flow-directed balloon-tipped catheter in 20 patients to evaluate the effects of vasodilator therapy. In the rest of 9 patients, heart rate, blood pressure, chest X-ray examination for heart size (CTR) and M-mode echocardiograms for ejection fraction (EF) were monitored. The hemodynamic responses to the combined vasodilator therapy in 20 patients showed significant decreases in afterload and preload concomitant with an increase in cardiac output. The noninvasive evaluation of combined vasodilator therapy in 9 patients resulted in significant improvement in CTR and EF. We also noted a significant improvement in their symptoms of 29 patients. Side effects and drug toxicity were uncommon during vasodilator therapy. It is concluded that the combined vasodilator therapy is most useful adjunctive therapy in the management of severe refractory heart failure. Moreover, long-term nonparenteral vasodilators can be administered even at outpatient clinic without hemodynamic monitoring.

Tumorigenicity of nasopharyngeal carcinoma hybrid cell line.

The spontaneous production of Epstein-Barr virus (EBV) and tumorigenicity in the BALB/c mutant nude mouse were studied with the use of the following 4 cell lines derived from the human nasopharynx: a) Ad-AH, an 8-azahypoxanthine-resistant epithelioid cell line; b) A2L, an EBV-carrying lymphoblastoid cell line; c) A2L/AH, an EBV-carrying epithelioid hybrid cell line established by fusion of Ad-AH cells with A2L cells; and d) NPC-KT, an EBV-carrying epithelioid hybrid cell line established by fusion of Ad-AH cells with nasopharyngeal carcinoma (NPC) primary culture cells. The NPC-KT hybrid cell line was the only cell line to produce EBV antigens of the lytic cycle and virus particles. Cloning efficiency in agarose was clearly related to tumorigenicity in nude mice. Especially high, frequent tumor formation was observed in the heterotransplantation of NPC-KT hybrid cells that presented poorly differentiated carcinoma, and the tumor cells were positive for EBV-associated nuclear antigen. These NPC-KT hybrid cells maintained the characteristics of a histologic type of NPC tumor cells. Thus experimental systems of NPC were established in vitro and in vivo by the application of NPC-KT hybrid cells to NPC model cells.

Defective cellular immune responses in lupus nephritis.

To determine whether patients with systemic lupus erythematosus (SLE) and active nephritis have more profound defects in cellular immune responses, we studied cell-mediated immunity (CMI) in 12 patients with lupus nephritis (LN) and 11 patients with chronic mesangial proliferative glomerulonephritis (CGN) without renal insufficiency. The LN patients had a significant depression in CMI, characterized by impaired delayed hypersensitivity skin reactivity to keyhole limpet haemocyanin (KLH), decreased percentage of T cells and diminished lymphocyte transformation to phytohaemagglutinin (PHA). The levels of concanavalin A (Con A)-induced suppressor cell activity (SCA) were significantly lower in LN patients compared with those in normal controls and those in CGN patients. The levels of all CMI parameters for the 6 LN patients with the nephrotic syndrome (NS) were significantly more decreased when compared with those of the 6 LN patients without NS. In contrast, the CMI levels in CGN patients with or without NS did not differ from normal subjects. These findings may lend further insight into the understanding of the immunoregulatory defect in LN.

43Ca and 67Zn NMR spectra of Ca2+, Zn2+-concanavalin A solutions.

The half-band width of 43Ca NMR of free aqueous Ca2+ was scarcely increased by adding more than equal molar apo-concanavalin A(apo-Con A), suggesting that slow chemical exchange, koff less than 10 s-1, occurs for the Ca2+ ion from Con A. In contrast with the 43Ca NMR findings, the half-band width of 67Zn NMR of free aqueous Zn2+ was markedly increased by adding apo-Con A. The 67Zn NMR half-band width of Zn2+ in the presence of apo-Con A was decreased by adding excess Ca2+, but was increased by adding excess D-mannose. These changes of the half-band width were influenced mutually by D-mannose or Ca2+, respectively. The broadened half-band widths of Zn2+ in the presence of Con A were decreased by adding Mn2+, suggesting that Mn2+ was substituted for Zn2+ at a metal binding site of Con A.
