Identification of erythroid colony progenitors in a subset of human peripheral null lymphocytes devoid of Fc receptors.

This study was aimed at purifying the progenitors of erythroid burst units (BFU-E) from human peripheral blood. Human mononuclear leukocytes from five normal donors were fractionated into several mononuclear cell subpopulations, including null lymphocytes with (null Fc+) and without (null Fc-) receptor for the Fc fragment of immunoglobulin G, through a succession of rosetting procedures and discontinuous Ficoll-Hypaque gradient centrifugations. The fractionated cells were separately cultured for 14 days in plasma clots in the presence of erythropoietin. Among fractionated cell subpopulations large and numerous BFU-E derived colonies grew only from the Fc- null lymphocyte subpopulation. This fraction, representing less than 4% of all mononuclear cells, also contains cells (42 + 11%) capable to differentiation towards the B-cell and plasma-cell lineages. The Fc+ null lymphocytes, representing less than 9% of all mononuclear cells, contained 15.2 + 3.3% cells capable of differentiation toward the T-cell lineage. The whole null lymphocyte subpopulation generated half the number of BFU-E colonies expected from its content in Fc- null lymphocytes. These data demonstrate that the progenitor of erythroid cells (BFU-E) resides in a small heterogeneous null Fc- subpopulation of circulating lymphocytes and suggest that its in vitro differentiation, though generally subjected to inhibitory and enhancing influences from other circulating cell subpopulations, does not necessarily require interaction with other peripheral blood cells.

Effects of smokeless tobacco and snuff on oral mucosa of experimental animals.

The effects of smokeless tobacco on oral mucosa were studied in 60 Syrian hamsters. Histologic study of the buccal pouch mucosa revealed no significant pathologic changes at either ten or 20 weeks after the start of the experiment. However, studies on epithelial whole mounts revealed a statistically significant decrease in mitotic activity and a statistically significant increase in the number of Langerhans' cells in animals subjected to daily use of tobacco or snuff for 20 weeks.

Animal model for theophylline--cimetidine drug interaction.

Cimetidine (300 mg I.V.) and theophylline (15 mg/kg I.V.) were studied in Beagle dogs for possible drug interaction. The drugs were administered alone and in combination using a crossover design. Although none of the determined pharmacokinetic parameters for theophylline changed significantly in the presence of cimetidine, a trend towards significance was found for the terminal half-life, area under the curve, and mean residence time. The magnitude in changes found in Beagles is representative of the changes reported in man.

[Bone nocardiosis. Apropos of a case in a young child].

Osteitis of the forearm due to Nocardia asteroides was noted in an 8 year old tunisian boy 14 months after an open fracture. This observation of direct inoculation of bone by Nocardia asteroides was atypical because of the remarkable nature of the hyperostotic reaction.

[Pseudotumoral and subacute pathology of the epididymis. Contribution of ultrasonics].

Three patients presented with atypical subacute epididymal disorders diagnosed clinically as a scrotal tumor. Ultrasound imaging provided satisfactory morphologic data on lesions. Cold surgery was performed, either because of major functional disturbance (multiple epididymal abscesses) or because of doubt with respect to diagnosis (spermatic granuloma and epididymal hematoma). Castration was the final procedure in each case. Even though this surgical attitude appears logical because of the serious nature of the lesions, the revised clinical diagnosis provided by ultrasonography and the very good correlation between pathologic and ultrasonic findings should lead to a more eclectic therapeutic attitude in other cases.

Binding of busulfan to plasma proteins and blood cells.

The reversible and non-reversible binding of busulfan to plasma proteins and to blood cells has been evaluated in man. The reversible binding to plasma proteins was insignificant in both healthy subjects and in patients with chronic myelocytic leukaemia. The percentage of busulfan irreversibly bound to plasma ligands was 32.4. Busulfan was distributed evenly between blood cells and plasma. The fraction of busulfan irreversibly bound to the blood cells was 46.9%. The major portion of busulfan that is degraded in whole blood reacts with the blood cells and the main part of that binds to macromolecular components in the cells.

[Echography of palpable abdominal masses in adults. Apropos of 92 cases].

A retrospective study was conducted of 92 cases of palpable abdominal masses in adults, lesions being of undetermined origin clinically but verified by ultrasound imaging. Final distribution of organs affected showed considerable divergence between the different intra- and retro-peritoneal organs and the various pathological conditions involved, with a predominance of malignant tumors. This dispersion means that a discriminating examination such as ultrasonography must be practised. Ultrasound imaging was able to identify the affected organ in 86 p. 100 cases using a precise semiological analysis (mass center, interface). Diagnostic difficulties (14 p. 100 cases) are due to restrictions in semiological factors, large masses, and certain topographic or morphologic clinical forms. Ultrasound regularly provides macroscopic data: shape of mass, and size reduced by approximately 15 p. 100 in all cases. Regional and metastatic extension is also underestimated in approximately 1/4 cases. If these limitations are accepted, the use of ultrasonography appears to be the primary procedure required, this determining the therapeutic decision.

Determination of busulfan in plasma by GC-MS with selected-ion monitoring.

A GC-MS technique with selected-ion monitoring is described for the determination of busulfan in plasma. Busulfan is extracted from plasma with methylene chloride and converted to 1,4-diiodobutane. Analysis by GC-MS with selected-ion monitoring (m/z 183) gave a relative standard deviation of +/- 4.3% (n = 5) at the 10-ng/ml level.

The radiologic diagnosis of thoracic aortic rupture on plain film.

This paper reviews the recent literature (1968-1982) describing the radiologic diagnosis of aortic rupture and attempts to define causal relationships between rupture and radiologic signs as well as assessing respective predictive values. Also presented is a case report of a patient with thoracic aortic rupture in whom esophageal (nasogastric tube) displacement occurred to the left rather than the right, in apparent violation of one of the more newly established radiologic signs of aortic rupture.

Busulfan kinetics.

Busulfan kinetics were studied in patients with chronic myelocytic leukemia after oral doses of 2, 4, and 6 mg. The plasma concentration-time data could be fitted to a zero-order absorption one-compartment open model. The elimination rate constant averaged 0.27 +/- 0.05 hr-1 (SD). The plasma AUC was linearly related to the dose. The lag time for the start of absorption, the time absorption ends, and the absorption rate constant showed some interindividual variations. About 1% of busulfan is excreted unchanged in urine over 24 hr.

A simple and economical method for the preparation of adsorbents for the thyroid hormone uptake test.

A low-cost and simple method has been developed for the preparation of human or animal protein-based adsorbents for use in the thyroid hormone uptake test. A combination of sodium sulphate precipitation and glutaraldehyde polymerisation of pooled whole serum or purified albumin preparations was employed to yield a fine solid-phase suspension which eliminates the need for capping and rotating assay tubes. Results for 107 samples obtained by this method correlated well with those obtained by use of a commercial kit.
