Simultaneous determination of bestatin and p-hydroxybestatin, a major metabolite, in human serum by gas chromatography mass spectrometry. Utilization of quantitative selected ion monitoring and deuterium labelled internal standard.

A specific and sensitive method for the simultaneous determination of bestatin and its major metabolite, p-hydroxybestatin in human serum has been developed by selected ion monitoring using the corresponding deuterated internal standards. The N-trifluoroacetamide-O-trimethylsilyl either derivatives of the trideuteromethyl esters of bestatin and p-hydroxybestatin were suitable for quantitation by selected ion monitoring. The determination limit of these compounds by the present method is 1 ng ml-1 in human serum.

Uptake and excretion of 67Ga-citrate in malignant tumors and normal cells.

Using in vivo and in vitro experimental models, the uptake and excretion of 67Ga-citrate in tumor cells and normal cells were studied. The time-lapse accumulation of 67Ga in the tumor of rats bearing Yoshida sarcoma reached its peak 24 h after the administration of 67Ga and gradually decreased thereafter. However, the excretion of 67Ga from the tumor was less than that from normal lung. For culture cells in vitro, the uptake of 67Ga increased with lapse of contact time between 67Ga and the cells, but there was no distinct difference between the results for tumor cells and normal skin fibroblasts. The excretion of 67Ga from the cells tended to decrease with prolongation of the contact time, the excretion from tumor cells being only about 10% after a contact time of 24 h. This indicated a significant delay in excretion in comparison with that of normal skin fibroblasts. This delay in the excretion of 67Ga may be an important factor in the tumor accumulation of 67Ga.

Smoking-induced body sway and its suppression by periodic saccades.

The effect of cigarette smoking on body sway, the everlasting displacement of the center of gravity while standing upright, was studied and smoking was found to induce an unusual, oscillatory body sway that was higher in frequency and greater in amplitude than that in controls and lasted in general for several minutes after smoking. This characteristic body oscillation was strongly suppressed by periodic, saccadic eye movements. These results suggest that nicotine absorbed into blood during smoking affects the structures in the brain stem related to the regulation of standing posture and the saccadic eye movement.

Synthesis and antibacterial activity of new 1-oxa-1-dethiacephalosporins.

A series of 3-(deacetoxymethyl)-1-oxa-1-dethiacephalosporins (1) bearing the 7-(alpha-alkoxyimono)acyl side chain were synthesized and their antibacterial properties were examined in comparison with that of FK-749 (2). (Z)-2-(Ethoxyimino)-2-(2 aminothiazol-4-yl)acetic acid was found to be a prefered side chain in the 1-oxa series, and the derivative 18b with this side chain proved to be a potential broad-spectrum antibiotic not at all inferior to 2.

Wall reinforcement with highly porous Dacron mesh in aortic surgery.

Wall reinforcement with highly porous synthetic Dacron mesh was devised and evaluated in various aortic operations. Excellent attachment of the mesh to the aortic wall, good preservation of pliability of the aorta, and efficient pressure resistance with minimal scar tissue formation were the usual findings. This type fo wall reinforcement was employed in 60 patients for the following indications: 1) prevention of suture line complications; 2) wall reinforcement of aortic aneurysm without resection; 3) reinforcement of adventitia or autogenous vein following thromboendarterectomy. It is concluded that reinforcement with highly porous synthetic Dacron mesh should be superior to reinforcement with conventional prosthetic Dacron graft.

Determination of haloperidol in human serum by radioimmunoassay.

A radioimmunoassay has been developed which enables accurate determination of haloperidol in human serum. Antiserum was prepared by immunizing guinea pigs with haloperidol (O-carboxymethyl)oxime derivative (III) coupled with bovine serum albumin. With the antiserum, 3H-haloperidol and dextran-coated charcoal, the assay of haloperidol in serum was possible over a concentration range of 1 to 50 ng/ml, using 0.1 ml of human serum without the need of an extraction procedure. Data obtained by radioimmunoassay are in good agreement with those obtained by gas chromatography. No appreciable cross-reactivity was observed neither with haloperidol metabolites nor with other butyrophenone neuroleptics. Serum levels of haloperidol in schizophrenic patients receiving single oral dosing (6 mg/subject) have also been determined.

Comparative study between steroidal and nonsteroidal anti-inflammatory drugs on the mode of their actions on vascular permeability in rat carrageenin-air-pouch inflammation.

In an attempt to clarify mechanisms of anti-inflammatory actions of steroidal and nonsteroidal anti-inflammatory drugs with special reference to the possible role of E type prostaglandins in the inflammatory process, mode of actions of these drugs was investigated by using acute exudative stage and chronic proliferative stage of carrageenin-air-pouch inflammation in rats. This inflammation was induced on the back of rats by subcutaneous injection with air and carrageenin solution. Plasma exudation in the inflammation locus was determined with the aid of radioiodinated serum albumin. In the acute exudative stage of this inflammation, a single oral administration of dexamethasone, indomethacin, phenylbutazone or aspirin all exerted potent anti-exudative effect, while in the chronic proliferative stage, only dexamethasone was effective in inhibiting the exudation of the labeled albumin. Prostaglandin E level in the exudate of the acute stage inflammation was suppressed by either dexamethasone or indomethacin in accordance with the inhibition of the vascular permeability. These results suggest (1) significant role of prostaglandin E in the acute exudative inflammation, (2) similarity between steroidal and nonsteroidal anti-inflammatory drugs in the mechanism of their anti-exudative actions in the acute inflammation, (3) independency of the exudative process in the chronic proliferative inflammation from prostaglandins and (4) role of extra-prostaglandin mechanism(s) for the anti-exudative action of the anti-inflammatory steroids.

Autoradiographic studies on the effect of allopurinol on 14C-hypoxanthine metabolism in the squirrel monkey.

The Effect of orally given allopurinol in the distribution of intravenously administered 14C-hypoxanthine radioactivity was studied in squirrel monkeys 8 hr after administration of the label by the whole body autoradiography. Although the distribution of radioactivity in the normal and allopurinol-treated animals was essentially similar to each other, more intense radioactivity was noted in the latter monkey; salvage of 14C-hypoxanthine was enhanced. Similarly to our previous observation in mice, significant radioactivity in monkeys was seen in tissues undergoing rapid nucleic acid synthesis except for slight species differences in some organs. 14C-Allantoin alone as the urinary metabolite of the hypoxanthine in the normal monkey whereas significant amounts of 14C-hypoxanthine and 14C-xanthine as well were detected in the urine of the drug-treated animal.

[Vaginal radical hysterectomy with and without systemic extraperitoneal pelvic lymphadenectomy for cervical cancer. A report of 666 patients operated upon between the years 1957 and 1973 (author's transl)].

The report presents 850 patients with cervical cancer operated with vaginal method 1957-1973. Vaginal radical hysterectomy and systemic extraperitoneal pelvic lymphadenectomy were done in 590 patients. A 5 year survival rate in Stage I was 90.4%, in Stage II 76.5%, and in Stage III 72.2%. The 10 survival rate for 462 patients were 86.1, 69.9 and 44.4% respectively. No operative mortality has occurred to date. The incidence of lymph node metastasis in stage I was 9.5%, in Stage II 25.9% and in Stage III 34.9%. The 5 year survival rate of the patients without lymph node metastasis was 88.2% and of those with lymph node metastasis 49.5%. Radical vaginal hysterectomy without the removal of regional lymph node were done in 76 patients. The 5 year survival rate was 86.8%. There was one postoperative death in this series. Vaginal hysterectomy were done in 184 patients. The 5 year survival rate was 92.4%.

Elevated activities and properties of arylsulfatases A and B and B-variant in human lung tumors.

The activities of arylsulfatases A and B were determined in human primary and secondary tumor tissues (total, 53 cases) of various histological types. Significantly higher activities of these sulfatases were found in almost all the primary lung carcinomas as compared to their corresponding uninvolved tissues. No significant correlation was demonstrated between the enzyme activities and histological figures (stroma amounts, etc.). Lung adenocarcinoma and squamous cell carcinoma showed the presence of an additional arylsulfatase component (B1) which was not detected in normal human lung. The tumor arylsulfatase B1 had an isoelectric point (pI) of 6.7 and was clearly distinguished from arylsulfatase A (pI 4.9) and arylsulfatase B (pI 9.1 to 9.2) in normal lung and lung tumor. The tumor B1 enzyme was demonstrated to be most probably an isoenzyme of arylsulfatase B, since this unusual enzyme was indistinguishable from arylsulfatase B in terms of Ag+ inhibition; its kinetic parameters of Km for p-nitrocatechol sulfate, which was 2.9 mM with B1; optimum pH of 6.3 for B1; heat stability; and substrate specificity for three synthetic and two physiological substrates.

Determination of (E)-2,3-dichloro-4-methoxyphenyl)-2-furanylmethanone-O-[2-(diethylamino) ethyl]oxime methanesulphonate (ANP-4364) in plasma using gas chromatography with electron-capture detection.

A gas chromatographic method has been developed that permits the accurate and specific determination of the antianginal agent ANP-4364 in plasma. ANP-4364 is extracted with n-hexane containing ethyl chloroformate and, after a clean-up procedure, derivatized to the trichloroethyl carbamate, which is assayed on a gas chromatograph equipped with an electron-capture detector. Accurate determinations are possible over a concentration range from 1 to 50 ng/ml of ANP-4364 in plasma with a relative standard deviation of 7.5%. The minimum detectable concentration is 0.5 ng/ml. Plasma levels of ANP-4364 in dogs receiving oral (10 mg/kg) or intravenous (0.1 mg/kg) dosing have been determined.
