An individual who is promoted to supervisor may experience new problems. His or her acceptance in a new role by former peers may depend on attitudes and interpersonal communication skills. This paper presents attitudes and interpersonal communication skills as being two major areas which can effect a smoother transition from peer to supervisor.

A DNA ligase has been purified from a subnuclear soluble replication complex isolated from adenovirus type 2-infected human KB cells. DNA ligase activity could not be demonstrated using an exogenous template until the complex was dissociated, suggesting that the ligase activity may be a component of the complex. The purified enzyme was free of endonuclease, exonuclease, 5'-nucleotidase, and phosphatase activities, and had a molecular weight of 105 000, as estimated by sedimentation in a glycerol gradient. The ligase requires ATP and a divalent cation for activity. The optimum of the reaction is at pH 7.8 in 50--100 mM Tris-HCl buffer and 10--20 mM MgCl2. Monovalent salts greatly stimulate ligase activity and the optimum was found at 150 mM. The reaction is very sensitive to high temperature; maximum activity was observed at 25--30 degrees C. ATP is the sole required cofactor and NAD, dATP and GTP could not replace the requirement for ATP. The Km for ATP is 60 microM. The Km for DNA is 250 microgram/ml or 1.6 nmol of terminal phosphate/ml and thus the enzyme shows relatively weak affinity for exogenous DNA. The maximum conversion of 32P into a phosphatase-resistant form is approximately 1.3% of the total, whereas T4 ligase, under the same conditions, can convert more than 25% of phosphate into a resistant form.

Death in ventricular fibrillation was induced consistently in desoxycorticosterone acetate-pretreated rats by the beta-adrenergic agonist isoproterenol but not by norepinephrine or epinephrine, both of which possess alpha- as well as beta-adrenergic activity. Aminophylline, which enhances beta-adrenergic activity, and phenoxybenzamine, an alpha-receptor blocking agent, were used to study the roles of alpha- and beta-adrenergic stimulation in the production of ventricular fibrillation. With the addition of aminophylline, both norepinephrine and epinephrine produced death in ventricular fibrillation, and the existing cardiotoxicity of isoproterenol was potentiated. Similarly, in the presence of phenoxylbenzamine, doses of norepinephrine and epinephrine that had been well tolerated became lethal. Internventions that favor beta-adrenergic preponderance, either by enhancing beta-effects or by blocking protective alpha-adrenergic activation, apparently increase the arrhythmogenic propensity of norepinephrine and epinephrine in steroid-pretreated rats. The similarity of some forms of stress to the experimental protocol of chronic steroid treatment followed by acute catecholamine exposure is discussed.

The reactivity of the large and small airways of the lung in a group of ten atopic subjects with rhinitis was compared with that in ten non-atopic control subjects and in 17 with atopic asthma. Atopic state was assessed by skin prick testing with 22 common allergens and by measurement of total serum immunoglobulin E (IgE) antibody. The atopic state of the rhinitis and asthma patients was similar. Exercise provocation tests were performed on each subject and changes in airway function were measured by peak expiratory flow rate, forced expiratory volume in one second, vital capacity, maximal expiratory flow volume curves and closing volume. There were no statistically significant differences between the changes in lung function after exercise in the rhinitis patients compared with the controls. There were, however, statistically significant changes in all the measurements of respiratory function in those with asthma compared with the other two groups. The falls in forced expiratory flow rates in the asthmatic subjects were greater at lower lung volumes than near the total lung capacity, perhaps indicating that narrowing of small, as well as large, airways was occurring. No correlation was found between the magnitude of the airway response to exercise and either the total serum IgE antibody or the mean weal diameter of positive skin tests in each subject. Atopic subjects with rhinitis but not asthma showed no evidence of bronchial hyper-reactivity on exercise testing. It is therefore suggested that single exercise provocation test can discriminate subjects with atopic asthma from those with other atopic disorders.
The pH dependence of the phototoxic effects of chloropromazine (CPZ) in Escherichia coli, Salmonella typhimurium and Chinese hamster cells were studied at pH 6--8. All three biological systems displayed higher photosensitization of the drug at lower pH-values. In S. typhimurium the combined action of drug and light also showed mutagenic activity which correlated with toxicity when exposed at pH 7 or 8. When solutions of protein or DNA and CPZ were exposed to near ultraviolet (UV) light, the drug became covalently attached to the macromolecules. This binding was pH dependent but did not correlate with the effects in vivo. It was found however that the permeability of the cells to the drug was enhanced at lower pH-values. It is suggested that the enhanced entrance of CPZ at lower pH-values into the cells facilitated the drug binding to DNA, RNA and proteins within the cells upon light exposure, and that this is the basis for the enhanced phototoxicity and mutagenicity of CPZ.
We compared four instruments, [Sequential Multiple Analyzer Computer (SMAC), AutoAnalyzer II (AAII), StatLyte and PVA-4] for the determination of Na, K, Cl and CO2 using the National Committee for Clinical Laboratory Standard's (NCCLS) protocol (PSEP-1) for evaluation of precision and accuracy of automated instruments. The protocol was applied concurrently to all four instruments. The repeatability results showed SMAC to have the highest precision among-days for K, Cl, and CO2 whereas StatLyte showed the highest precision among-days for Na. The PVA-4 had the best within-run precision for Na and Cl while the AAII and SMAC had the best within-run precision for K and CO2 respectively. Systematic biases among the instruments were small for Na and K. The AAII exhibited a nearly constant bias at all concentrations of Cl relative to the other three instruments. For CO2 the SMAC showed a large proportional bias compared with the other three instruments.
The predictive values of Fourier analysis and local-feature analysis of spatial stimuli were compared in an orientation-specific adaptation experiment. Observers adapted to checkerboard patterns, which have fundamental Fourier components oriented 45 degrees away from the edges. Detection of gratings was found to be maximally impaired when fundamental Fourier components of adaptation and test patterns were in the same orientation and minimal when edges were aligned. The orientation spread and amount of adaptation effect were similar to that found in previous experiments which employed sinusoids as adaptation and test stimuli.

The physical state of the JC virus (JCV) genome was studied in two clonal cell lines (clones 2 and 7) derived from a tissue culture cell line (HJC-15) established from a hamster brain tumor induced by JCV. Saturation-hybridization and reassociation kinetic analyses, using in vitro (32)P-labeled JCV DNA, indicated that clone 7 and 2 cells contain 9 to 10 and 4 to 5 copies per cell, respectively, of all or most of the viral genome. Both cell DNAs were analyzed by using the Southern blotting procedure with three restriction endonucleases: XhoI, which does not cleave JCV DNA; EcoRI, which cleaves once; and HindIII, which cleaves three times. With each DNA, a variety of JCV-specific DNA fragments were detected. The following conclusions are possible: (i) JCV DNA is integrated into cell DNA in both clonal lines; (ii) both clonal lines contain multiple copies of the viral genome integrated in a tandem head-to-tail orientation; (iii) neither clonal line contains detectable free-form I, II, or III JCV DNA; (iv) each clonal line contains multiple independent sites of JCV DNA integration; and (v) most or all of the sites of integration on the cellular or the viral genome, or both, are different in clone 7 DNA than in clone 2 DNA. Thus, although both clone 7 and clone 2 cells were established from the HJC-15 tumor cell line, they differ in the copy number and integration pattern of JCV DNA.
Fifty-six fibreoptic bronchoscopies were performed on 42 patents with Hodgkin's disease, lymphoma, or leukaemia and pulmonary complications which did not respond to conventional antibiotics. All these patients had received chemotherapy, radiotherapy, or both for the treatment of their underlying conditions. Twenty-two bronchoscopic procedures were complicated by thrombocytopenia and neutropenia, requiring platelet transfusion before bronchoscopy, and many patients were hypoxaemic. Visual examination of the tracheobronchial tree, alveolar lavage, bronchial brushing, and transbronchial biopsy were carried out as approximate. Three patients had minor pulmonary haemorrhage, and three developed a pneumothorax after transbronchial biopsy. A specific diagnosis was obtained in 14 of 18 patients (78%) with diffuse chest radiographic abnormalities, in seven of 11 patients (64%) with lobar or segmental (focal) abnormalities, in two of eight patients with small (local) lesions, and in three of five patients with hilar abnormalities. In only three patients were infections diagnosed. It is concluded that fibreoptic bronchoscopy is a useful and safe diagnostic procedure in this situation but its value depends upon the type of radiological abnormality.
