Patterns of reversion to bobbed condition of magnified bobbed loci in D. melanogaster.

The number of genes coding for the ribosomal RNA (rDNA) can increase in D. melanogaster by means of a process called magnification. In this way, a partial deletion in this locus, termed bobbed, can reach a wild type condition. A newly magnified locus, in turn, reverts to a deficient bobbed condition if it is kept in a phenotypically wild type genotype for several generations. We studied bobbed loci at different magnification steps, analysing their behaviour through the reversion process and the way they carry out a second round of magnification. Results based on the analysis of the reversion process led to the conclusion that magnification consists of a progressive integration into the bobbed locus of free rDNA copies. Moreover, evidence is supplied that the extent of this integration affects the way a reverted locus goes through a second magnification cycle. The extensive characterization of reverted bobbed loci lends substantial support to the extra copies model of rDNA magnification.

Identification of a DNA polymerase beta-like form in Drosophila melanogaster adult flies.

DNA polymerase beta is widely distributed in the eukariotes. So far, few examples are known in which a DNA polymerase alpha -like form alone is reported. Surprisingly, DNA polymerase beta was not detected in Drosophila embryos, while it is present in the cells of multicellular species from sponge to mammals. In view of the relevance of Drosophila as a model biological system for studying the role of the various DNA metabolism enzymes in vivo we have reinvestigated the presence of the DNA polymerase beta-like form in Drosophila adult flies. Here we report the occurrence in Drosophila melanogaster adult flies of a DNA polymerase activity that, for its NEM(1) resistance, template specificity, sensitivity to ddTTP, sedimentation coefficient and nuclear localization can be classified as a beta-like form.

Inhibition of amylases from different origins by albumins from the wheat kernel.

The amylase activity of water extracts from 18 insect species, from 23 marine species and from 17 different species of birds and mammals was determined quantitatively. The inhibition of amylase in these extracts by three albumin fractions from the mature wheat kernel, which had been separated according to their molecular weights (60 000, 24 000 and 12 500 D), was determined as well. The inhibition activity of the three albumin fractions toward amylases extracted from a number of cereal species or from immature and germinating wheat kernel was also tested. The extracts from insects that are destructive of wheat grain and stored wheat products showed much higher amylase activities as compared to the other insect species that do not attack wheat and wheat products. On the basis of the effectiveness with which the three albumin fractions inhibit their activities, the amylase preparations tested were divided into susceptible, partially susceptible and resistent. Susceptible amylases, inhibited by any of the three albumin fractions, were found mainly in insects that attack wheat and in marine species. Partially susceptible amylases, inhibited by only one or two of the three albumin fractions, were present in a few avain and mammalian species including man. Resistent amylases were largely distributed in cereal, avian and mammalian species as well as in insect species that do not usually attack wheat grain or wheat flour products. At no stage of development, wheat alpha-amylase was inhibited by the albumin fractions from the mature kernel. The 12 500 dalton albumin fraction was the most effective in inhibiting insect amylases, but it was inactive toward avian and mammalian amylases. The 24 000 dalton albumin fraction was the most effective in inhibiting amylases from marine avian and mammalian species and inhibited as much as 33 amylases over 66 different amylases tested. It is suggested that protein inhibitors of amylase contributed to natural selection of polyploid wheats by giving some insect resistence to such wheats, even though some insect species were able to overcome this biochemical defense toa large degree by producing higher amylase activities.

Bone mineral metabolism in juvenile chronic arthritis: the influence of steroids.

Calciotropic hormone assays and bone mineral density (BMD) evaluations were carried out on a group of 36 children affected with juvenile chronic arthritis (JCA) (13 with pauciarticular, 8 with polyarticular and 15 with systemic onset subtype) and in a matched control group. The plasma levels of calcium, phosphate and alkaline phosphatase were normal in all the groups. Basal 25-OH D plasma levels were significantly decreased both in the polyarticular and in the systemic groups; the latter showed a further decrease after 1 year. There was a significant decrease in both the basal and the 1-year plasma levels of 1-25-OH D only in the systemic group. The BMD was decreased in the systemic group alone and the percentage increase after 1 year was significantly inferior to that in the other groups and in the controls. There was a clear relationship between the decrease in hormones and the cumulative dose of steroids administered, as well as between the BMD decrease and the physical activity of the patients (according to Steinbrocker's classification).

[Pars plana phacoemulsification. Technics, indications and results].

Pars plana phacoemulsification allows cataract extraction through a 3 mm posterior sclerotomy. The indications of these technics are the combined cataract-vitrectomy, the cataracts on vitrectomised eyes and cataracts on eyes filled with silicone oil. The alteration of corneal endothelium is very reduced with the endocapsular pars plana phacoemulsification. After the emulsification, the capsular bag is extracted with a microforceps through the pars plana. It is also possible to save the anterior capsule to reduce corneal alteration during endocular surgery. We present the indications and the advantages of these technics.
