Alpha-fetoprotein subfractions in amniotic fluid identified by a modification of the method of concanavalin A, lentil lectin or phytohemagglutinin-E affinity crossed-line immunoelectrophoresis.

Using a modified method of concanavalin A (Con A), lentil lectin (LCH) or phytohemagglutinin-E (PHA-E) affinity crossed-line immunoelectrophoresis, alpha-fetoprotein (AFP) subfractions were studied in 33 samples of human amniotic fluid obtained between 41 and 287 days of gestation. Fetal tissues (yolk sac, liver, stomach and small intestine) obtained from a fetus of 68 days' gestation were incubated for 24 hours and AFP subfractions in the culture fluid examined. AFP in control amniotic fluids yielded two subfractions (types a and b) with Con A, three subfractions (types A, B and C) with LCH, and four subfractions (types W, X, Y and Z) with PHA-E. Serial changes of AFP subfractions in the amniotic fluid, as well as in the incubation study, indicated that the yolk sac and the gastrointestinal tract were responsible for the production of the Con A non-reactive subfraction (type b), the LCH weakly-reactive subfraction (type B) and the PHA-E reactive subfraction (types W and X), at an early stage of gestation. The Con A reactive subfraction (type a), LCH reactive subfraction (type A), PHA-E weakly-reactive subfraction (type Y) and PHA-E non-reactive subfraction (type Z) were assumed to be produced mainly by yolk sac, liver or gastrointestinal tract. We also found that the LCH non-reactive subfraction (type C) was synthesized either by liver or by the gastrointestinal tract at an early stage of gestation. At term, type a of Con A, type C of LCH and types Y and Z of PHA-E were the main subfractions in amniotic fluid, assumed to be produced by the fetal liver.

Serum alpha-fetoprotein subfractions in pregnant women identified by the modified method of lectin affinity crossed-line immunoelectrophoresis.

Using a modified method of lectin affinity crossed-line immunoelectrophoresis, we studied the alpha-fetoprotein (AFP) subfractions in sera from 12 pregnant Japanese women, at 22 and 42 weeks of gestation. The method involves the first dimension electrophoresis in agarose gel containing concanavalin A (Con A) or lentil lectin (LCH), the second dimension immunoelectrophoresis in agarose gel containing polyclonal antibody against AFP, reaction with peroxidase-conjugated Protein A and staining with 4-methoxy-1-naphthol. We found that type a of Con A or type C of LCH was the only subfraction present in maternal circulation at the second or third trimester. These AFP subfractions were assumed to be of fetal liver-origin. The minimum concentration which yielded an immunoprecipitation peak was approximately 100 ng/ml, being twenty times more sensitive than the conventional lectin affinity crossed-line immunoelectrophoresis.

[Antitumor activities of orally administered 4-carbamoylimidazolium-5-olate (SM-108)].

We investigated the antitumor effect of orally administered SM-108. The drug showed strong antitumor activities against Ehrlich carcinoma, sarcoma 180, P388 leukemia, L1210 leukemia, colon 26 adenocarcinoma, colon 38 adenocarcinoma, and Lewis lung carcinoma. The antitumor activity of SM-108 against Ehrlich carcinoma was so remarkable, in fact, that all mice in a group survived for a long time. The antitumor effect of SM-108 depends on its administration schedule. Treatment involving a schedule of one dose every 6 h for 24 h at intervals of 3 days brought about a much stronger effect than daily single treatment. The maintenance of a high serum level of SM-108 for 24 h by the former treatment is responsible for the strong therapeutic effects, because the action of SM-108 is time-dependent. The antitumor activities of SM-108 administered orally are excellent enough to be comparable with those obtained by intraperitoneal administration as previously reported.

CT findings in patients with gliomas, surviving more than 10 years.

After reviewing the latest CT scans of 23 mostly low grade glioma patients who survived more than 10 years after treatment, it was suggested that loss of the original tumour bulk and frequently the presence of porencephalic cavity replacing the original tumour, with a smooth lining which is not contrast-enhanced is essential to assume a clinical "cure" in the case of intrinsic cerebral gliomas. In the case of gliomas growing within the ventricle or subarachnoid space, tissue with calcium deposit but without contrast-enhancement was noted, although the number of cases was limited.

[Ophthalmoscopic observations of ocular fundus in colony-born cynomolgus monkeys aged from 0 day to 90 days].

Apparently healthy 242 colony-born cynomolgus monkeys (Macaca fascicularis) aged from 0 day to 90 days were examined for the findings of ocular fundus by using ophthalmoscope. One drop of mixed solution of 0.5% tropicamide and 0.5% phenylephrine hydrochloride was instilled into each eye of the monkey. Then, those monkeys were anesthetized with ketamine-HC1 at the dose level of 10mg/kg B. W. Regular and fluorescein photographs were taken with Kowa RC-II ophthalmoscope-camera by using daylight typed color film. Following findings were obtained in each age class: Retinal color was salmon pink with 0 to 3-days-old neonates, salmon pink and blue to green with 7 days to 14-days-old animals and blue to green with 60-days to 90-days-old monkeys. As regards optic disc, 0- to 14-days-old animals were observed to be light orange in color, and the infant aged more than 28-days showed orange color. Retinal arteries and veins were lightly reddish in color with every age class. Macular color was salmon pink in 0-day-old cases, slightly dark in 3-days-old neonates and very dark after 14-days of age. Lightly retinal reflex was noted in 0- and 7-days-old animals. The reflex was observable in 14-days-old animals without any case of exception. Retinal hemorrhages were recorded in 22 (67%) of 36 neonates born in natural condition and 10 (33%) of 30 neonates born by cesarean section. These findings will be useful as the criteria for ophthalmoscopic observations of the cynomolgus monkeys as laboratory use.

[Persistence and disappearance of hyaloid artery in newborn cynomolgus monkeys].

Apparently healthy 45 newborn cynomolgus monkeys (Macaca fascicularis) were examined for the persistence and disappearance of the hyaloid artery by using an ophthalmoscope. On the day of birth, the hyaloid artery was observed in all the newborn animals. Between 21 and 45 days of age, the hyaloid artery faded without exception. The persistence of the hyaloid artery in the newborns and its disappearance during an early period of life can be regarded as a normal phenomenon in the developmental process of infant cynomolgus monkeys.

Reversal of multidrug resistance by non-antitumor anthracycline analogs.

It was found that three synthetic anthracycline analogs lacking not only antitumor activity but also calcium-antagonizing action possessed an activity to potentiate vincristine cytotoxicity against vincristine-resistant P388 leukemia. ID-8279, one of these analogs, significantly reversed resistance to vincristine and daunorubicin by increasing their intracellular accumulation.

Interferon effects on multiplication, cytoplasmic protein and GFAP content, and morphology in human glioma cells.

Beta-type human fibroblast interferon inhibited the multiplication of human glioma cells in a dose-dependent manner. The inhibitory effects were thought to be due to suppression of the cells entering the S-phase. After interferon treatment with 5 X 10(3) IU/ml for ten days, the mean cell volume, the soluble protein, and the glial fibrillary acidic protein increased to 970%, 190%, and 380% that of the controls, respectively. The interferon-treated cells changed shape and resembled mature astrocytes. It is presumed that beta-type human fibroblast interferon inhibits DNA synthesis of cultured glial cells and subsequently induces specific differentiated proteins and morphologic alteration.

Supernumerary lactate dehydrogenase isozymes in human gliomas.

Supernumerary bands in agarose gel electrophoresis of lactate dehydrogenase (LDH) were frequently observed in extracts of human gliomas. The supernumerary fractions which migrated cathodic to LDH-2 and/or cathodic to LDH-3 were designated LDH-2' and LDH-3'. These extra bands were clearly seen in certain gliomas and less distinctly in others, being more frequent in primitive or undifferentiated tumours. The extra bands seen in gliomas differ from the LDH-X of the testis, but LDH-2' seemed correspond to LDH-Z in placenta, hydatidiform mole, and choriocarcinoma. These sub-bands are interpreted as being produced by gliomas and as oncofetal enzymes.

Cavernous hemangioma in the pineal region.

A cavernous hemangioma in the pineal region of a 22-year-old man is reported. Because the patient had diabetes insipidus and a contrast-medium-enhanced mass in his pineal region on computed tomography, a germinoma was suspected. A ventricular shunt was made and the tumor was irradiated, but without a response. After irradiation the tumor was entirely removed, and postoperatively the diabetes insipidus improved.

Metabolism of dibucaine: isolation and identification of urinary basic metabolites in the rat, rabbit and man.

The metabolism of dibucaine was studied in the rat, rabbit and man. A total of ten basic metabolites other than dibucaine were detected in the urine samples of three species by thin-layer chromatography (TLC) and gas chromatography (GC), and structures of these metabolites were identified by comparison of the properties given by TLC, GC and gas chromatography-mass spectrometry (GC-MS) with those of authentic compounds. Four of these metabolites were new metabolites which were found in the rabbit or human urine; two were identified as the 2', 3'-dihydroxybutoxy product (M-6, diol) and its N-deethyl product (M-2), and others were identified as the 2'-hydroxyethoxy product (M-8, alcohol) and its N-deethyl product (M-3). One of the two hydroxyl groups on M-5 was at 6-position on the quinoline ring, while another was assumed to be at 3'-position on the O-alkyl side chain. There were apparent species differences with regard to the major metabolites found in each species; i.e. M-10 and M-5 in rat, M-6 and M-4 in rabbit, and M-8 and M-4 in man. Small amounts of the conjugated basic metabolites were observed in the urine of all three species. The new metabolic pathways to the diols (M-2 and M-6) or the alcohols (M-3 and M-8) were also discussed.
