Ehrlich carcinoma and P388 leukemia cells were rendered resistant to 4-carbamoylimidazolium 5-olate (SM-108), and assessments were made of biochemical and pharmacological determinants for the sensitivity to SM-108 using both sensitive and resistant sublines. We observed that the treatment of cells with SM-108 in vitro caused a remarkable decrease in the intracellular guanosine 5'-triphosphate pool level in sensitive but not in resistant sublines. There was no difference in the ability to take up SM-108 between sensitive and resistant sublines, but the cellular conversion of SM-108 to its nucleotide, which is the putative active anabolite of SM-108, proceeded only in sensitive sublines. Enzymological studies revealed that the activity of adenine phosphoribosyltransferase (EC 2.4.2.7), which is believed to conjugate SM-108 with 5-phospho-alpha-D-ribose 1-diphosphate, was very low in the resistant sublines. These results strongly support our previous hypothesis that SM-108 is activated by adenine phosphoribosyltransferase to SM-108-nucleotide which then inhibits hypoxanthine-5'-monophosphate dehydrogenase (EC 1.2.1.14), a key enzyme for the de novo synthesis of guanosine 5'-monophosphate.

A case is reported of an infant with a giant cerebellar vermis hemangioma which was totally removed. The excised specimen contained both cavernous angioma-like and telangiectasia-like portions with intervening cerebellar tissue. The tumor was considered to be a mixture of cavernous hemangioma and telangiectasia.
The supraclinoid segments of the internal carotid artery (ICA) and their surrounding structures were examined under magnification in 25 adult cadavers. Attention was paid to anatomical variations and relationships concerning ipsilateral and contralateral pterional microsurgical approaches to these regions, especially to the origin of the ophthalmic artery. Eighty-four percent of the ophthalmic arteries arose from the supraclinoid segment of the ICA. In the ipsilateral pterional approach, mobilization of the ipsilateral optic nerve was required to see the origin of the ipsilateral ophthalmic artery and the medial aspect of the proximal portion of the supraclinoid segment of the ICA. In the contralateral pterional approach, on the other hand, these areas on the contralateral side could be identified under the optic nerve with minimal or without retraction of the contralateral optic nerve. This was because 71% of the ophthalmic arteries arose from the supero-medial aspect of the ICA, and because there was nothing to intercept the view of the medial aspect of the ICA under the optic nerve. This study supports the usefulness of the contralateral pterional approach to the origin of the ophthalmic artery and the medial aspect of the supraclinoid segment of the ICA. This approach could be useful in certain cases of carotid-ophthalmic aneurysm. The authors' experience with the contralateral pterional approach to carotid-ophthalmic aneurysms is also described.
A case of so-called intraspinal neurenteric cyst is described. A single epithelial cyst, associated with a dimple in the sacral region, was located ventral to the spinal cord in the lower cervical region. On light microscopy, a single epithelial layer lining the cyst wall contained a few ciliated cells and squamous cells, in addition to many goblet cells. An electron microscopic study also demonstrated three different kind of cells. The goblet cells contained many secretory granules, the ciliated cells had many cilia, and the squamous cells were characterized by abundant tonofilaments and desmosomes.
A mixed colloid cyst-xanthogranuloma of the third ventricle is described. The xanthogranuloma associated with the cyst completely occupied the third ventricle. On light microscopy, a single epithelial layer with a basal lamina was seen lining the cyst wall. Electron microscopic study revealed three different kinds of cells in the epithelial layer: ciliated, nonciliated, and basal cells. Many lipid droplets were present in the superficial epithelial cells, macrophages, pericytes, and endothelia. The pathogeneses of the cyst and of the xanthogranuloma are discussed.
Differences in clinical features of arteriovenous malformations (AVM) in six children with ruptured AVM (RAM) and three with unruptured AVM (URAM) are herein reported. The age at onset ranged from the neonatal period to 8 yrs in cases of URAM and 10 to 12 yrs in those with RAM. The children with RAM had a sudden onset of intracranial hemorrhage, except for one with prodromal symptoms such as vertigo or headache. The clinical features of URAM were chronic mild symptoms such as a certain degree of delayed development, focal neurologic signs or increased intracranial pressure in addition to macrocephalus and cranial bruit. CT scan revealed an evenly enhanced AVM and dilatation of ventricles in cases of URAM. The RAMs were angiographically relatively small while the URAMs were large.
High molecular weight DNAs prepared from a variety of human tumors maintained in nude mice were assayed for their ability to transform NIH 3T3 cells. DNAs from 4 of 21 tumors tested induced transformed foci in cultures of NIH 3T3 cells. They were from a Ewing sarcoma line, a glioblastoma line, a leiomyosarcoma line, and a lung carcinoma line. Hybridization analyses of the NIH 3T3 transformant DNAs with a human repetitive sequence as probe revealed that four distinct transforming DNA sequences were transferred to NIH 3T3 cells from the four tumor lines. The transforming DNA in a lung carcinoma line was a human homologue of the oncogene of Kirsten murine sarcoma virus (Ki-ras). On the other hand, the three other transforming DNAs showed no similarity to any known human transforming gene detected by the NIH 3T3 transformation assay. Further analyses with a series of cloned oncogenes as probes revealed that the transforming DNA in a glioblastoma line was a human homologue of the oncogene of 3611-murine sarcoma virus (raf). However, the two transforming DNAs in a Ewing sarcoma line and a leiomyosarcoma line had no sequence homology to any of the cloned oncogenes.
A case of accessory nerve neurinoma presenting with a so-called "intraaxial mass sign" on the angiograms is reported. The tumor originated from the intracranial portion of the accessory nerve and displaced the lateral medullary segment of the posterior inferior cerebellar artery posterolaterally. The displacement of the artery can be understood by the microanatomical relations.

Three thousand and five apparently healthy cynomolgus monkeys (Macaca fascicularis) were examined for the finding of the anterior part of the oculi by using an ophthalmoscope. One drop of the mixed solution of 0.5% tropicamide and 0.5% phenylephrine hydrochloride was instilled into each eye of the animal. Then, those monkeys were anesthetized with ketamine-HC1 at the dose level of 10 mg/kg B. W.. One monkey had opaque membranes of tan to brown color, extending from some part of collarette of the iris to the other part of collarette like the network over the pupil. This finding was diagnosed to be bilateral persistent pupillary membranes. Further breeding studies will be carried to determine if this case in hereditary.
The ocular fundi of 1,151 apparently healthy colony-born cynomolgus monkeys (Macaca fascicularis) ranging in age from newborn to 19 years were examined using an ophthalmoscope. Two hundred and thirty-eight abnormal findings were recorded in 219 of the 1,151 monkeys. Of these, 23 were related to the optic disc and 115 to the retinal vessels. Of the remaining 100 abnormal findings, 91 were retinal degenerations and 9 were retinal hemorrhages. The 23 optic disc abnormalities consisted of 3 cases of micropapilla, 4 of ectasia and 16 of myelination of the retinal nerve fibers. Of the 115 retinal vascular abnormalities, 87 were arterial tortuosity, one was venous tortuosity, 2 were tortuosity of both artery and vein, 2 were artery-vein crossing, 20 were copper-wire artery, one was inosculation of the artery, one was vascularization of the vein and one was persistent hyaloid artery. Of the 91 retinal degenerations, one was degeneration of the periphery of the macular and the optic disc, 8 were macular degeneration and 82 were peripheral degeneration. Nine cases of retinal hemorrhages appeared under 6 years of age.
Serologic survey and virus isolation of feline herpesvirus (FHV) and feline calicivirus (FCV) were performed on cats used for research at the Laboratory Animal Research Centre, The University of Tsukuba, over the period from 1978 to 1981. Of the 507 mature and immature cats, 4 months old or older, 102 (20.1%) had HI antibody against FHV and 412 (81.3%) SN antibody against FCV. Some 23 (16.2%) and 76 (53.5%) kittens among 142 younger than 4 months had antibodies against FHV and FCV, respectively. Both the antibodies in kittens were considered to be maternally derived. The FCV antibody rate was especially high in cats weighing 2.5 kg (males) and 2.0 kg (females) or more, which were regarded as 8 to 10 months of age. An attempt was made to isolate the viruses from the oropharynx and conjunctiva of clinically healthy mature or immature cats and kittens. As the result, either one or both of the viruses were isolated from 31 of 75 mature and immature cats, and isolation rates of FHV and FCV were 6.7% and 36.0%, respectively. On the other hand, no virus was detectable in 16 kittens.
Tri-combinant vaccine consisting of attenuated feline herpesvirus (FHV) and feline calicivirus (FCV) and inactivated feline panleukopenia virus (FPLV), were evaluated for safety and efficacy, using Japanese native cats and the viral strains isolated in Japan. Thirty-eight 9- to 12-week-old kittens were inoculated intramuscularly and subcutaneously with the vaccine. Consequently, no adverse reaction was found, and protective efficacy was confirmed by challenge tests with the virulent strains of each virus. Serum-neutralizing antibodies against FCV and FPLV were maintained for at least one year after vaccination, whereas antibody against FHV disappeared in two cases at 24 weeks after vaccination. Application of this vaccine seemed effective for control of feline viral disease in cats for experimental use.

Using a modified method of Con A, LCH or PHA-E affinity crossed-line immunoelectrophoresis, we studied AFP subfractions in 78 sera including 58 from patients with primary hepatoma, 11 from patients with hepatic metastasis of gastric cancer and 9 from patients with germ cell tumors of the gonads (yolk sac tumor, immature solid teratoma or mature solid teratoma). It was found that AFP in primary hepatoma, metastatic hepatoma or germ cell tumors of the gonads were differently glycosylated, and different patterns of AFP subfractions identified by Con A, LCH or PHA-E affinity crossed-line immunoelectrophoresis facilitated a differential diagnosis of such AFP related malignancies.
