Electron microscopic observation on the epithelial-connective tissue junction of pemphigus vulgaris. A case report.

An electron microscopic observation on the erosive buccal mucosa of a buccal mucosa of a patient with typical pemphigus vulgaris was carried out. The ultrastructural features of the basal cells and the basal lamina in the advanced oral lesion varied from section to section of the biopsy specimen. The basal lamina showed various morphologic alterations indicating the presence of both destructive and regenerative processes in different sections. The filamentous bodies were also observed both in the basal cell layer and in the upper lamina propria.

[A case of clear cell carcinoma of the minor salivary gland].

A rare case of clear cell carcinoma of the palatal salivary gland in a 69-year-old man is reported. Histopathologically, nests and cords of tumor cells, which showed predominantly clear cytoplasms, and occasionally eosinophilic cytoplasms, proliferated on the overlying epithelium and capsules of the fibrous connective tissue. The clear cell cytoplasms were found to be PAS positive, diastase soluble, and Alcian-blue negative, indicating the presence of glycogen. Ultrastructurally, the tumor cells were found to contain glycogen and cytoplasmic filaments in various proportions. These observations suggest that clear cell carcinoma is myoepithelial in origin.

[Prognostic factors in nonresected adenocarcinoma of the lung].

We have studied the prognostic factors of 117 cases of unresected adenocarcinoma of the lung, and have examined these cases according to the following categories: age, sex, clinical stage, performance status (P.S.), serum albumin on admission, serum CEA, the presence of complication, the present status of an abnormal chest shadow from a year ago at the same location, and differentiation of cells. Significant differences were detected in the P.S., the serum albumin and the presence of complications by survival analysis through the use of covariate-Cox models. We also have noted that the difference between stage I and the other stages of the clinical stage was found to be significant by a generalized Wilcoxon test. In conclusion, the determining prognostic factors of an unresected adenocarcinoma of the lung are the P.S., the serum albumin, the presence of complications and the clinical stage.

Interaction between human albumin polymers and the envelope polypeptide of hepatitis B virus (P31) containing the translation product of the pre-S2 region.

Two preparations of human serum albumin (Cohn fraction V), when subjected to high-performance liquid chromatography, were found to contain albumin polymers with molecular sizes of 35 X 10(4) daltons or greater (breakthrough fraction), as well as tetramers, trimers and dimers. They bound to the envelope polypeptide of hepatitis B virus composed of translation products of the pre-S2 region and the S gene (P31), carrying the receptor for polymerized human serum albumin, with an activity increasing in parallel with the degree polymerization.

[Bilateral renal cell carcinoma associated with von Hippel-Lindau disease].

A 52-year-old man was referred to the department of neurosurgery of our hospital for evaluation of left occipital pain on September 27, 1982. Brain CT and arteriography showed midcerebellar tumor and an operation was performed on October 7, 1982. Histopathologically, the tumor was hemangioblastoma (solid type). His whole-body CT showed bilateral multiple renal tumors but no angioma retinae were found. Angiography revealed that the lesions were bilateral multiple (more than 20) renal tumors, bilateral adrenal tumors and left retroaortic renal vein. The patient underwent bilateral radical nephrectomy and lymphadenectomy on December 15, 1982. Renal cell carcinoma (grade II greater than III) with adrenal and left renal vein involvement were noted on the pathologic specimen. Postoperatively, he received supplement therapy with hydrocortisone and hemodialysis, but died of bleeding from the recurrent hemangioblastoma on July 7, 1983.

[Results of pulmonary resection for metastatic renal cell carcinoma].

Nine cases of metastatic renal cell carcinoma to the lung were studied to assess the efficacy of surgical management. Between January, 1965 and December 1981, 116 cases of metastatic renal cell carcinoma to the lung were treated. Nine of these cases (7.8%), were treated with surgical resection for the pulmonary metastases. The overall crude survival rate after pulmonary resection was 33.3% (3/9) at 3 years and 22.2% (2/9) at 5 years. Two patients are long-term survivors, one still being in good health 108 months, and the other 72 months after pulmonary resection. Pulmonary resection for metastatic renal cell carcinoma was considered effective in some selected slow-growing cases as protection against metastasis from a metastasis.

Asn-linked oligosaccharides in lectin-resistant tumor-cell mutants with varying metastatic potential.

MDW4, a wheat germ agglutinin-resistant mutant of the metastatic murine tumor MDAY D2 has previously been shown to be poorly metastatic when injected intravenously and non-metastatic when injected subcutaneously into syngeneic mice. W4EB8, a Bandeiraea simplicifolia (BSII) lectin-selected subline of MDW4 has previously been shown to be intermediate between that of MDAY-D2 and MDW4 cell for sensitivity to lectin and metastatic phenotype when injected intravenously into mice. The Asn-linked oligosaccharides from MDAY-D2, MDW4 and W4EB8 cells were released enzymatically with peptide N-glycosidase, reduced with tritiated sodium borohydride and fractionated by Concanavalin-A--Sepharose affinity chromatography and high-performance liquid chromatography (HPLC). Structures of the major fractions were determined by a combination of glycosidase digestion and sizing, gas-liquid chromatography/mass spectrometry and by proton nuclear magnetic resonance. Wild-type and mutant cells processed high-mannose-type structures to biantennary (GlcNAc)2(Man)3(GlcNAc)2. In MDAY-D2 cells this structure was processed further to sialylated tetra-antennary complex with polylactosamine-containing antennae terminating in either sialic acid or alpha 1-3-linked galactose. MDW4 cells had four or five times more (GlcNAc)2(Man)3(GlcNAc)2 than MDAY-D2 cells and a major component of tri-antennary (GlcNAc)3(Man)3(GlcNAc)2 (i.e. 2,2,6-substituted tri-mannosyl core) that was not found in wild-type cells. The partial revertant, W4EB8 had intermediate levels of mutant (GlcNAc)3(Man)3(GlcNAc)2 and sialylated complex-type carbohydrates. The results indicate that a shift in expression from incomplete complex type to sialylated tri/tetra-antennary complex-type carbohydrates in tumor cell may enhance the metastatic potential of tumor cells in the experimental metastasis assay. In addition, somatic cell hybridization analysis indicated that the defect in MDW4 cells was identical to that of the Chinese hamster ovary mutant Lec8: a deficiency in UDP-galactose transport into the golgi.

Isolation and characterization of leukosialin, a major sialoglycoprotein on human leukocytes.

A major sialoglycoprotein (previously called gp105) on the human erythroleukemic cell line K562 was purified, and specific antibodies were raised in a rabbit. A number of different hematopoietic cell lines belonging to erythroid, myeloid, T-lymphoid, and B-lymphoid cell lineages were found to possess glycoproteins that were immunoprecipitated by these antibodies. However, the apparent molecular weights differed between cell lines, ranging from 113,000 to 150,000. In almost all cases, the immune precipitated molecule corresponded to the major sialoglycoprotein of the respective cell. Pulse-chase experiments showed that all cells produced an early precursor form of the molecule of 54 kDa, which was susceptible to endo-beta-N-acetylglucosaminidase H to give an apoprotein of 52 kDa. Neuraminidase treatment of the mature forms resulted in a characteristic decrease of the mobility in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (apparent molecular weights from 150,000 to 183,000). Amino acid analysis of the glycoprotein isolated from HL-60 cells showed a high content of serine, threonine, and proline, and the carbohydrate composition was compatible with the presence of a large number (approximately 90) of O-linked carbohydrate chains. The name leukosialin is proposed for this sialoglycoprotein, which seems to be widely distributed, but differently glycosylated, on leukocytes with diverse functions. In the following paper (Carlsson, S.R., Sasaki, H., and Fukuda, M. (1986) J. Biol. Chem. 261, 12787-12795), we demonstrate that the structures of O-linked oligosaccharides vary significantly depending on the cells from which leukosialin was isolated.

Structural variations of O-linked oligosaccharides present in leukosialin isolated from erythroid, myeloid, and T-lymphoid cell lines.

Structures of O-linked oligosaccharides of leukosialin isolated from K562 erythroid, HL-60 promyelocytic, and HSB-2 T-lymphoid cell lines were examined. Leukosialin was isolated by specific immunoprecipitation from cells which were metabolically labeled with [3H]glucosamine, and glycopeptides were isolated after Pronase digestion. O-Linked oligosaccharides were released by alkaline borohydride treatment, and the structures of purified oligosaccharides were elucidated by specific exoglycosidase digestion, Smith degradation, and methylation anaylsis. Oligosaccharides from K562 cells were found to be GalNAcOH, Gal beta 1----3GalNAcOH, NeuNAc alpha 2----6GalNAcOH, NeuNAc alpha 2----3Gal beta 1----3GalNAcOH, Gal beta 1----3(NeuNAc alpha 2----6)GalNAcOH, and NeuNAc alpha 2----3Gal beta 1----3(NeuNAc alpha 2----6)GalNAcOH. On the other hand, oligosaccharides from HL-60 and HSB-2 cells were found to be NeuNAc alpha 2----3Gal beta 1----3GalNAcOH, NeuNAc alpha 2----3Gal beta 1----4GlcNAc beta 1----6(Gal beta 1----3)GalNAcOH, Gal beta 1----4GlcNAc beta 1----6(NeuNAc alpha 2----3)Gal beta 1----3)GalNAcOH, and NeuNAc alpha 2----3Gal beta 1----4GlcNAc beta 1----6(NeuNAc alpha 2----3Gal beta 1----3)GalNAcOH. These results clearly indicate that leukosialin can be differently glycosylated with O-linked chains, and each erythroid or myeloid (and T-lymphoid) cell line expresses a characteristic set of O-linked oligosaccharides which differ in core structures as well as in sialylation.

Structures of O-linked oligosaccharides isolated from normal granulocytes, chronic myelogenous leukemia cells, and acute myelogenous leukemia cells.

O-Linked oligosaccharides were isolated from normal granulocytes, chronic myelogenous leukemia cells, and acute myelogenous leukemia cells by alkaline borohydride treatment. Oligosaccharides were fractionated by Sephadex G-50 gel filtration and QAE-Sephadex column chromatography, and their structures were elucidated by fast atom bombardment-mass spectrometry after permethylation and methylation analysis before and after specific exoglycosidase treatments. Results show that normal granulocytes and chronic myelogenous leukemia cells contain a series of O-linked oligosaccharides with the following structure, (formula: see text) where, in normal granulocytes n = 0 is major and n = 1 or 2, and thus polylactosaminyl oligosaccharides are present as minor components. However, these polylactosaminyl oligosaccharides were barely detectable in chronic myelogenous leukemia cells. On the other hand, acute myelogenous leukemia cells, which represent poorly differentiated myeloid cells, mainly contain short O-linked oligosaccharides with 2----6-linked sialic acid as follows. (formula: see text) These results suggest that structures of O-linked oligosaccharides vary in the different maturation stages along the same cell lineage.

Human glycophorin A and B are encoded by separate, single copy genes coordinately regulated by a tumor-promoting phorbol ester.

Human glycophorin A belongs to a family of structurally related cell surface glycoproteins which are expressed in erythroid cells. We have recently isolated several human glycophorin A-specific cDNA clones and have determined their nucleotide sequence (Siebert, P. D., and Fukuda, M. (1986) Proc. Natl. Acad. Sci. U. S. A. 83, 1665-1669). In this report by using cDNA and exact sequence synthetic oligodeoxyribonucleotides as hybridization probes, we provide evidence that human glycophorin A and B are encoded by separate and distinct genes present as single copies in the human genome. Furthermore, we show that the expression of the glycophorin A and B genes are coordinately regulated by tumor-promoting phorbol ester, 12-O-tetradecanoylphorbol-13-acetate.

Ultrastructural evidence of enkephalinergic input to glucoreceptor neurons in ventromedial hypothalamic nucleus.

A combination of intracellular injection of horseradish peroxidase into the ventromedial hypothalamic nucleus glucoreceptor neurons and immunohistochemical staining for enkephalin allowed ultrastructural visualization of interrelations between these electrophysiologically identified neurons and enkephalin-like immunoreactive (ENK-IR) terminals. The results show that glucoreceptor neurons receive direct inputs from ENK-IR neurons. This is the first evidence that glucoreceptor neurons, identified electrophysiologically, can be affected monosynaptically by endogenous enkephalin, probably released from ENK-IR terminals.

Visual responses related to food discrimination in monkey lateral hypothalamus during operant feeding behavior.

Single neuron activity was recorded from monkey lateral hypothalamic area (LHA) to relate neuronal events to food discrimination and initiation of procurement movement in operant bar press feeding behavior. Of 429 neurons tested, 68 (16%) responded during visual phase. Of these, 30 (7%) responded selectively to the sight of food or non-food associated with a juice reward, but not to the sight of meaningless non-food or food associated with aversive saline. Neuronal activity related to discrimination was readily influenced by extinction, reversal or satiation. The strength of visual responses was correlated with latency of bar press initiation and speed of bar pressing, but was not related directly to bar press movement. These suggest that the LHA is deeply involved in discrimination of reinforcement or non-reinforcement, and might be associated with higher functions to regulate internal states such as physiological need to get food during operant feeding behavior.

Demonstration of high molecular weight forms of inhibin in bovine follicular fluid (bFF) by using monoclonal antibodies to bFF 32K inhibin.

Monoclonal antibodies specifically recognizing each 20K and 13K subunit of bovine follicular fluid (bFF) 32K inhibin have been prepared. By immunoblotting procedures using these antibodies, we have demonstrated in bFF at least six inhibin forms, the apparent molecular weights of which are estimated to be 120K, 108K, 88K, 65K, 55K and 32K daltons, respectively. Amongst them 65K, 55K and 32K inhibin forms comprise two polypeptide subunits linked by disulfide bridge(s). In these inhibin forms a polypeptide of 13K daltons, a shorter subunit of bFF 32K inhibin, is attached by disulfide linkage(s) to polypeptides of 57K, 44K and 20K daltons, which are immunologically related to a larger subunit of the 32K inhibin, to yield 65K, 55K and 32K inhibins, respectively. On the other hand the higher molecular weight forms, 120K, 108K and 88K inhibins, are composed of three polypeptide subunits. In these forms a polypeptide of 62K daltons, immunologically related to the shorter subunit of bFF 32K inhibin, is attached by disulfide bridge(s) as the third component to the respective smaller inhibin forms which are composed of two subunits. These findings suggest that the complex formation of the subunit precursors may be extremely important and that restricted proteolytic cleavages following the complex formation may yield such divergent forms of inhibin in bFF.
