Hemorheological factors in the pathophysiology of acute and chronic cerebrovascular disease.

The hemorheologic changes in three groups of patients suffering from acute and chronic cerebrovascular diseases were studied. Firstly, a horizontal study on 57 patients with definite stroke and on 49 patients with TIA was made. Plasma viscosity, whole blood filtration rate, fibrinogen concentration and hematocrit were evaluated as markers of the rheological property of blood. Blood samples were drawn within 6 h from the onset of vascular syndrome. The findings were compared with values obtained in 112 as controls. At the same time, washed red cell filtration rate, together with lactoferrin, betaglucuronidase and beta-thromboglobulin plasma level were assayed. In both groups the onset of the vascular storm was associated with a marked increase of plasma fibrinogen and of blood and plasma viscosity and a significant decrease of whole blood filterability. Lactoferrin, betaglucuronidase and beta-thromboglobulin levels were also significantly increased. Following this, a longitudinal study was performed on 27 patients with definite stroke and 32 patients with TIA. The clinical regression of acute stroke was associated with the progressive reduction of rheological abnormalities. Finally, 81 patients with clinical diagnosis of cerebrovascular disease due to previous stroke or repeated TIA were studied together. An increase of blood viscosity, of fibrinogen concentration and of hematocrit and a decrease of blood filtration rate together with higher levels of beta-thromboglobulin were registered. These results confirm the existence of an association between CVD and hemorheological alterations and suggest more in depth research directed towards identifying the significance of these alterations in the pathogenesis of tissue ischemia.(ABSTRACT TRUNCATED AT 250 WORDS)

Uptake of tritiated phosphatidylcholine by isolated rat liver nuclei studied by electron microscope autoradiography in albumin embedded specimens.

The transfer of phosphatidylcholine from multilamellar vesicles to isolated rat liver nuclei was studied by means of electron microscope autoradiography. To avoid the possible loss or the artifactual redistribution of the phospholipid occurring during dehydration with organic solvents and plastic embedding, the fixed specimens were embedded in aqueous albumin, which was then hardened by glutaraldehyde and dehydrated physically. The quantitative analyses of the autoradiograms demonstrated that part of the labelled phosphatidylcholine was taken up by the isolated nuclei and was transferred inside the nucleoplasm. The uptake corresponded to the loss of the vesicular arrangement, probably owing to the formation of a lipoprotein complex with the nuclear proteins. The results provide evidence that the lipid-induced changes of transcriptional activity occur upon the actual interaction of the exogenous phospholipid with the inner nuclear components.

Pharmacokinetics and distribution of a fluoresceinated glycosaminoglycan, sulodexide, in rats. Part II: Organ distribution in rats.

Fluorescein-labelled sulodexide tissue distribution was studied in the rat after intravenous administration at the dosage of 15 mg/kg. A globular fluorescence was observed mainly in the extracellular matrix and a diffuse one was present in liver and kidney parenchyma lasting for a long time, up to 4 h after administration. In particular in the kidney cortex the fluorescent material was present at the higher time intervals. Fluorescence was also observed in the spleen and in the perivascular areas and in the media of the aortic wall.

Immunodiagnosis in gynaecologic oncology: possibilities and limitations.

The theory of Immunological Surveillance suggested by Burnet has been the object of many criticisms over the last years, but the study and the clinical applications of tumours immunology still show a remarkable interest with attracting prospects for the future. Some markers for human tumours cells, detected by RIA assay (TA4 for cervical cancer, CEA for ovarian cancer and alpha-FP for Endodermal Sinus Tumour) allow accuracy in neoplastic patients. The study of the tumoural patient's immunologic status could point out diagnostic and prognostic criteria and is likely to be used in the future for screening groups at risk for developing neoplasia.

[Relation between the distribution and orientation of collagen fibers and the inorganic phase of calcification processes in the anterior leaflet of the human mitral valve].

The collagen fibers observed in calcified valves appear randomly distributed without any preferential arrangement. In these areas also the hydroxylapatite crystals do not show any orientation. It is suggested that the disorder of the organic phase is a factor favouring the deposition of the calcium crystals.

Arrangement of microfibrils in collagen fibrils of tendons in the rat tail. Ultrastructural and x-ray diffraction investigation.

The microfibrillar arrangement in collagen fibrils of tendons in the tail of the rat was examined by electron microscopy and X-ray diffraction. Fresh and air-dried collagen fibers were examined in unstretched and stretched conditions. The results demonstrate that the microfibrils have a course parallel to the longitudinal axis of the collagen fibrils. The influence of stretching and hydration of the samples on the orientation of fibrils and microfibrils is also assessed.

T-lymphocyte subpopulations in pregnancy.

T-lymphocyte subpopulations were evaluated in 44 pregnant women at different stages of gestation by means of monoclonal antibodies. A total of 32 healthy women of the same age served as a control group. OKT3-positive cells (more than 95% of the circulating T-lymphocytes) decrease markedly in pregnancy between the 14th and the 28th week. Such a decrease seems to be due to a specific lowering in the number of OKT4-positive cells (which seem to represent helper T-lymphocytes). OKT8-positive cells (which seem to represent cytotoxic-suppressor T-lymphocytes) show no meaningful alteration. Possibly due to helper decrease, maternal immunity is impaired between the 14th and 28th week.

T-lymphocyte subsets in normal menstrual cycle.

OKT3, OKT4, OKT8 positive cells levels were evaluated in 27 healthy volunteers, during preovulatory phase, luteal phase and menstrual period. OKT3, OKT4 positive cells showed no statistical difference over the three periods, while OKT8 positive cells decrease in preovulatory phase compared to both luteal phase (p less than 0.05) and menstrual period (p less than 0.02). A possible immunoregulatory function of oestrogens is discussed.

[The relation between erythrocyte filterability and platelet aggregation (in vitro studies in normal subjects and those with vascular disease before and after isotonic exercise)].

The aim of this study was to evaluate the possible action in vitro of the platelets on the rheological behavior of red blood cells in healthy subjects and in patients with peripheral obliterative arterial disease before and after an isotonic exercise. Red blood cells and platelets from control subjects and from patients were tested in several ratios before and after the exercise studying the erythrocyte filterability. beta-thromboglobulin, a platelet-specific protein, was determined in vivo in all subjects before and after the exercise and in vitro as percent of increase between PPP and PRP activated, to monitor the platelet release. The results showed the interrelationship between platelet activation and rheological behavior of red blood cells.

[The effect of lysine acetylsalicylate on hemorrheological changes induced through ischemic exertion in patients with vascular disease].

This study has been performed to evaluate the in vivo effect of acetylsalicylic acid (ASA) i.v. infusion at two different dosages: 500 and 2,000 mg on the main hemorheological parameters (blood viscosity and filtrability, hematocrit) and on beta-thromboglobulin (a platelet-specific protein) in patients with peripheral obliterative arterial disease, before and after an ischemic exercise. The results obtained before and 3 and 24 h after the drug administration induced us to think that platelet-specific substances released during the ischemic exercise can affect the hemorheological parameters and that ASA may be able to interfere with these reactions by inhibiting the platelet release.

[Risk factors in maternal-fetal infections. Research conducted in 93 cases].

The results of microbiologic cultures from 186 vaginal samples obtained from 93 labouring women have been compared with the results of the microbiologic cultures from meconium of their newborns, to test possible risk-factors for IMF. The high frequency of positive cultures from vaginal samples (83.3%), constantly with strong bacterial charge, and the proved direct mother to foetus transmission for many potential pathogens (23.6%), cause various interpretative problems and require further studies.
