Age-dependent accumulation of thymic hormone-sensitive cells.

T-cell subsets were investigated in the thymus and spleen of BALB/c mice from 1 to 9 mth of age. It was found that in both of these organs, the proportion of T cells with a high surface concentration of Thy 1 antigen increased with age. The intensity of the graft-versus-host reaction induced by spleen and lymph node cells in F1 hybrids also increased with age. On the other hand, the response of thymocytes to Con A measured by migration inhibition decreased with age. All of these parameters could be reversed to the 1-mth-old levels by preincubation of the cells with a thymic hormone preparation. This indicates that in the time span under study, the changes observed could be attributed to an accumulation of thymic hormone-sensitive cells.

[Bronchial arteriography in the localization and treatment of haemoptysis. Report of 34 observations (author's transl)].

34 bronchial arteriograms were performed in 34 patients over two years. The authors tried to assess the value of this technique in the localization and treatment of haemoptysis. The study confirms the excellent results which are obtained immediately by selective embolisation to achieve haemostasis in severe cases of haemoptysis studied acutely (12 out of 14 cases). The value of arteriography was mediocre when used to localize the lesion (12 times out of 31 assessable cases). Accordingly the authors currently limit bronchial arteriography to those cases where embolisation is indicated and cases in which previous investigation had failed to yield a cause.

Central respiratory drive in acute respiratory failure of patients with chronic obstructive pulmonary disease.

Control of breathing was studied in patients with chronic obstructive pulmonary disease, both in the chronic state and during acute respiratory failure. The results were compared to those in a group of age-matched normal subjects. In patients breathing air, minute ventilation was not different during acute and chronic states, and was similar to that in normal subjects. The pattern of breathing, however, was different: acutely ill patients took shorter and smaller breaths, with a breathing frequency higher than that of normal subjects. The pattern of the chronic group was intermediate between that of acutely ill patients and that of normal subjects. Mouth occlusion pressure, an index of neuromuscular respiratory drive, was 5 times greater in acutely ill patients than in normal subjects. Administration of O2 at a flow of 5 L/min caused a small (14%), bus significant, decrease in minute ventilation due to decreased respiratory frequency. The tidal volume did not change, so the decrease in minute ventilation was the result of decreased inspiratory flow. This was associated with a decreased mouth occlusion pressure that was still 3 times greater than that of normal subjects. The increase in arterial PCO2, observed after administration of O2 was not correlated with the decrease in ventilation, indicating that other factors must be responsible for the increase in arterial PCO2. We concluded that (1) despite the poor mechanical advantage of the respiratory muscles in acute respiratory failure, the increased drive to breathe results in high mouth occlusion pressure and inspiratory flow, and (2) the increase in arterial PCO2, observed during administration of O2 is not related solely to changes in respiratory drive.

Evidence for circadian variations in serum thyrotropin, 3,5,3'-triiodothyronine, and thyroxine in the rat.

We previously observed that under a 12-h light, 12-h dark schedule (lights off at 1900 h), male Sprague-Dawley rats showed a circadian rhythm for serum TSH with a zenith near midday. In the present work we further characterized the serum TSH rhythm by appropriate mathematical analysis. The peak of serum TSH occurred at 1130 +/- 0105 h (mean of five experiments), with an amplitude of 0.2 +/- 0.1 microgram/ml, while the TSH level was minimum at the beginning of the dark period; the period of the TSH rhythm was 24.5 +/- 0.6 h. A circadian rhythm was also demonstrated for serum T3 and T4; these rhythms were characterized by peaks occurring 1.5 and 2.2 h after that of TSH, respectively. As expected, characteristics of the rhythm were more narrowly defined for T3 than for T4. Serum concentrations peaked at 1256 +/- 0206 and 1346 +/- 0308 h for T3 and T4, respectively; the serum T3 concentration varied from 78.7 +/- 7.8 to 54.2 +/- 2.7 ng/100 ml (P < 0.001) and serum T4 varied from 7.3 +/- 1.1 to 5.1 +/- 0.5 microgram/100 ml (P < 0.05) for zenith and nadir values, respectively. It is suggested that the diurnal peaks of thyroid hormones might be related to TSH-induced changes in thyroid secretion.

[Extensive bronchial dystrophy. Physiopathology and treatment of suppurative complications (author's transl)].

Extensive bronchial dystrophy differs by its histopathology, course and treatment from chronic obstructive bronchitis. This dystrophy leads to cavities which are virtually constantly infected. The extension of infection to adjacent alveoli may be dramatic, contrasting with underlying slight chronic respiratory failure. The existence of these cavities explains the preponderant role in therapy of drainage and the fact that antibiotic therapy would appear to be indicated only in the case of extension of infection to the alveoli and the peribronchial tissues.

Effect of different components of the thymic stroma on the onset of AKR leukaemia.

Spontaneous leukaemia in AKR mice is characterized by an accumulation of thymocyte precursors prior to the onset of the disease. An insoluble thymic fraction (ITF), previously shown to provoke such an accumulation when injected to normal mice, caused a significant acceleration of death by leukaemia when given to pre-leukaemic AKR mice. Conversely, the injection of a soluble thymic factor (STF), believed to further differentiate thymocyte precursors, was shown to markedly delay the onset of leukaemia. These results are interpreted as reflecting the specific immune functions of T cells at different stages of maturation.

[Abnormalities in bronchial permeability after induction of a chronic SO2 bronchopathy in rat (author's transl)].

Our aim was to study the effects of a chronic exposure to SO2 on the permeability of bronchial epithelium in the rat, in vivo and in vitro, in relation with a marker (ferritin). The anatomical lesions observed in light and electron microscopy considerably increased the passage of ferritin both in vivo and in vitro through the epithelium of trachea and primary bronchi. This increase was still found although more discrete three months after the end of exposure while the histological appearance returned to normal. Those results showed that SO2 induces in upper airways functional abnormalities persisting beyond the necessary lapse of time for the repairing of histological lesions which could therefore intervene in the pathogeny of chronic bronchopathies.

Primary structure of bovine liver tRNATrp.

Purified tRNATrp from bovine liver, accepting 1700 pmol tryptophan per A260nm unit, was completely digested with pancreatic ribonuclease and T1 ribonuclease. The sequences of the resulting oligonucleotides were determined and the primary structure of the tRNA was deduced. These analyses showed numerous incomplete post-transcriptional modifications, and several positions heterogenously occupied by two different nucleotides, which lead us to think that in bovine liver there exist a mixture of several tRNATrp.

[Pan-lobular emphysema: relationship with serum alpha-1-antitrypsin levels, Pi phenotype and the HLA system (author's transl)].

Pi phenotypes have been studied in a group of 433 patients. Patients with panacinar emphysema and/or bullae were identified from careful analysis of their clinical, physiological, radiological and anatomical characteristics. Twenty-five p.cent of the emphysematous patients were MZ; there was no significant difference in the alpha-1-antitrypsin plasmatic levels between the groups. The HLA antigens were studied in order to try to identify the possible cofactors in the development of emphysema. The role of occupational pollutants and/or of tobacco is discussed. The frequency of the MZ phenotype in our series differs from previous publications. This discrepancy is discussed on the basis of the criterious used to identify the emphysematous patients.

[Study of the thyroid function by the I-M TRH test (2 mg) (author's transl)].

The responses of TSH, T3 and T4 to intramuscular administration of 2 mg of TRH were compared in the same subjects to those observed after intravenous injection of 200 microgram of TRH. In 12 normal subjects the TSH and T3 peaks were significantly higher after intramuscular injection. There was no difference in T4 levels. In 15 patients: 5 toxic adenomas after surgery and 10 Graves' disease after treatment the results were comparable for TSH and T3 by the two routes.

tRNATrp (bovine) binding to the reverse transcriptase of avian myeloblastosis virus and function as a heterologous primer.

The primary structures for tTNATrp (bovine) and primer tRNATrp (avian) show only minor differences in nucleotide sequence. The heterologous tRNATrp (bovine) appears to have properties similar to the tRNATrp (avian) in its ability to bind the alphabeta from of RNA-dependent DNA nucleotidyltransferase of avian myeloblastosis virus. A stable enzyme-tRNA complex has been isolated by gel filtration. In addition, tRNATrp (bovine) can hydridize to the avian viral 35S RNA and act as a primer for transcription of the RNA. tRNATrp (bovine) can be obtained in larger amounts than the avian primer and can be used to study the interactions between the primer and the viral enzyme.
